최근에는 GPCR 활성화를 모니터링하기 위한 라벨없는 검정법이 등장하고 있으며, 임피던스 기반 기술 또는 공진 도파관격자를적용하는 광학 적 방법 등5,6. 세포의 라벨링은 실험적으로 이러한 접근법에 필요하지 않습니다. 이러한 물리적 판독은 낮은 진폭 신호에서 작동하므로, 이러한 방법은 비침습적으로 간주되며, 잠재적으로 실시간으로 연속 적인 세포 모니터링을 허용하고 관찰 시간은 판독이 아닌 셀 배양에 의해서만 제한된다. 전체 기관에서 판독과 유사, 라벨없는 접근은 일반적으로 전체적인 세포 응답에 보고, 전체 신호 네트워크를 따라 통합 할 때 수용체 활성화의 멀리 하류는 시간 의존하지만 오히려 비특이적 인 변화로 이어질 세포 형태 또는 대량 재분배. 임피던스 계 측정은 세포 형상7,8의변화의 유전체 서명을 측정하는 반면, 공진 도파관 격자를 이용한 측정은 동적 질량 재분배(DMR) 9에서발생하는 세포 기판 계면에서굴절률의 변화에 민감하다. 통합 문자는 G 단백질의 유형 (Gq,Gi/0,Gs, G12/13)또는 β-arrestin 신호 캐스케이드6에 관여하는 수용체 매개 사건에 관계없이 매우 민감하고 수용체의 내인성 발현 수준에 매우 적합한 무첨가 방법을 만든다.
표준 라벨 프리 임피던스 계 분석에서 세포는 각 웰10의바닥에 증착된 코플라나 금막 전극을 가진 다중 웰 플레이트에서 부착적으로 성장한다. 이러한 전극 어레이는 임피던스 분석기와 연결되어 있으며 실험 자극에 대한 셀 응답은 시간 해결된 임피던스 판독값에 의해 개별 우물에서 기록됩니다. 전형적인 GPCR 분석에서 리간드가 각 개인에게 개별적으로 상이한 농도로 잘 첨가된다. 임피던스 시간 코스에서 리간드-유도 변화는 리간드의 효능 및 효능을 정량화하기 위해 최대 신호 변화, 곡선 아래 영역, 지정된 시간 간격 또는 곡선의 기울기 내의 신호 변화와 같은 특성 곡선 특징에 대하여 분석된다(11).
