肝臓に対する毒性侮辱の頻度は、特にアルコールや薬物乱用によるもので、増加している。急性肝障害(ALI)は高い死亡率に関連しており、臨床的な懸念を引き起こしている1,2。毒性傷害は肝臓の死シグナル伝達経路につながり、肝細胞アポトーシス、壊死、またはピロプトーシスを引き起こします。ALIは肝不全の発症において重要な役割を果たしており、肝性脳症やタンパク質合成障害などの合併症を含む重度の肝機能障害を特徴とする33,4。4最近の研究では、肝不全に伴う生理学的および病理学的変化に関する知識が増えているが、細胞死のメカニズムに影響を与える病的分子の特徴を完全に説明しているわけではない。さらに、ALI患者の進行性悪化を逆転させる薬は現在のところ利用できない。現在、唯一の有意に有効な治療法は、肝臓移植55,66である。
ALIのメカニズムと病態生理を調査し、異なる肝保護戦略をテストするために、異なる動物モデルがALIを誘導するために使用される。ALIの好ましい動物モデルは、信頼性の高い、検証済み、安価で適用しやすい方法を介して疾患の病理学的プロセスを模倣すべきである。実験モデルの例としては、肝毒性剤、全肝切除術などの外科的処置、完全または一過性の脱血管検査、および感染性処置77、8、98,9が含まれる。既知の肝毒性物質は、ガラクトサミン、アセトアミノフェン、チオアセアセタミド、アゾキシメタンおよびCCl4を含む。このうち、CCl4は、10、11、12、13,11,12をまだ十分に特徴づけられていないが広く用いられている13。4
CCl4は、甘い香りと低い温度でほとんど燃焼性のない有機無色液体化合物です。CCl4の高濃度への暴露は、肝臓および腎臓の悪化を含む中枢神経系への損傷を引き起こす可能性がある。CCl4は、活性酸素種を形成する肝臓での生体変換を通じてALIを誘導する。これは、P450シトクローム酵素2E1を介して起こり、活性代謝産物を形成し、高分子結合による細胞損傷をもたらし、脂質過酸化の増強および細胞内カルシウム恒常性14の障害をもたらす。また、CCl4モデル4は、RNA合成15のレベルでアストロサイトを刺激するために使用することができる。この肝毒素は、腹腔内、門内、口腔内、および胃内経路16によって投与されている。
