In deze studie presenteren we een betrouwbare methode voor de isolatie, cultuur en adipogene inductie van NCADSCs uit de PVAT van Wnt-1 Cre+/-; Rosa26RFP/+ transgene muizen ontworpen om RFP+ ADSCs te produceren. De prevalentie van obesitas neemt wereldwijd toe, wat het risico op gerelateerde chronische ziekten, waaronder hart- en vaatziekten en diabetes1, verhoogt. PVAT omringt bloedvaten en is een belangrijke bron van endocriene en paracrine factoren die betrokken zijn bij vasculatuur functie. Klinische studies tonen aan dat een hoog PVAT-gehalte een onafhankelijke risicofactor is van hart- en vaatziekten2,3, en de pathologische functie ervan hangt af van het fenotype van de van bestanddelen afgeleide stamcellen (ADSCs)4.
Hoewel ADSC-cellijnen zoals de murine 3T3-L1, 3T3-F442A en OP9 nuttige cellulaire modellen zijn om adipogenese of lipogenese5te bestuderen, verschillen de regulerende mechanismen voor adipogenese tussen cellijnen en primaire cellen. De ADSCs in de stromal vasculaire celfractie (SVF) rechtstreeks geïsoleerd van vetweefsel en ertoe aangezet om zich te onderscheiden in adipocyten waarschijnlijk samenvatten in vivo adipogenese en lipogenese6. Echter, de kwetsbaarheid, drijfvermogen, en de variaties in grootte en immunophenotypes van de ADSCs maken hun directe isolatie uitdagend. Bovendien kunnen de verschillende isolatieprocedures ook een aanzienlijke invloed hebben op het fenotype en het adipogene potentieel van deze cellen7,waardoor de nadruk wordt gelegd op de noodzaak van een protocol dat de ADSC-integriteit handhaaft.
Vetweefsel wordt meestal geclassificeerd als ofwel de morfologische en functioneel onderscheiden wit vetweefsel (WAT), of de bruine vetweefsel (BBT)8, die verschillende ADSCs herbergt9. Terwijl ADSCs geïsoleerd van perigonadale en inguinal onderhuidse WAT's zijn gekenmerkt in eerdere studies9,10,11,12, minder bekend is met betrekking tot de ADSCs van PVAT die voornamelijk bestaat uit BAT13.
In een recente studie, vonden we dat een deel van de ingezetene ADSCs in de periaortische boog vetweefsel (PAAT) zijn afgeleid van neurale crest cellen (NCCs), een voorbijgaande populatie van migrerende voorlopercellen die afkomstig zijn van de ectoderm14,15. Wnt1-Cre transgene muizen werden gebruikt voor het traceren van neurale crest cel ontwikkeling16,17. We staken Wnt1-Cre+ muizen over met Rosa26RFP/+ muizen om Wnt-1 Cre+/-te genereren; Rosa26RFP/+ muizen, waarin NCC's en hun nakomelingen zijn gelabeld met rood fluorescerend eiwit (RFP) en gemakkelijk in vivo en in vitro15worden bijgehouden. Hier beschrijven we een methode voor het isoleren van neurale kam afgeleide ADSCs (NC-afgeleide ADSCs, of NCADSCs) van muis PAAT en induceren de NCADSCs om zich te onderscheiden in witte adipocyten of bruine adipocyten.

isolation, culture, and adipogenic induction of neural crest original adipose-derived stem cells from periaortic adipose tissue

Een overmatige hoeveelheid vetweefsel rond de bloedvaten (perivasculair vetweefsel, ook bekend als PVAT) wordt geassocieerd met een hoog risico op hart- en vaatziekten. Adscs afgeleid van verschillende vetweefsel tonen verschillende kenmerken, en die van de PVAT zijn niet goed gekarakteriseerd. In een recente studie, rapporteerden we dat sommige ADSCs in de periaortische boog vetweefsel (PAAT) afstammen van de neurale crest cellen (NCCs), een voorbijgaande populatie van migrerende cellen afkomstig uit het ectoderm.
In dit artikel beschrijven we een protocol voor het isoleren van rode fluorescerende eiwitten (RFP)-gelabelde NCCs van de PAAT van Wnt-1 Cre+/-; Rosa26RFP/+ muizen en het opwekken van hun adipogene differentiatie in vitro. Kortom, de stromal vasculaire fractie (SVF) is enzymatisch gescheiden van de PAAT, en de RFP+ neuralcrest afgeleide ADSCs (NCADSCs) worden geïsoleerd door fluorescentie geactiveerde cel sortering (FACS). De NCADSCs onderscheiden zich in zowel bruine als witte adipocyten, kunnen worden cryopreserved, en behouden hun adipogene potentieel voor ~ 3-5 passages. Ons protocol kan genereren overvloedige ADSCs uit de PVAT voor het modelleren pvat adipogenese of lipogenese in vitro. Zo kunnen deze NCADSCs een waardevol systeem bieden voor het bestuderen van de moleculaire schakelaars die betrokken zijn bij PVAT-differentiatie.

Met behulp van het hierboven beschreven protocol hebben we ~0,5–1,0 x 106 ADSCs van 5-6 Wnt-1 Cre+/-verkregen; Rosa26RFP/+ muizen (48 weken oud, mannelijk of vrouwelijk).

Isolation, Culture, and Adipogenic Induction of Neural Crest Original Adipose-Derived Stem Cells from Periaortic Adipose Tissue

We presenteren een protocol voor de isolatie, cultuur en adipogene inductie van neurale kam afgeleide vet-afgeleide stamcellen (NCADSCs) uit de periaortische vetweefsel van Wnt-1 Cre+/-; Rosa26RFP/+ muizen. De NCADSCs kunnen een gemakkelijk toegankelijke bron van ADSCs voor het modelleren van adipogenese of lipogenese in vitro.

Onze eenvoudige en praktische methodologie kan worden gebruikt om overvloedige ADSCS te verkrijgen voor het bestuderen van PVAT adipogenese in vitro en om normale geneesmiddelen te testen tegen obesitas en gerelateerde hart- en vaatziekten. Het voordeel van deze methode is de inherente fluorescentie van de NCADSCs van biotine die u nodig hebt om de cellen te labelen met geconjugeerde antilichamen met flou-chroom voor FACS, of magnetische geactiveerde celsorde. Deze methode kan worden gebruikt om overvloedige ADSC afgeleid van neurale kuifcellen uit het ectoderm te verwerven om PVAT adipogenese, of lipogenese in vitro te bestuderen.Het gebruik van jonge muizen en het hebben van ervaring met cautery en geïnduceerde adipogenese betekent dat 3T3-cellen cruciaal zijn voor het succes van dit protocol. Het aantonen van de procedure zal Yiding Qi, een grad student, en Lian Xu, een technicus van mijn laboratorium. Voor PAAT dissectie, na het steriliseren van de muis karkas in 75%ethanol gedurende vijf minuten, gebruik schaar te knippen en scheiden van de huid op de buik.Snijd langs de ventrale middellijn van bekken tot de nek, en open de buik. Verplaats de lever om het middenrif bloot te leggen, en snijd het middenrif en de ribben aan beide zijden van de middellijn. Pel de ribben terug om het hart en de longen bloot te leggen.En verwijder de longen en de thymus. Haal de PAAT samen met de aorta en het hart in een petrischaaltje. En snijd de aorta af bij de aortawortel om het hart te verwijderen.Maak vervolgens een snede tussen de aortaboog en dalende aorta. Zorgvuldig scheiden van het vetweefsel rond beide structuren, en de linker en rechter gemeenschappelijke halsslagader van de achterste borstwand. Het is belangrijk om de PAAT van de wortel te scheiden en als een grotere vasculaire vaten zorgvuldig, is de aanwezigheid van vasculaire wandcellen kan facs efficiëntie beïnvloeden.Plaats het weefsel in een petrischaaltje met ijskoude HBSS-buffer. En gebruik steriele tangen om zo veel van de vasculatuur en fascia mogelijk te verwijderen. Breng het vetweefsel vervolgens over in een twee milliliter Eppendorf-buis, met punt vijf milliliter ijskoude HBSS-buffer op ijs.Wanneer alle PAAT is verzameld, breng het geoogste vetweefsel over in een nieuwe twee milliliter microcentrifugebuis met een milliliter vers bereid digestiemedium. En gebruik een chirurgische schaar om de weefsels te gehakt. Breng de weefseldrijfmest over in een 50 millimeter buis met negen milliliter vers spijsverteringsmedium.En gebruik een 1 milliliter pipet om de weefsels 10 keer te tritureren. Wanneer een homogene oplossing is verkregen, incuberen de buis gedurende 30 tot 45 minuten bij 37 graden Celsius en 100 omwentelingen per minuut, controle om de vijf tot 10 minuten om overdigestie te voorkomen. Wanneer een lichtgele homogene vetweefseloplossing kan worden waargenomen bij zachte werveling van de buis, stop de spijsvertering met vijf milliliter HDMEM aangevuld met 10% foetaal runderserum.Na een grondige menging, centrifugeren het vetweefsel monster. De stromale vasculaire celfractie zal zichtbaar zijn als een bruinachtige pellet. Schors de pellet opnieuw in 10 milliliter cultuurmedium en filter de celsuspensie door een celzeef van 70 micrometer.Verzamel de cellen door een andere centrifugatie. En voorzichtig opnieuw op te schorten de pellet in vijf milliliter van erythrocyten lyse buffer. Breng de ophanging over op een buis van 15 milliliter.Na 10 minuten stopt u de reactie met twee centrifugaties in 10 milliliter PBS, aangevuld met 1%foetaal runderserum. Na de tweede centrifugatie, opnieuw opschorten de pellet in vijf milliliter van cultuur medium op ijs. En verzamel de cellen met een laatste centrifugatie.Dan, opnieuw opschorten van de cellen in vijf milliliter facs buffer voor het tellen. Het opzetten van een NCADSC cultuur, na hun isolatie door FACS volgens standaard protocollen, plaats de cellen op een vijf keer 10 tot de derde cellen per centimeters kwadraat concentratie in elke put, een twaalf put cultuur plaat. Onvolledig kweekmedium bij 37 graden Celsius en 5% kooldioxide gedurende 20 tot 24 uur.De volgende dag, was de cellen met 37 graden Celsius PBS, en voeden de cultuur met verse cultuur medium. Wanneer de cultuur bereikt 80 tot 90%samenvloeiing, behandel de cellen met twee milliliter van 0,25% trypsine EDTA per goed gedurende drie tot vijf minuten bij 37 graden Celsius. Wanneer de cellen zijn losgemaakt van de bodem van de plaat, neutraliseren van de enzymen met twee milliliter gekweekt medium, en het verzamelen van de geoogste cellen door centrifugatie.Schort de pellet opnieuw op in een milliliter vers kweekmedium om te tellen. En zaad de cellen in een nieuwe 12 goed kweekplaat in een vijf keer 10 tot de derde cellen per centimeter in vierkante concentratie. Dan, opnieuw opschorten de resterende cellen in cultuurmedium aangevuld met 10%dimethylsulfoxide voor bevroren opslag in vloeibare stikstof.Om adipogene differentiatie te induceren, wanneer de cellen een samenvloeiing van 80 tot 90% bereiken, behandelen de cellen met bruin of wit adipogeen inductiemedium gedurende twee dagen, naar gelang van het geval. Aan het einde van de incubatie, voorzichtig wassen de cellen twee keer met PBS voor het toevoegen van de juiste verse adipogene inductie medium aan elke put, en de terugkeer van de cellen naar de celcultuur incubator voor een extra drie tot vijf dagen van incubatie. Gekweekte NCADSCs bereiken een 80 tot 90% samenvloeiing na zeven tot acht dagen van cultuur.En een uitgebreid glasvezel zoals morfologie demonstreren. Om hun adipogene potentieel verder te bevestigen, kan olierode kleuring van de gedifferentieerde NCADSCs worden uitgevoerd om volwassen adipocyten op te sporen. Typisch, volwassen adipocyten worden waargenomen na acht dagen van witte of bruine adipogene inductie met meer dan 60% van de NCADSCs vertonen adipogene differentiatie.Merk op dat NCADSCs een sterk verminderd adipogeen potentieel na passaging aantonen. Immunoblotting en kwantitatieve real-time PCR blijkt dat de expressie van adipocyten specifieke relatieve eiwitten en genen aanzienlijk toeneemt in adipogenisch gedifferentieerde NCADSCs na acht dagen van witte adipogene inductie. Ook de expressie van adipocyten specifieke genen, en bruine adipocyten specifieke genen, neemt ook aanzienlijk toe na acht dagen van bruine adipogene inductie.Aangezien de meeste NCADSC isolatie protocol is eenvoudig, economisch, en vereist niet het gebruik van antilichamen, voor dat, de sterke vloeistof redenen intensiteit van IFP kan de FACS voeding efficiëntie te verbeteren.
