In dit protocol wordt CCl4 gebruikt als levertoxine om ALI bij ratten op te wekken. ALI wordt gekenmerkt door verlies van hepatische parenchyma en daaropvolgende dysregulatie van de metabolische en synthetische functies van de lever. Geneesmiddelen, virussen, toxines, auto-immuunziekten, metabole ziekten en vasculaire aandoeningen veroorzaken allemaal hepatocyte dood, en de daaropvolgende ontstekingsreactie draagt bij aan de pathogenese van ALI.
De eerste belediging van de lever lei...

De frequentie van giftige beledigingen aan de lever, vooral als gevolg van alcohol- en drugsmisbruik, neemt toe. Acuut leverletsel (ALI) wordt geassocieerd met hoge sterftecijfers en heeft klinische problemen veroorzaakt1,2. Giftig letsel leidt tot dodelijke seinen trajecten in de lever, wat resulteert in hepatocyte apoptose, necrose, of pyroptose. ALI speelt een cruciale rol in de ontwikkeling van leverfalen, dat wordt gekenmerkt door ernstige leverdisfunctie, waaronder complicaties zoals hepatische encefalopathie en verminderde eiwitsynthese3,4. Hoewel recent onderzoek onze kennis over de fysiologische en pathologische veranderingen bij leverfalen heeft vergroot, heeft het de pathomoleculaire kenmerken die de mechanismen van celdood beïnvloeden niet volledig verklaard. Bovendien zijn er momenteel geen medicijnen beschikbaar om de progressieve verslechtering van ALI-patiënten om te keren. Momenteel is de enige aanzienlijk effectieve behandeling levertransplantatie5,6.
Om het mechanisme en de pathofysiologie van ALI te onderzoeken en verschillende hepatoprotectiestrategieën te testen, worden verschillende diermodellen gebruikt om ALI te induceren. Een voorkeursdiermodel van ALI moet het pathologische proces van de ziekte nabootsen via een betrouwbare, gevalideerde, goedkope en gemakkelijk toe te passen methode. Voorbeelden van experimentele modellen zijn hepatotoxische middelen, chirurgische ingrepen zoals totale of gedeeltelijke hepatectomie, volledige of voorbijgaande devascularisatie, en infectieve procedures7,8,9. Bekende hepatotoxische stoffen zijn galactosamine, paracetamol, thioacetamide, azoxymethaan en CCl4. Van deze, CCl4 wordt veel gebruikt, hoewel het nog niet goed is gekenmerkt10,11,12,13.
CCl4 is een organische kleurloze vloeibare verbinding met een zoete geur en bijna geen ontvlambaarheid bij lagere temperaturen. Blootstelling aan hoge concentraties CCl4 kan schade aan het centrale zenuwstelsel veroorzaken, waaronder verslechtering van de lever en nieren. CCl4 induceert ALI door zijn biotransformatie in de lever, die reactieve zuurstofsoorten vormt. Dit gebeurt via het P450 cytochroomenzym 2E1, dat een actieve metaboliet vormt en resulteert in celschade door macromolecuulbinding, verbetering van lipideperoxidatie en verstoring van intracellulaire calciumhomeostase14. Daarnaast kan het CCl4 model worden gebruikt om de astrocyten te stimuleren op het niveau van RNA synthese15. Dit hepatotoxine is toegediend door de intraperitoneale, intraportal, orale en intragastrische routes16.
In dit protocol beschrijven we in detail CCl4-geïnduceerde ALI bij ratten via een orogastrische buis. Deze methode induceert robuuste en reproduceerbare ALI die kan worden gebruikt om de pathogenese van ALI te onderzoeken. De bepaling van de ernst van de leverziekte wordt gecontroleerd door meting van serumglutamaat-pyruvaattransaminase (GPT), glutamic oxaloacetische transaminase (GOT) enzymen en totale bilirubine (TB) evenals definitieve histologische diagnose door hematoxylinen en eosine (H&E) gekleurde leverweefsels. Blootstelling aan CCl4 via een intragastrische toegang zorgt voor een praktische, goedkope, minimaal invasieve methode met minimaal risicorisico.

Acute leverschade (ALI) speelt een cruciale rol in de ontwikkeling van leverfalen, dat wordt gekenmerkt door ernstige leverdisfunctie, waaronder complicaties zoals hepatische encefalopathie en verminderde eiwitsynthese. Geschikte diermodellen zijn van vitaal belang om het mechanisme en de pathofysiologie van ALI te testen en verschillende hepatoprotectiestrategieën te onderzoeken. Vanwege zijn vermogen om chemische transformaties uit te voeren, wordt koolstoftetrachloride (CCl4)op grote schaal gebruikt in de lever om ALI te induceren door de vorming van reactieve zuurstofsoorten. CCl4 blootstelling kan intraperitoneally worden uitgevoerd, door inademing, of via een nasogastrische of orogastrische buis. Deze methode is goedkoop, gemakkelijk te uitgevoerd, en heeft een minimaal risico risico. Het model is zeer reproduceerbaar en kan op grote schaal worden gebruikt om de werkzaamheid van potentiële hepatoprotectieve strategieën te bepalen en markers van leverletsel te beoordelen.

De TB, GOT, en GPT niveaus aanzienlijk toegenomen 24 uur na het induceren van ALI (meer bij hogere CCl4 doses) in vergelijking met sham-operated controles (p < 0,001) (Figuur 1). Dit protocol beschrijft een gemeenschappelijke en haalbare methode voor het induceren van acute leverschade (ALI) via CCl4 blootstelling via een orogastrische buis. Ik wil ons werk presenteren over geïnduceerde acute leverletsel bij ratten via blootstelling aan koolstoftetrachloride via orogastrische buis. Selecteer volwassen mannelijke Sprague-Dawley ratten met een gewicht van 300 tot 350 gram. Zorg voor rat chow en water ad libitum.Voer alle experimenten uit tussen zes uur 's zes en 12 uur. Onderzoek van leverenzymen en het totale bilirubineniveau werd uitgevoerd in het biochemisch laboratorium van het Soroka Medical Center.Let op: blootstelling aan een hoge concentratie koolstoftetrachloride, waaronder damp of huidabsorptie, kan het centrale zenuwstelsel aantasten, de lever en de nieren degenereren en langdurige blootstelling kan leiden tot coma of de dood. Bereid een 50 procent oplossing van koolstoftetrachloride gemengd met olijfolie als voertuig in een op een verhouding. Induceer de pathotoxiciteit in vivo door koolstoftetrachloride toediening via een orogastrische buis.Steek een orogastrische buis door de mondholte van de rat. Stel de ratten bloot aan verschillende doses koolstoftetrachloride. Een milliliter per kilo voor lichte acute leverletsel, twee punt vijf milliliter per kilo voor matige acute leverletsel, of vijf milliliter per kilo voor ernstige acute leverletsel.Voor sham groep, bloot stellen de ratten tot vijf milliliter per kilo alleen olijfolie. Verzamel bloedmonsters op 24 uur van blootstelling aan koolstoftetrachloride. Bloed biochemische analyse omvatten de meting van serum GOT, GPT, en totale bilirubine.Plaats dode rat op ontleden boord in dorsale recombinant met de buik naar boven gericht en scheren haar buik. Met behulp van scalpel, insnijden de volledige lengte van het ventrum van de anus naar de kin. Reflecteer de huid.Met de scalpel, insnijden de buikwand van de anus naar de xiphoid kraakbeen om de buikwandcera bloot. Breng de lever in een petrischaaltje. Voer een hematoxyline en eosine kleuring.Bestudeer de plakjes onder microscoop vergroting van 200 keer met een 20 millimeter objectieve lengte. De leversectie moet worden beoordeeld door een gespecialiseerde patholoog, verblind door het behandelprotocol. De score van nul geeft geen leverafwijkingen aan, een tot twee duidt op lichte leverletsel, drie tot vier duidt op matig leverletsel en vijf tot zes wijst op ernstig leverletsel.De verschillende groepen ratten op verschillende tijdstippen worden op de regeling weergegeven. 24 uur na het opwekken van acuut leverletsel, was er een aanzienlijke toename van got, GPT, en de totale bilirubine niveaus, afhankelijk van de koolstof tetrachloride blootgesteld in vergelijking met de controlegroep. P-waarde was minder dan nul punt nul nul een.Histopathologische veranderingen werden na 24 uur in alle groepen waargenomen na koolstoftetrachlorideintoxicatie in verschillende doses. De histologische cijferscore van 24 uur tussen alle drie de interventiegroepen naar de controlegroep was aanzienlijk hoger. P-waarde was minder dan nul punt nul vijf.Onze onderzoeksgroep presenteerde de gemeenschappelijke en fysische methode van geïnduceerde acute leverletsel bij ratten via blootstelling aan koolstoftetrachloride via orogastrische buis. Deze methode wordt gebruikt om de pathofysiologie van acute leverletsel te onderzoeken en om anders te testen over de beschermende strategie. Hartelijk dank voor uw aandacht.
