Inflammatoire darmziekte (IBD) is een inflammatoire chronische ziekte in het maagdarmkanaal (DARM). In de Verenigde Staten zijn er ongeveer 1.400.000 IBD patiënten. Het is algemeen aanvaard dat een disgereguleerde immuunrespons op darmbacteriën de ziekte initieert en de mucosale epitheliale barrière1,2verstoort. Om deze reden, momenteel beschikbare U.S. Food and Drug Administration (FDA)-goedgekeurde drugs remmen de functies van inflammatoire mediatoren of blokkeren het homing van immune cellen in de darm. Echter, de ontstekingsmediatoren en immuuncellen die gericht zijn zijn ook nodig voor immuun afweer. Dientengevolge, de inflammatoire mediator remmers compromitteren systemische immuun verdediging en de immuuncel homing blokkers verzwakken gut immuun verdediging, die beide kunnen leiden tot ernstige gevolgen3,4. Bovendien, de immuuncel homing blokkers kunnen ook blokkeren het homing van regelgevende T (Treg) cellen in de darm en dus kan verergeren de reeds gecompromitteerde gut immuun tolerantie bij IBD patiënten. Bovendien, blokkeren van Treg cel homing in de darm kan ook leiden tot systemische immuunsuppressie als gevolg van de accumulatie van Treg cellen in het bloed5. Tot slot, remmers en blokkers functioneren tijdelijk en, daardoor, vereisen frequente toedieningen. Frequente toediening van deze remmers en blokkers kan verder verergeren de ongewenste neveneffecten.
Onlangs, we hebben voorgesteld een nieuwe strategie die potentieel kan verzachten of zelfs elimineren van de bijwerkingen geassocieerd met de huidige drugs voor IBD behandeling6. Deze strategie vergroot de inductie van gut-homing Treg cellen in perifere lymfoïde weefsels6. De logica van deze strategie is dat gut-homing Treg cellen specifiek de thuisbasis van de darm en dus zal niet in gevaar brengen systemische immuun afweer. Bovendien, aangezien Treg cellen potentieel geheugen kunnen vormen7,8, gut-homing Treg cellen kunnen mogelijk zorgen voor een stabiele controle van de chronische darmontsteking bij IBD patiënten en, daardoor, de behandeling moet niet zo frequent worden toegediend. Bovendien, aangezien deze strategie de inductie van gut-homing Treg cellen in vivo vergroot, heeft het niet de zorg van in vivo instabiliteit in een zeer proinflammatoire omgeving die gepaard gaat met adoptie overdracht van in vitro gegenereerde Treg cellen9,10. In dit verband, in vitro gegenereerde Treg cellen zijn een van de voorgestelde strategieën voor de behandeling van auto-immuunziekten11,12,13 en transplantatie afstoting14,15. Tot slot, in deze strategie, dendritische cellen (DCs) zijn ontworpen om lokaal hoge concentraties van twee moleculen de Novo te produceren: actieve vitamine D (1,25-dihydroxyvitamin D of 1,25 [OH]2D) en actieve vitamine A (retinoïnezuur of RA). We kozen voor 1,25 (Oh)2d en RA omdat 1,25 (Oh)2d de expressie van regelgevende moleculen kan induceren (bijv. Forkhead Box P3 [foxp3] en interleukine-10 [Il-10])16,17 en dat Ra de expressie van gut-homing-receptoren in T-cellen kan stimuleren18. Omdat zowel 1,25 (Oh)2D en RA ook de DCS28,29kunnen vertolderen, reden we dat de engineered DCS in vivo in een tolerogene toestand zal worden gehandhaafd en daarmee de in vivo instabiliteit problemen die geassocieerd zijn met in vitro gegenereerde tolerogene dc's (toldc's)19,20,21te omzeilen. In dit opzicht zijn toldc's ook een van de voorgestelde strategieën voor in vivo vergroting van de Treg-celfuncties19,20,21. Ter ondersteuning van onze redenering, hebben we aangetoond dat de engineered DCs, bij in vivo levering, kan de inductie van gut-homing Treg cellen in perifere lymfoïde weefsels te vergroten6.
Een bijkomend voordeel van onze voorgestelde strategie is dat 1,25 (OH)2D ook andere functies heeft die mogelijk ten goede komen aan IBD-patiënten. Deze andere functies omvatten het vermogen van 1,25 (OH)2D om de secretie van antimicrobiële geneesmiddelen22 te stimuleren en carcinogenese23te onderdrukken. Infecties en kankers worden vaak geassocieerd met IBD24,25.
Om de DCs te genereren die lokaal hoge concentraties van zowel 1,25 (OH)2D en Ra de Novo kan produceren, gebruiken we een lentivirale vector om DCS te overdrukken om twee genen over te brengen. Een gen is de cytochroom P450 familie 27 subfamilie B lid 1 (CYP27B1) die codeert 25-hydroxyvitamin D 1α-hydroxylase (1α-hydroxylase), die fysiologisch katalyseert de synthese van 1,25 (OH)2D. Het andere gen is de aldehyde-dehydrogenase 1-familielid a2 (ALDH1a2) die de retinaldehyde dehydrogenase 2 (RALDH2) codeert, wat de synthese van RA6fysiologisch katalyseert.
Omdat in vivo augmentatie van gut-homing Treg Cell inductie is mogelijk belangrijk bij de behandeling van IBD, in het volgende protocol zullen we de procedures voor het genereren van de 1α-hydroxylase-RALDH2-overuitdrukken DCs (DC-CYP-ALDH cellen) die kan gebruikt worden voor het toekomstig onderzoek van de Treg cellen in vivo.

in vivo augmentation of gut-homing regulatory t cell induction

Inflammatoire darmziekte (IBD) is een inflammatoire chronische ziekte in het maagdarmkanaal (DARM). In de Verenigde Staten zijn er ongeveer 1.400.000 IBD patiënten. Het is algemeen aanvaard dat een disgereguleerde immuunrespons op darmbacteriën de ziekte initieert en de mucosale epitheliale barrière verstoort. We hebben onlangs laten zien dat gut-homing Regulatory T (Treg) cellen een veelbelovende therapie voor IBD zijn. Dienovereenkomstig, dit artikel presenteert een protocol voor in vivo augmentatie van gut-homing Treg Cell inductie. In dit protocol zijn dendritische cellen ontworpen voor de productie van lokaal hoge concentraties van twee moleculen de Novo, actieve vitamine D (1,25-dihydroxyvitamin D of 1,25 [OH]2D) en actieve vitamine A (retinoïnezuur of RA). DC-CYP-ALDH cellen uitgedrukt significant toegenomen hoeveelheid 1α-hydroxylase. In Vivo Augmentation of Gut-Homing Regulatory T Cell Induction

Dit protocol beschrijft een potentieel nieuwe therapeutische strategie die gemakkelijk kan worden gewijzigd voor menselijke toepassingen. Deze techniek verbetert direct de inductie van darm-homing regulerende T-cellen in perifere lymfeweefsels en kan zelfs in zeer pro-inflammatoire omgevingen stabiel worden geïmplementeerd. Deze technieken kunnen ook worden gebruikt om de rol van actieve vitamine D en vitamine A binnen het perifere immuunsysteem te onderzoeken.Deze visuele demonstratie geeft stapsgewijze instructies voor het genereren van hoogwaardige ontworpen dendritische cellen die cruciaal zijn voor het succes van deze techniek. Begin met het zaaien van een keer 10 tot de zeven 293T cellen in 20 milliliter CM-10-D cel cultuur medium in 150 door 25 millimeter kweekplaten voor een 24-uurs incubatie bij 37 graden Celsius en 5% kooldioxide. Voeg de volgende dag 1,62 milliliter vers bereide tweevoudig geconcentreerde HBS-oplossing, 9,5 microgram enveloppe plasmid, 17,5 microgram verpakkingsplasmid en 27 microgram van de LENTI-CYP-ALDH plasmid toe aan een conische buis van 50 milliliter.Breng het uiteindelijke volume van de buisinhoud tot 3,0375 milliliter met water, gevolgd door de toevoeging van 202,5 microliter van twee molaire calciumchloride dropwise. Laat het DNA-neerslagmengsel 20 minuten op kamertemperatuur met af en toe mengen voordat u 3,24 milliliter van de oplossing dropwise per 293T-cultuur toevoegt terwijl de schotel zachtjes wervelt. Concentreer vervolgens het VSV-G pseudotyped virus door centrifugatie gedurende 24 uur.De volgende dag decanteren de supernatants. Laat de buizen enkele minuten op een papieren handdoek in een steriele bioveiligheidskast uitlekken en zet de pellets opnieuw op in 33,3 microliter steriele PBS met 5%glycerol per kweekgerecht. Voor beenmerg afgeleide dendritische cel generatie, plaats de scheenbeen en femurs, geoogst van BALB / c muizen, in een 100 bij 20 millimeter cultuur schotel met verse cultuur medium.Gebruik ontleden schaar en tangen om alle weefsels te verwijderen uit de botten. Wanneer alle botten zijn gereinigd, gebruik de schaar om beide uiteinden van elk bot te snijden om de beenmergholtes bloot te leggen. Gebruik een 10 milliliter spuit uitgerust met een 30-meter naald om elk bot te spoelen met 10 milliliter vers kweekmedium, het verzamelen van het beenmerg in een 15 milliliter buis.Wanneer al het beenmerg is verzameld, sediment het beenmerg door centrifugatie en resuspend de pellet in twee milliliter van rode bloedcel lyse buffer. Na vier tot vijf minuten bij kamertemperatuur met af en toe schudden, stop de reactie met 10 milliliter celkweekmedium en verzamel de cellen door centrifugatie. Resuspend de pellet in 10 milliliter cm-10-R cultuurmedium voor het tellen en aanpassen van de cellen voor een keer 10 tot de zes cellen per milliliter van CM-10-R cultuur middelgrote concentratie.Voeg vervolgens 100 eenheden per milliliter recombinant murine GM-CSF en 10 eenheden per milliliter IL-4 toe aan de cellen. Plaat vier milliliter cellen in individuele putten van een zes goed kweekplaat voor hun incubatie bij 37 graden Celsius en 5% kooldioxide gedurende 48 uur. Aan het einde van de incubatie, zachtjes aspirate de niet-aanhangende cellen.Voeg vers medium aangevuld met GM-CSF en IL-4 toe aan de culturen voordat u de cellen 48 uur naar de celkweek incubator terug brengt. Oogst aan het einde van de tweede incubatie de niet-aanhangende beenmerg afgeleide dendritische cellen in 50 milliliter conische buis voor hun centrifugatie. Om BMDC-CYP-ALDH cellen te genereren, hersuspend de culturen op een een keer tien tot de zesde dendritische cellen per 500 microliters van CM-10-R medium per put concentratie, en plaat 500 microliter cellen in elke put van een nieuwe zes put cultuur plaat.Voeg 50 microliter LENTI-CYP-ALDH virus en 8 microgram per milliliter protamine toe aan elke put en plaats de plaat vervolgens 24 uur in de couveuse. De volgende dag, vervang de supernatants door verse cultuur medium en breng de platen terug naar de cel cultuur incubator voor nog eens 24 uur. Aan het einde van de tweede incubatie kunnen de cellen gedurende 24 uur voor de oogst worden geactiveerd met 100 nanogram per milliliter lipopolysaccharide per put voor functionele analyses.In vergelijking met ouderlijke beenmerg afgeleide dendritische cellen, BMDC-CYP-ALDH cellen vertonen een verbeterde expressie van 1-alpha-hydroxylase. De concentraties van de actieve vorm van vitamine D in de kweeksupernatants van BMDC-CYP- en BMDC-CYP-ALDH-cellen zijn ongeveer 20 keer hoger dan die welke worden waargenomen in de door ouderlijke beenmerg afgeleide dendritische cel en de BMDC-ALDH-celculturen. De gemiddelde fluorescerende intensiteiten van BMDC-CYP-ALDH cellen behandeld met het substraat zijn ongeveer zes keer hoger dan die van ouderlijke beenmerg afgeleide dendritische cellen behandeld met het substraat.Dit suggereert dat de BMDC-CYP-ALDH cellen aanzienlijk verbeterde retinale dehydehydrogenase-2 enzymatische activiteit uitdrukken. DC2.4 cellen behandeld met 25-hydroxy vitamine D en retinale veroorzaken geen significante verandering in de overvloed van foxp3 positieve CDR9 positieve CD4 positieve T-cellen. Dc2.4 CYP-ALDH-cellen behandeld met 25-hydroxy vitamine D en retinale veroorzaken daarentegen aanzienlijk hogere aantallen darm-homing regulerende T-cellen op een concentratieafhankelijke manier.Bovendien veroorzaken DC CYP-ALDH-cellen, in vergelijking met intraperitoneally geïnjecteerde controlecellen, ook een aanzienlijke toename van het aantal foxp3-positieve CCR9-positieve CD4-positieve T-cellen na intraperitoneale injectie in BALB/c-ontvangende dieren. Het succes van de procedure hangt af van de bereiding van gezonde T-cellen en een correct voorbereid DNA-mengsel.
