Er zijn een aantal belangrijke punten om in gedachten te houden voor de SP-test. De eerste is de selectie van een goede blokker, zoals Verapamil of Reserpine, voor elke cellijn, omdat de "gate" locatie van de SP-cellen wordt bepaald op basis van de positie waarin een groot aantal SP-cellen verdwijnt na de toevoeging van de blocker. Voor de MDA-MB-231 cellijn werkt Reserpine goed. Voor andere cellijnen kunnen verschillende blokkers echter beter werken.
De tweede is de concentratie van Hoechst 3...

Kankerstamcellen (CSC's) zijn deelverzamelingen van cellen met zelfvernieuwingsvermogen en multipel differentiatiepotentieel, die een vitale rol spelen bij tumorgroei, metastase en recidief1,2. Momenteel zijn CSC's geïdentificeerd om te bestaan in een verscheidenheid van kwaadaardige tumoren, met inbegrip van long-, hersen-, alvleesklier-, prostaat-, borst- en leverkankers3,4,5,6,7,8,9. Identificatie van CSC's in deze tumoren is voornamelijk gebaseerd op de aanwezigheid van oppervlaktemarkereiwitten, zoals hoge en/of lage expressie van CD44, CD24, CD133 en Sca-19,10, maar een unieke marker die CSC's van niet-CSC's kan onderscheiden, is tot nu toe niet gerapporteerd. Momenteel worden verschillende technieken gebruikt om CSC's in tumorweefsel of tumorcellijnen te identificeren en te zuiveren. Deze technieken zijn ontworpen op basis van de specifieke eigenschappen van CSC's. Onder hen, assays en sortering van zijpopulatie (SP) cellen zijn twee van de meest gebruikte methoden.
SP-cellen werden oorspronkelijk ontdekt door Goodell et al.11, toen ze hematopoëtische stamcellen in beenmergcellen van muizen kenmerkten. Toen de beenmergcellen van de muis werden gelabeld met de fluorescerende kleurstof Hoechst 33342, verscheen een kleine groep Hoechst 33342 zwak bevlekte cellen in de tweedimensionale puntplot van een flowcytometrietest. Hoechst 33342 is een DNA-bindende kleurstof en heeft ten minste twee bindingsmodi die leiden tot verschillende spectrale kenmerken. Bij het bekijken van fluorescentie-emissie op twee golflengten tegelijkertijd, kunnen meerdere populaties worden onthuld12. In hun test werd de Hoechst 33342 opgewekt bij 350 nm en werd de fluorescentie gemeten met behulp van het 450/20 nm band-pass (BP) filter en 675 nm edge filter long-pass (EFLP)11. Vergeleken met de hele populatie beenmergcellen werd deze groep cellen verrijkt met hematopoëtische stamcellen genaamd SP-cellen11. SP-cellen kunnen Hoechst 33342 snel verdrijven. De efflux van deze kleurstof is gerelateerd aan ATP-binding cassette (ABC) transporters13, die kunnen worden geremd door sommige middelen zoals Fumitremorgin C14, Verapamil en Reserpine15,16. Daarna werden verschillende verhoudingen SP-cellen gedetecteerd in een verscheidenheid aan weefsels, organen, tumorweefsels en tumorcellijnen17,18,19. Deze SP-cellen hebben veel kenmerken van stamcellen17,19.
Dit manuscript beschrijft Hoechst 33342 etikettering en kleuring van gekweekte tumorcellen en de analyse van SP-cellen door flowcytometrie. Bovendien wordt optimalisatie van de Concentratie Hoechst 33342 en de juiste blokkerselectie voor een specifieke tumorcellijn met behulp van deze aanpak getoond. Ten slotte worden de effecten van stemheidsbevordering of remmingssignalen op het aandeel SP in tumorcellen aangetoond. De experimentele voorbeelden tonen aan dat analyse van SP kan worden gebruikt om de effecten van verschillende signalen te onderzoeken, zoals genexpressie, kleine remmers, activatoren, cytokinen en chemokinen, op tumorstamheid. In vergelijking met andere methoden voor isolatie en zuivering van CSC's, zoals het sorteren van CD44+/ CD24- populatie, aldehydedehydrogenase (ALDH) analyse en tumorbolvormingstesten, is deze methode gemakkelijker te manipuleren en is het kosteneffectief.

Kankerstamcellen (CSC's) zijn een belangrijke oorzaak van tumorgroei, metastase en recidief. Isolatie en identificatie van CSC's zijn van groot belang voor tumoronderzoek. Momenteel worden verschillende technieken gebruikt voor de identificatie en zuivering van CSC's uit tumorweefsels en tumorcellijnen. Scheiding en analyse van zijpopulatiecellen (SP) zijn twee van de meest gebruikte methoden. De methoden zijn gebaseerd op het vermogen van CSC's om fluorescerende kleurstoffen snel te verdrijven, zoals Hoechst 33342. De efflux van de kleurstof wordt geassocieerd met de ATP-binding cassette (ABC) transporters en kan worden geremd door ABC transporter remmers. Methoden voor het kleuren van gekweekte tumorcellen met Hoechst 33342 en het analyseren van het aandeel van hun SP-cellen door flowcytometrie worden beschreven. Deze test is handig, snel en kosteneffectief. Gegevens die in deze test worden gegenereerd, kunnen bijdragen aan een beter begrip van het effect van genen of andere extracellulaire en intracellulaire signalen op de stameigenschappen van tumorcellen.

Volgens deze methode werden vier experimentele SP-analyses uitgevoerd. In de eerste ontdekten we het aandeel SP-cellen in MDA-MB-231, een drievoudige negatieve menselijke borstkankercellijn, onder normale omstandigheden. Na het tellen van de cellen werd Hoechst 33342 toegevoegd aan één buis met 1 x 106 cellen tot een eindconcentratie van 3 μg/ml. Reserpine en Hoechst 33342 werden aan een andere buis toegevoegd aan eindconcentraties van respectievelijk 40 μM en 3 μg/ml. PI werd aan beide buizen toegevoegd....

Analyse van zijpopulatiecellen is een van de meest gebruikte methoden om kankerstamcellen uit tumorweefsels en tumorcellijnen te isoleren en te identificeren. Deze test is handig, snel en kosteneffectief. Deze methode kan bijdragen aan een beter begrip van het effect van genen of andere extracellulaire en intracellulaire signalen op de stameigenschappen van tumorcellen.Veel factoren kunnen het percentage SP-cellen in deze analyse beïnvloeden, dus het is van cruciaal belang om de juiste omstandigheden te verkennen vóór het formele experiment. Zaadtumorcellen in een zes-put plaat, en incubeer ze in een 37 graden Celsius incubator geleverd met 5%kooldioxide. Wanneer de celdichtheid ongeveer 90% bereikt, aspireer dan het kweekmedium en was de cellen tweemaal met drie milliliter PBS.Voeg vervolgens 500 microliters van 0,25% trypsine-EDTA toe aan elke put en incubeer de plaat gedurende één tot drie minuten op 37 graden Celsius. Tik voorzichtig op de plaat om de cellen los te maken, voeg drie milliliter PBS aangevuld met 2% FBS toe aan elke put en pipetteer de cellen drie tot vijf keer op en neer om de celklonters te verspreiden. Onderzoek de celsuspensie onder de microscoop.Als er celklonters aanwezig zijn, laat u de suspensie door een zeef van 70 micrometer lopen. Breng de cellen over in een centrifugebuis van 15 milliliter en centrifugeer ze gedurende vijf minuten op 200 maal g. Verwijder het supernatant en resuspend de cellen in drie milliliter PBS aangevuld met 2%FBS, waarbij de cellen drie tot vijf keer op en neer worden geleid om te mengen.Tel cellen onder de microscoop met behulp van een hemocytometer en verdun ze in PBS aangevuld met 2%FPS tot een uiteindelijke concentratie van één keer 10 tot de zes cellen per milliliter. Bereid twee reageerbuizen met een ronde bodem van vijf milliliter, polystyreen, ronde bodem en voeg één milliliter van de celsuspensie toe aan elke buis. Label de ene buis als reageerbuis en de andere als blokkeerbuis.Bereid een oplossing van blokker voor door deze tot de juiste concentratie te verdunnen. Voeg het toe aan de blokker controlebuis en meng goed, incubeer de buis vervolgens op 37 graden Celsius gedurende 30 minuten. Schud de buis om de 10 minuten.Bereid een oplossing van Hoechst 33342 door deze tot de juiste concentratie te verdunnen. Voeg het apart toe aan de reageerbuis en blokkerregelbuis en meng goed en incubeer de buizen vervolgens gedurende 60 minuten op 37 graden Celsius. Schud de buizen om de 10 minuten.Centrifugeer de cellen na de incubatie gedurende vijf minuten bij 200 keer g en vier graden Celsius en aspireer vervolgens voorzichtig het supernatant. Resuspendeer de cellen in elke buis met een milliliter ijskoude PBS aangevuld met 2%FBS en pipetteer de cellen vervolgens op en neer om te mengen. Voeg één microliter propidiumjodide van één milligram per milliliter, of PI, toe aan elke buis en meng.Klik op het tabblad Parameters in de flowcytometersoftware. Kies eerst de parameters van respectievelijk FSC, SSC, Hoechst Blue, Hoechst Red en PI. Kies ten tweede de logaritmische schaal voor de PI-parameter.Kies ten slotte gebieds- en breedteschalen voor respectievelijk de parameters FSC, SSC, Hoechst Blue, Hoechst Red en PI. Voer de cellen eerst uit de reageerbuis en voer vervolgens de cellen uit de blokkeerbesturingsbuis met dezelfde spanningen en poorten. Verzamel 20 tot 100.000 gebeurtenissen uit elk monster om het percentage SP-cellen te analyseren.Deze methode werd gebruikt om het aandeel SP-cellen in de MDA-MB-231 menselijke borstkankercellijn te detecteren. De dot plot van FSC-A versus PI-A toonde een populatie van dode cellen, celresten en de belangrijkste populatie van PI negatieve cellen, die werd onderworpen aan verdere analyse. Een eencellige populatie die afkwam van het puntplot van FSC-A versus FSC-W en het puntplot van SSC-A versus SSC-W werd gebruikt om het aandeel SP-cellen te analyseren, dat ongeveer 0,9% was in onbehandelde cellen en ongeveer 0,09% in cellen die met reserpine werden behandeld.Verschillende kleuringsconcentraties van Hoechst 33342 werden getest op MDA-MB-435 cellen, en er werd vastgesteld dat twee microgram per milliliter optimaal was voor SP-analyse. Lage concentraties maakten het moeilijk om SP-cellen te onderscheiden van andere populaties, terwijl hoge concentraties ervoor zorgden dat SP-cellen verdwenen. Verapamil en reserpine werden getest om de juiste blokker voor de MDA-MB-435 cellen te bepalen.Ongeveer 0,4% van de cellen verdreef de kleurstof na de verapamil-behandeling, maar de verhouding daalde tot ongeveer 0,1% na reserpinebehandeling, wat aantoont dat reserpine een geschiktere blokker is voor deze cellijn. Dit protocol werd ook gebruikt om het aandeel SP-cellen te bepalen in A549 menselijke longadencarcinoomcellen behandeld met STAT3 activator colivelin en in T47D menselijke borstkankercellen behandeld met FRA1-remmer SKLB816. Deze test geeft aanwijzingen voor het regulerende effect van signaleringsroutes op de kenmerken van kankerstamcellen en vergemakkelijkt de ontdekking van nieuwe mechanismen, die uiteindelijk de gerichte therapie van tumoren kunnen begeleiden.
