単眼的な視覚剥奪は、一次視覚皮質応答可塑性を誘導する優れた実験プロトコルである。通常、マウスの一次視覚皮質の双眼鏡セグメントのニューロンは、両側眼に対する対側眼の刺激に約2倍強く反応する。しかし、縫合は、眼支配可塑性の重要な期間中に対して対側眼を閉じ、V1ニューロンによる対側眼刺激への応答性の急速な喪失をもたらし、その後、イプシラショナル眼への応答の増加も続く。ここでは、単眼剥奪の実験プロトコルを紹介し、視覚剥奪時の眼支配の動向を分析するために一般的に使用される方法を提案する。手術用具をアルミボックスに入れ、120度以下で30分間オートクレーブします。水浴釜で2%アガロース溶液を調製し、摂氏75度に保ちます。両眼に目軟膏の薄い層を適用します。照明付きの解剖顕微鏡の下で、まぶたを縫合する。約4針を作ります。スレッドのノードを 2 ~ 3 個作成します。ノードの安定性を高めるために、ノードの頭部に3マイクロリットルの瞬間乾燥接着剤502を塗布する。その後、スレッドを十分に短くカットします。マウスが完全に目を覚ましたら、それらを分離された保持ケージに入れます。電気生理学的記録の前に、イオブルランを使用してマウスを麻酔してください。ステッチを取り外し、眼球を露出させる。まぶたを慎重にトリミングします。コンタクトレンズ溶液で目を洗い、目を明瞭にチェックしてください。眼球の良好な状態のマウスのみ使用できます。マウスを麻酔した後、ステレオタキシックフレーム上にマウスを置きます。加熱パッドを使用して、手順全体を通して体温を維持します。目の表面に目の軟膏を塗布して、しっとりと保ちます。マウスの毛を取り除き、皮膚を露出させます。頭蓋骨を露出し、頭皮組織を除去するために、両耳の間に8ミリメートルの皮膚領域を掛けた8ミリメートルを切り開きます。次に、過酸化水素で上にある結合組織を取り除きます。小脳の上の頭蓋骨に穴を開けます。穴の小さな骨ねじを参照として固定します。V1双眼領域で直径1ミリの小さな頭蓋切開術を行う。頭蓋の断片を脳を傷つけることなく慎重に取り除いてください。乾燥を防ぐために露出皮質表面を2%アガロースで覆います。ステレオタキシックフレーム上のタングステン電極を固定します。タングステン電極を双眼鏡V1領域に垂直に配置し、記録された細胞が両眼に反応することを確認します。刺激はランダムブロックで30度分離されます。各測定で合計3~5ブロックが提示された。透明でないプラスチックプレートで片目をマスクします。LCD1をマウスの目から23センチメートルに位置させます。油油圧マイクロマニピュレータでゆっくりとマイクロ電極を進めます。電極が4層に進み、測定を開始する場合は、信号の進歩を止めます。スパイク活動を1,000倍に増幅し、300~10キロヘルツで絞り込み、40キロヘルツでサンプリングします。その後、もう一方の目をマスクし、対側の目の応答を測定します。単一電極で測定される信号は集団活動である。ソフトウェアを使用して、さまざまなセルのスパイクを分離します。まず、500~5,000ヘルツの信号をフィルタリングします。正の偏向用と負の偏向スパイク用の 2 つのカーソルを設定します。スパイクテンプレートを設定します。さまざまなクラスのスパイク間で最大のバリエーションを示すテンプレート領域を設定します。主成分分析を使用して、それらをクラスターに分離します。K 平均アルゴリズムを使用して境界のスパイクを分類します。スパイクの発射速度と方向を相関させ、イプシラショナルおよび対側の眼の向きチューニング曲線をプロットします。次に、対側応答および両側応答を表す単一セルの OD インデックスを計算 OD インデックスに従って各セルに OD スコアを割り当てます。数式に従って、対側バイアスインデックスを計算します。この図は、単眼奪われたマウスおよび奪われたマウスにおける、イプシラショナルアイと対側眼の方向調整曲線を示している。このプロトコルは、重要な時期に奪われたマウスと奪われたマウスからの単眼的な視覚剥奪および眼支配測定を成功に導く。この実験を行う方法を示したばかりで、視覚剥奪時の眼支配の変化を分析しました。実験中、電気生理学的記録中の縫合過程や相互作用における感染を避けることは重要である。私たちのビデオを見てくれてありがとう。
