1. 准备解决方案
<ol><li>在汉克平衡盐溶液（HBSS）中制备500 mL的洗涤缓冲液A（WBA）：10 mM HEPES溶液。储存在4°C。</li>

</ol>2. 脑组织恢复和脑脑切除
<ol><li>为保持一致性，在每批萃取中使用年龄相仿和性别相同的小鼠。使用无病原体的动物庇护所，并提供获得水的机会。为了确保动物的效率和使用，避免组织材料的损失。</li>
	<li>安乐死C57BL/6J，4-6周大，雄性小鼠（扬维尔实验室，法国圣岛勒）。</li>
	<li>在无菌条件下快速切除脑组织，避免对组织的任何损伤。小心地将组织置于40 mL的冷磷酸盐缓冲盐水 （PBS） 中。</li>

</ol>5. 脑血管扩散

即使生产，这些方法也有几个限制，从使用大量动物进行单批次隔离到输出量非常有限。

从P0到P2，有特定的形态特征，通过这些特征可以观察到内皮细胞（用白色箭头表示）和围细胞的逐渐增加（用黑色箭头表示）。在P0中，从微血管发育的细长内皮细胞是丰度（图2A，P0：第3天），而这种拉长细胞的丰度在P1中减少，在P2中不存在。相反，在P2中，围卵体表现为丰富的四边形细胞（图2A，P2：第18天）。
为了确认P2中过冰菌培养物的纯度，我们使用定量PCR（图2B）、免疫细胞化学（图2C）和西印（图3）检查了NG2、CD146和PDGFR-beta作为过核子标记的表达。与小鼠大脑总（Ms Br）提取物中的表达相比，P2中的围杨表示更高的CD146、NG2和PDGFR-+水平。作为对照，在P2中也观察到内皮标记物闭塞和CD31、星形细胞标记物胶质纤维酸性蛋白（GFAP）和微胶合标记CD11b的表达。
