がん幹細胞(CSC)は、自己再生能および複数の分化能を有する細胞のサブセットであり、腫瘍の増殖、転移、再発1、2において重要な役割を果たす。現在、CSCは、肺、脳、膵臓、前立腺、乳房、および肝臓癌3、4、5、6、7、8、9を含む様々な悪性腫瘍に存在することが確認されている。これらの腫瘍におけるCSCの同定は、主にCD44、CD24、CD133、およびSca-19、10の高低発現などの表面マーカータンパク質の存在に基づいているが、CSCと非CSCを区別できるユニークなマーカーは、これまで報告されていない。現在、腫瘍組織または腫瘍細胞株のCSCを同定し、精製するためにいくつかの技術が使用されている。これらの手法は、CSC の特定のプロパティに基づいて設計されています。中でも、アッセイおよびサイド集団(SP)細胞の並べ替えは、一般的に使用される方法の2つである。

がん幹細胞(CSC)は、腫瘍の増殖、転移、再発の重要な原因です。CSCの単離と同定は、腫瘍研究にとって非常に重要です。現在、腫瘍組織および腫瘍細胞株からのCSCの同定と精製にはいくつかの技術が用いられている。側母集団(SP)細胞の分離および分析は、一般的に使用される方法の2つである。これらの方法は、Hoechst 33342のような蛍光色素を迅速に排出するCSCの能力に依存します。色素の流出は、ATP結合カセット(ABC)トランスポーターに関連付けられており、ABCトランスポーター阻害剤によって阻害され得る。Hoechst 33342で培養腫瘍細胞を染色し、フローサイトメトリーによってそれらのSP細胞の割合を分析する方法が記載されている。このアッセイは、便利で、高速で、費用対効果が高い。このアッセイで生成されたデータは、腫瘍細胞の幹細胞性に対する遺伝子または他の細胞外および細胞内シグナルの影響をよりよく理解するのに役立つ。

側の集団細胞の分析は、腫瘍組織および腫瘍細胞株から癌幹細胞を単離し、同定するために一般的に使用される方法の1つである。このアッセイは、便利で、高速で、費用対効果が高い。この方法は、腫瘍細胞の幹細胞性に対する遺伝子または他の細胞外および細胞内シグナルの影響をよりよく理解するのに役立つ。この分析では、多くの要因がSP細胞の割合に影響を与える可能性があるため、正式な実験の前に適切な条件を調べるのが重要です。腫瘍細胞を6ウェルプレートに播種し、5%の二酸化炭素を供給する摂氏37度のインキュベーターでインキュベートする。細胞密度が培地を約90%吸引すると、3ミリリットルのPBSで細胞を2回洗浄した。その後、0.25%トリプシン-EDTAの500マイクロリットルを各ウェルに加え、37°Cでプレートを1〜3分間インキュベートします。プレートを軽くタップして細胞を取り外し、各ウェルに2%FBSを加えたPBSを3ミリリットル加え、細胞を上下に3~5回ピペットして細胞塊を分散させます。細胞懸濁液を顕微鏡で調べます。細胞の塊が存在する場合は、70マイクロメートルのストレーナーを通して懸濁液を通過する。細胞を15ミリリットルの遠心分離管に移し、200倍gで5分間遠心分離する。上清を取り除き、2%FBSを補充したPBSの3ミリリットルで細胞を再懸濁し、細胞を上下に3〜5回ピペットして混合する。ヘモサイトメーターを用いて顕微鏡下で細胞を数え、2%FPSを加えたPBSで希釈し、1ミリリットル当たり6個の細胞に10倍の最終濃度にする。5ミリリットルのポリスチレン、ラウンドボトム試験管を2本用意し、各チューブに1ミリリットルの細胞懸濁液を加えます。一方のチューブを試験管として、もう一方のチューブにブロッカーコントロールチューブとしてラベルを付けます。適切な濃度に希釈することにより、ブロッカーの溶液を調製.ブロッカーコントロールチューブに加えてよく混ぜ、37°Cで30分間チューブをインキュベートします。チューブを10分おきに振ります。適切な濃度に希釈してHoechst 33342の溶液を調製する。試験管とブロッカーコントロールチューブに別々に加え、よく混ぜ、摂氏37度で60分間インキュベートします。10分ごとにチューブを振ります。インキュベーション後、細胞を200倍gと摂氏4度で5分間遠心分離し、上清を慎重に吸引する。2%FBSを補った氷冷PBSの1ミリリットルで各チューブ内の細胞を再懸濁し、その後、細胞を上下にピペットして混合します。各チューブに1ミリグラム/ヨウ化プロピジウム(PI)を1マイクロリットル加え、混合します。フローサイトメーターソフトウェアの[パラメータ]タブをクリックします。まず、FSC、SSC、ホークストブルー、ホーヒストレッド、PIのパラメータをそれぞれ選択します。次に、PI パラメータの対数スケールを選択します。最後に、FSC、SSC、Hoechst ブルー、ホークスト レッド、PI パラメータの面積と幅のスケールをそれぞれ選択します。まず、試験管からセルを実行し、次に同じ電圧とゲートを使用してブロッカー制御チューブからセルを実行します。各サンプルから20~100,000個のイベントを収集し、SPセルの割合を分析します。この方法は、MDA-MB-231ヒト乳癌細胞株におけるSP細胞の割合を検出するために使用した。FSC-A対PI-Aのドットプロットは、死んだ細胞、細胞デブリ、およびPI陰性細胞の主な集団の集団を示し、さらなる分析を行った。FSC-A対FSC-WのドットプロットとSSC-A対SSC-Wのドットプロットからゲートされた単一の細胞集団は、未治療細胞で約0.9%、レセルピンで処理された細胞で約0.09%であったSP細胞の割合を分析するために使用された。MDA-MB-435細胞で異なる染色濃度のHoechst 33342を試験し、1ミリリットル当たり2マイクログラムがSP分析に最適であると判断した。低濃度ではSP細胞を他の集団と区別することが困難となり、高濃度ではSP細胞が消失しました。ベラパミルおよびレセルピンはMDA-MB-435細胞に対する適切なブロッカーを決定するために試験した。約0.4%の細胞がベラパミル処理後に色素を排出したが、レセルピン処理後に約0.1%に低下し、レセルピンがこの細胞株にとってより適切なブロッカーであることを示した。このプロトコルは、STAT3アクチベーターコリブリンで治療されたA549ヒト肺腺癌細胞およびFRA1阻害剤SKLB816で治療されたT47Dヒト乳癌細胞におけるSP細胞の割合を決定するためにも使用された。このアッセイは、がん幹細胞の特徴に対するシグナル伝達経路の調節効果の手がかりを提供し、最終的に腫瘍の標的療法を導くことができる新しいメカニズムの発見を容易にする。
