本報告では、親油性色素のマイクロピペット媒介送を用いて胚における運動ニューロンの逆行標識を可能にする技術を提示する。この技術により、産卵後15時間で30個の体壁筋肉のそれぞれを15時間でヘミセグメントにインジェインする38個の運動ニューロンをトレースすることができます(AEL)14.この技術を用いることで、当社のグループは多数の機能獲得/機能喪失アレス15、16、17を徹底的に調査しました。我々は最近、運動デンドライト接続の開始を駆動する分子機構を解明し、Dscam1-Dock-Pak相互作用がaCC運動ニューロン17における樹状突起伸の部位を定義することを実証した。一般に、この技術は、野生型または変異型株における任意の胚性運動ニューロンの表現型解析に適応可能であり、ショウジョウバエ神経系の機能設計に関する新しい洞察を提供する能力を高める。

このプロトコルで説明されているトレース技術は、神経形態形成を研究し、ショウジョウバエの運動ニューロン系における接続性をスナップするのに役立ちます。親油性染料は、非常に高密度の細胞膜のあらゆる部分に急速に拡散する可能性があります。このため、私たちのプロトコルは、ラベル付きニューロンの構造を見つけるために効率的に解決します。軸索末端が注射マイクロピペットでアクセス可能な場合、この技術はハエの幼虫および成体段階または他の生物の任意のニューロンに適用することができる。解剖やマイクロインジェクターを使用した経験がない場合は、試料の操作を正確に行うことは困難です。胚の解剖学を理解することは、解剖または注射を行うためのツールの適切な部分へのガイダンスに役立つ。開始するには、すべての胚収集材料を準備し、レイアウトします。50ミリリットルのチューブを切断し、キャップに穴を開けて切断して濾過装置を準備します。次に、チューブとキャップの間に100マイクロメートルの細孔を入れてメッシュフィルターをセットします。同じキャピラリーチューブから、内径1.2ミリメートルの色素注入マイクロピペットと解剖針を準備します。マイクロピペットでチューブを引っ張り、170ボルトの最大出力から7%の引き手で、長さが約0.4センチメートルのテーパーで鋭い針を作ります。染料注入マイクロピペットを調製するには、粉砕機に湿潤剤を浸し、針をマイクロピペットクランプに25~30度置きます。次に、先端をベベルサーフェスの中心から半径の3分の2に下げます。チューブを持つ注射器が空気を押し込んでいる間、針を粉砕します。マイクロピペットに細かい先端の永久マーカーを付けて、斜めで形成された狭い開口部を見つけるのが難しい場合があるため、ベベル後の先端の開口部の位置を示します。ハエが室温で一晩卵を産み、午前中に卵と一緒にパイプを集めます。胚を収集するには、5分間50%漂白剤でプレート上の卵をデクロリン化します。塩素がクリアされたら、濾過装置または細胞ストレーナーを通してプレートの内容物を注ぎ、胚を単離する。水の絞りボトルを使用してプレートに残された漂白剤を希釈し、混合物をフィルターにデカンスしてできるだけ多くの胚を集めます。漂白剤の臭いが消えるまで、胚を水で3~4回洗います。装置からフィルターを取り出し、水で別のきれいなプレートに洗います。その後、胚が置かれている新しいプレートから水をデカントします。産卵後15時間で5~10個の胚を選択し、後側を上に向けた両面テープに貼ります。脱剖プールに昆虫リンガー生理を加え、胚を乾燥から保護します。解剖顕微鏡の下でガラス針を使用して、中途半端な膜からガラスに胚を引きずり出し、胚の内部組織に損傷を与えないのに注意してください。その後、後部から前端まで表面の単一の胚の中間線を切断します。上皮組織を中央から反転させ、表皮の縁をガラススライドの表面に取り付けます。約300マイクロメートルの先端開口部を持つチューブ接続された針を使用して吸引または空気を吹き飛ばし、後部縦方向の気管幹と残りの腸を取り除きます。4%PFAとPBSを使用して、軌道シェーカーの室温で5分間胚を固定します。その後、PBSでそれらを3回洗います。シアニン3色素と共役した抗西洋ワサビペルオキシダーゼ抗体1マイクロリットル、軌道シェーカーで1時間PBSの200マイクロリットルで胚を染色する。そして、PBSでのすきを繰り返します。注射マイクロピペットを充填するには、毛細管ホルダーに入れる。次に、染料スライドをステージに配置し、マイクロマニピュレータを使用してスライドの上に配置します。次に、ステージを調整して、マイクロピペットを染料の上に置きます。注入圧力を200~500ヘクトパスカル、0.1~0.5秒の射出時間、補償圧力を5分間ゼロにして、マイクロピペットに染料を回収します。染料を採取したら、色素スライドを取り出し、サンプルを顕微鏡ステージに置きます。その後、補償圧力を30〜60ヘクトパスカルの範囲に上げ、マイクロピペットをサンプルに下げます。10倍の対物レンズを使用して胚を見つけ、マイクロピペットを胚と位置合わせします。対物レンズを水浸漬40回レンズに変更します。そして、PBSにレンズを水没.蛍光顕微鏡を使用して、抗HRP Cy3によってマークされた神経形態をチェックし、注射部位を決定します。胚が焦点を合わせるとき、目的の軸索の先端と穏やかに接触するようにマイクロピペットの位置を変更する。次に、注射中に明視野顕微鏡を使用して、染料滴を見ます。diDまたはDiOのいずれかでaCCまたはRP3の神経筋接合部で右腹部のヘミセグメントに染料をドロップします。手制御を使用して染料を放出し、マイクロピペットを取り除きます。次の注射部位に移動します。イメージングの前に軌道シェーカーの室温で1時間サンプルをインキュベートします。細かい鉗子を使用して、ガラススライドからテープを取り出します。サンプルを取り付けるには、4つのコーナーに少量の真空グリースを入れてカバースリップを用意します。慎重にサンプルの上に置き、気泡を避けるようにしてください。カバースリップを押し下げて、サンプルのスペースを調整して、対物レンズとスライドの間の作業距離を調整します。タスクワイプで余分なPBSを取り除きます。カバースリップの端をマニキュアで完全に密封します。共焦点顕微鏡で10倍と100倍の倍率で画像を画像Jソフトウェアで処理し、画像を処理します。このプロトコルは、aCC運動ニューロンインナーバディング筋肉1とRP3運動ニューロンインナーバディング筋肉6と7のラベルを逆行するために使用されました。ACCおよびRP3運動ニューロンからの樹状枝は、広範な重複を示す。両方のニューロンは双極性であり、樹状突起の2つの異なる集団を確立する。この方法の定量的能力を実証するために、樹状突起先端の総数を野生型およびDscam1変異体で数えた。aCCのイプシララルデンドライトは野生型に対して示されているが、Dscam1変異体に対しては少数のイピシアラショナルデンドライトが観察された。この手順を試みる際には、約10〜20マイクロメートルの色素滴の大きさが重要であることを覚えておくことが重要です。両面テープのサイズをテストし、注射部位に適用します。この手順を用いて、DiDプローブのフォト切り替え可能な性質を利用して、プラズマ膜の超解像度画像を得ることができます。このようにして、神経筋接合部におけるシナプス膜の超構造的特徴を研究することができます。私たちはクリックをしました。変異株中のaCCデンドライトのフェロチ分析を行い、Dscam1-Dock-Pak注入がaCCにおける樹状突起の部位を定義することを発見した。
