Immunohistochemie is een veelzijdig hulpmiddel dat kan worden gebruikt om de spatio-temporele expressie van vrijwel elk eiwit van belang in een organisme te karakteriseren. De zebravis (Danio rerio) is ontstaan als een krachtig model voor de studie van biologische processen om verschillende redenen, waaronder korte generatie tijd, snelle ontwikkeling, en beaambaarheid aan genetische technieken. Als gevolg hiervan worden zebravissen vaak gebruikt in hoge doorvoer kleine molecuul schermen voor toxicologisch onderzoek en drugs ontdekking. Zebravissen zijn ook een aantrekkelijk model voor de studie van ontwikkelingsprocessen, aangezien één vrouwtje routinematig 50-300 eieren tegelijk kan produceren en de optisch heldere embryo's zich extern ontwikkelen, waardoor ontwikkelingsprocessen efficiënt kunnen worden gevisualerd. Echter, vroeg onderzoek baseerde zich meestal op voorwaartse genetische schermen met behulp van N-ethyl-N-nitrosourea (ENU) of andere mutageens als gevolg van uitdagingen bij het vaststellen van omgekeerde genetische technieken. Ongeveer twee decennia geleden werden morpholinos voor het eerst gebruikt bij zebravissen om gerichte genen neer te halen1. Morpholinos zijn kleine antisense oligonucleotiden die de vertaling van doelmRNA remmen na micro-injectie in een embryo in een vroeg ontwikkelingsstadium. Een grote zwakte van morpholinos is dat ze worden verdund als de cellen te verdelen en over het algemeen verliezen effectiviteit door 72 uur na de bevruchting (hpf). Terwijl morpholinos een krachtig instrument blijven voor verstoring van het zebravisgen, worden transcriptionor-achtige effector nucleases (TALENs), zink-vinger nucleases (ZFNs), en geclusterd regelmatig interspaced korte palindromic herhalingen (CRISPRs) meer recent gebruikt om direct gericht op de zebravis genoom2,3. Deze omgekeerde genetische strategieën, in combinatie met voorwaartse genetica en hoge doorvoerschermen, hebben de zebravissen vastgesteld als een krachtig model om genexpressie en functie te bestuderen.
De capaciteit om genfunctie te bestuderen vereist over het algemeen een evaluatie van de spatio-tijdelijke distributie van gen of genproductuitdrukking. De twee meest gebruikte technieken om dergelijke expressiepatronen te visualiseren tijdens de vroege ontwikkeling zijn in situ hybridisatie (ISH) en hele mount immunohistochemie (IHC). In situ hybridisatie werd voor het eerst ontwikkeld in 1969 en is gebaseerd op het gebruik van gelabelde antisense RNA sondes om mRNA expressie in een organisme te detecteren4. In tegenstelling, gelabelde antilichamen worden gebruikt in immunohistochemie om eiwitexpressie te visualiseren. Het idee van het etiketteren van eiwitten voor detectie dateert uit de jaren 19305 en de eerste IHC experiment werd gepubliceerd in 1941 toen FITC-gelabelde antilichamen werden gebruikt om pathogene bacteriën in geïnfecteerde weefsels op te sporen6. ISH en IHC zijn in de daaropvolgende decennia sterk geëvolueerd en verbeterd en worden nu zowel routinematig gebruikt in het moleculaire en diagnostische onderzoekslaboratorium7,8,9,10,11. Hoewel beide technieken voor- en nadelen hebben, biedt IHC verschillende voordelen ten opzichte van ISH. Praktisch, IHC is veel minder tijdrovend dan ISH en is over het algemeen minder duur, afhankelijk van de kosten van de primaire antilichaam. Bovendien, mRNA expressie is niet altijd een betrouwbare metrische van eiwitexpressie als het is aangetoond bij muizen en mensen dat slechts ongeveer een derde van de eiwit overvloed variatie kan worden verklaard door mRNA overvloed12. Om deze reden is IHC een belangrijk supplement om ISH-gegevens te bevestigen, indien mogelijk. Ten slotte kan IHC subcellulaire en co-lokalisatiegegevens verstrekken die niet kunnen worden bepaald door ISH13,14,15. Hier beschrijven we een stapsgewijze methode om eiwitten betrouwbaar te detecteren door immunohistochemie in hele mount zebravisembryo's en larven. Deze technologie maakt gebruik van antigeenspecifieke primaire antilichamen en fluorescerend gelabelde secundaire antilichamen. Het protocol is gemakkelijk aan te passen aan gebruik op dia-gemonteerde weefselsecties en voor gebruik met chromogene substraten in plaats van fluorescentie. Met behulp van dit protocol, we aantonen dat de ontwikkeling van zebravissen skeletspier drukt ionotropische glutamaat receptoren, in aanvulling op acetylcholine receptoren. NMDA-type glutamaat receptor subunits zijn detecteerbaar op de longitudinale spier op 23 pkf.

Immunohistochemie is een veel gebruikte techniek om eiwitexpressie en lokalisatie te verkennen tijdens zowel normale ontwikkelings- als ziektetoestanden. Hoewel veel immunohistochemie protocollen zijn geoptimaliseerd voor weefsel- en weefselsecties van zoogdieren, vereisen deze protocollen vaak modificatie en optimalisatie voor niet-zoogdiermodelorganismen. Zebravissen worden steeds vaker gebruikt als een modelsysteem in fundamenteel, biomedisch en translationeel onderzoek om de moleculaire, genetische en celbiologische mechanismen van ontwikkelingsprocessen te onderzoeken. Zebravissen bieden veel voordelen als modelsysteem, maar vereisen ook aangepaste technieken voor optimale eiwitdetectie. Hier bieden we ons protocol voor de gehele mount fluorescentie immunohistochemie in zebravissen embryo's en larven. Dit protocol beschrijft bovendien verschillende montagestrategieën die kunnen worden gebruikt en een overzicht van de voor- en nadelen die elke strategie biedt. We beschrijven ook wijzigingen in dit protocol om detectie van chromogene substraten in hele mountweefsel en fluorescentiedetectie in gesectioneerd larveweefsel mogelijk te maken. Dit protocol is in grote lijnen van toepassing op de studie van vele ontwikkelingsstadia en embryonale structuren.

Hele mount immunohistochemie is een relatief snelle en eenvoudige techniek die het mogelijk maakt voor de ruimtelijke en temporele observatie van eiwitten direct in een weefsel of organisme met behulp van gelabelde antilichamen. Het nut van deze techniek is dat het zorgt voor de kleuring van een intact weefsel of een organisme zonder eerst sectie en confocale microscopie kan worden gebruikt om een driedimensionale weergave van het eiwitexpressie patroon te genereren. Immunohistochemie wordt veel gebruikt in biomedisch onderzoek om de etiologie en moleculaire mechanismen die ten grondslag liggen aan de ziekte te begrijpen.Het is ook een waardevol diagnostisch hulpmiddel om tumoren in pathologische monsters te identificeren. Om te beginnen, bereiden paaitanks gevuld met systeemwater en met een mesh of sleuven voering op zijn plaats. Plaats volwassen zebravissen gemengde geslacht paren in groepen in de tanks 's nachts.Gebruik een 14-uurs/10-uur licht/donker cyclus met lichten die om 9.m aan gaan. De volgende ochtend, met de lichten aan, verander de paaitank water met vers systeem water om uitwerpselen te verwijderen. Zodra eieren zijn gelegd, de volwassenen terug naar huis tanks.Verzamel de eieren door ze op te stellen met behulp van een transfer pipet. Breng 50 eieren naar elke petrischaal gevuld halverwege met embryo medium. Verwijder alle ondoorzichtige en troebele eieren die dood zijn of niet te verdelen.Incubeer de schotel van eieren bij 28.5 graden Van Celsius tot 23 uren na bevruchting. Als embryo's ouder zijn dan 24 uur na de bevruchting, met een pipet brengen de embryo's over naar 200 micromolar PTU in embryomedium om melanogenese te voorkomen. Onder een stereomicroscoop, dechorionate de unhatched embryo's met behulp van ultra fijne tip tang.Met behulp van brandgepolijste Pasteur pipetten om schade te minimaliseren, breng ze in glas of plastic Petri schotels bedekt met 1-2%agarose opgelost in embryo medium. Breng de embryo's vervolgens over op 1,5 milliliter centrifugebuizen met een plastic of vuurgepolijste pipet. Verwijder embryomedium met een micropipet.Laat alleen genoeg vloeistof om gewoon de embryo's te dekken. Bevestig de embryo's in 4%PFA gedurende een tot twee uur met zacht schommelen bij kamertemperatuur. Was vervolgens de embryo's drie keer in 1X PBS en 1%PBTriton gedurende vijf minuten.Gebruik de embryo's onmiddellijk of bewaar maximaal een week op vier graden Celsius. Voor langdurige opslag de hydrateren en opslaan van de embryo's in 100% methanol bij min 20 graden Celsius. Om de embryo's te hydrateren, voert u seriële incubaties uit vijf minuten per stuk bij kamertemperatuur met afnemende hoeveelheden methanol en PBS en PBTriton voor de uiteindelijke wasbeurt.Ga verder met het embryopreparaat volgens het manuscript. Kies een blokkeringsoplossing die overeenkomt met de secundaire antilichaamgastheersoort, bijvoorbeeld 10% geitenserum in PBTriton met of zonder twee milligram per milliliter BSA. Voeg 0,5 milliliter van de blokkeeroplossing toe in de buis met het embryo en plaats de buis één tot drie uur op een tuimelschakelaar bij kamertemperatuur.Dan incubeer het embryo in primaire antilichaam verdund in blokkeren oplossing en PBTriton 's nachts op vier graden Celsius tijdens het schommelen. In de ochtend, was het embryo vijf keer in PBTriton gedurende 10 minuten elk op kamertemperatuur tijdens het schommelen. Selecteer vervolgens een secundair antilichaam op basis van de gastheersoort van het primaire antilichaam en de gewenste golflengte op 488 nanometer.Voeg het secundaire antilichaam verdund in het blokkeren van oplossing in de buis met het embryo. Bedek de buis met aluminiumfolie en broed gedurende twee uur op kamertemperatuur tijdens het schommelen. Daarna, was de embryo's drie keer in PBTriton gedurende 10 minuten elk op kamertemperatuur, terwijl bedekt met aluminiumfolie en schommelen.Breng de embryo's met een pipet over naar een 50%glyceroloplossing in PBS over een bed van 2%agarose in embryomedium. Om de dooier te verwijderen, breng je 200 microliter 1X PBS over naar een depressieschuif of een gewone glazen schuif. Gebruik een plastic transfer pipet om een of meer embryo's naar de PBS druppel te verplaatsen.Gebruik ultra fijne tangen en dubbele nul insectenspelden om de dooier uit elkaar te halen en dooierkorrels zeer voorzichtig van het ventrale oppervlak van het embryo te schrapen. Verwijder de dooierkorrels en vul aan met PBS als dat nodig is. Na montage, plaats het monster op de microscoop stadium.Zoek het interessegebied door een relatief helder voorbeeld te selecteren. Pas de belichting van de camera aan en verkrijg de belichting zodat het signaal voldoende helder is zonder te verzadigen. Gebruik bij het vergelijken tussen de experimentele embryo's met antilichaam en controleantilichaamembryo's dezelfde blootstellingsinstellingen.Dit protocol van hele mount immunohistochemie is een waardevol instrument voor de studie van ruimtelijke en temporele eiwitexpressie met behulp van zebravis ontwikkeling. De GluN1 subunit van het NMDA type glutamaat receptor bleek expressie van glutamaat receptor subeenheden tijdens de ontwikkeling van zebravis spieren op 23 uur na de bevruchting. Bevroren delen van 72-uurs postbevruchting embryo's werden gemonteerd op dia's en immunovlekken voor fosfo-H3, een marker van zich vermenigvuldigende cellen.Verscheidene cellen drukden fosfo-H3 uit en de uitdrukking was het opmerkelijkst bij de ventriculaire streken. Muis IgG controle antilichaam en met uitzondering van primaire antilichamen bleek een laag niveau van niet-specifieke expressie. Het is belangrijk om geschikte blokkeringsoplossingen en secundaire antilichamen te selecteren op basis van de primaire antistoffen gastheersoorten om detectie te garanderen en niet-specifieke kleuring te minimaliseren.Immunohistochemie moet worden geverifieerd om ervoor te zorgen dat het waargenomen signaal daadwerkelijk het eiwit van belang is. Follow-up experimenten meestal te betrekken met behulp van genetisch of ecologisch gemodificeerde organismen. Immunohistochemie verleent onderzoekers de mogelijkheid om eiwitten in situ te observeren, waardoor ons begrip van talrijke systemen in zowel de basis- als de toegepaste biologie sterk wordt versneld, met name tijdens de ontwikkeling van embryo's.Methanol en formaldehyde zijn giftig en moeten zorgvuldig worden behandeld. Afval moet worden ingezameld en afgevoerd volgens institutionele procedures.
