FDAはCD3を標的とする最初のモノクローナル抗体を承認しました (OKT3, ムロモナブ)1,2.それ以来、次の20年間、免疫チェックポイント阻害剤3に対する抗体の圧倒的な成功により、抗体工学の分野で急速な爆発が起こった。免疫系の間接的な活性化に加えて、抗体は、免疫エフェクター細胞を正確に関与させるために癌細胞に直接フラグを付け、死の受容体アゴニストを介して細胞毒性を引き起こし、腫瘍細胞生存シグナル伝達をブロックし、血管新生(血管の成長)を妨害し、免疫チェックポイント調節因子を制約し、放射性同位体、化学療法薬およびsiRNAを共役剤として送達することを目的としている。また、患者由来T細胞やNK細胞(CAR-TおよびCAR-NK)の表面上の様々な抗体の単鎖可変フラグメント(scFv)を研究することは、細胞ベース療法のための臨床研究の急速な成長領域である4。
抗原発現腫瘍細胞への選択性を提供する抗体系薬物の超高親和性は、それを魅力的な薬剤にする。同様に、治療用抗体(または化学薬品)の標的送達および腫瘍保持は、毒性に対する有効性のバランスをとるための鍵である。したがって、二重特異性5および三特異的抗体6を含むがこれらに限定されない多数のタンパク質工学ベースの戦略が、静脈内に(IV)注入された治療薬のアビディティ最適化腫瘍保持を有意に増強するために利用されている。ここでは、潜在的に有効な抗癌抗体の腫瘍および組織分布に対処するための単純な蛍光ベースの方法について述べた。
動物組織は可視スペクトルで励起されると自己蛍光を有するため、抗体は当初、近赤外色素(例えばIRDye 800CW)で標識された。概念実証調査では、フォールレスツズマブと呼ばれる抗体を標的とする葉酸受容体α-1(FOLR1)とその誘導体である二重特異性アンカー細胞毒性活性化剤(BaCa)7抗体を1つの組換え抗体にCOR1および死受容体-5(DR5)8を共同標的とする抗体を用いた。FOLR1は、卵巣およびTNBC癌細胞、腫瘍異種移植片および患者腫瘍9において明確に定義された過剰発現標的受容体である。特に、抗体ベースのアプローチを用いてFOLR1を臨床的に利用して、卵巣癌および乳癌に対する免疫エフェクター細胞および抗体薬物コンジュゲート(ADC)を関与させる複数の取り組みがある。
本方法論文では、CHO発現系を用いた他の対照抗体とともに、臨床抗FOLR1(farletuzumab)をクローニング、発現、精製した。IgG1アイソタイプおよびアバゴボマブ12 と呼ばれる臨床抗イディオタイプムチン-16抗体を陰性対照として使用した。タンパク質A精製後、示された抗体はIRDye 800CWで標識され、卵巣腫瘍異種移植片または安定してトランスフェクトされたヒトFOLR1を発現するヒトFOLR1マウス大腸癌異種移植片のいずれかを有するヌードマウスの尾静脈に投与された。抗体の局在化は、複数の異なる時点点7でin vivoイメージングスペクトルを用いたライブイメージングによって追跡した。この方法は、発光を可能にするために基質の遺伝子組み換えまたは注入を必要とせず、大幅に迅速かつ費用対効果が高く、効率的である。以下に記載の一般的なクローニング、発現、精製および標識プロトコルは、重鎖および軽鎖配列が利用可能な場合、任意の臨床および非臨床抗体に適用することができる。

このプロトコルは、腫瘍および組織における抗体分布の発現と精製および抗体分布のリアルタイムモニタリングを実証する。記載されたプロトコルを用いた抗体のスクリーニングは、限定的な非特異的組織分布を有する抗体の選択を助け、腫瘍の局在を豊かにする。記載された技術は、迅速かつ費用対効果が高い。単光子放射コンピュータ断層撮影や陽電子放出断層撮影などの他の技術は非常に高価であり、放射性および他の洗練されたトレーサーを必要とする。この技術は、がん標的抗体に限定されない。これは、体内の抗体分布の分析のために、肺や心臓病などの他の疾患に適用することができます。手順を実証すると、私の大学の獣医技術者ジェレミー・ゲイツマンになります。200ミリリットルのメディアの生のCHO細胞は、デロンで、エルレンマイヤーバッフルフラスコ。CHOフリースタイル培地5ミリリットルと可変重クローンDNA50マイクログラム、15ミリリットルチューブ内の可変光クローンDNA75マイクログラムを組み合わせます。次いで、チューブを渦に入れ。混合物を室温で5分間放置します。1ミリリットルの750マイクロリットルを1ミリリットルのポリエチレンアミンストックをDNA溶液に加え、30秒間積極的に渦液化します。さらに5分間室温で混合物を残します。フラスコを手動で振りながら、CHOセルに全体の混合物を追加します。130 RPMで振りながら、すぐに37°Cで細胞をインキュベートします。翌日、100X抗凝集剤を2ミリリットル、100X抗菌抗菌性抗菌溶液を2ミリリットルを細胞に加えます。フラスコを摂氏32~34度でインキュベートし、130RPMで揺れ動かします。10〜11日間細胞を培養し続け、トリプトンN1飼料10ミリリットル、5日ごとに100 Xグルタミンサプリメント2ミリリットルを加えます。細胞の生存率が10日または11日に80%を超えることを確実にするために、3日ごとに細胞を数え、培養物を3000倍Gで遠心し、40〜60分間摂氏4度で抗体精製用培地を収穫する。次に、0.22マイクロメートルのボトルフィルターで透明な媒体を濾過します。抗体を精製するために、結合バッファーの2列体積でプロテインAカラムを平衡化する。1 分間に 1 ミリリットルの流量で、フィルターされたメディアをカラムに通します。次に、2段の結合バッファーでカラムを洗います。溶出バッファーの 5 ミリリットルを使用して、抗体を 500 マイクロリットル分数に溶出させます。そして、分画のために中和バッファーの10マイクロリットルを添加することによって、ほのめかされた抗体のpHを中和する。IgGのデフォルトプロトコルを使用して、分光光度計で精製された抗体の濃度を測定します。1リットルの共役バッファーに透析カセットを用いた抗体の透析0.5ミリリットル。4時間後、透析カセットを新鮮なバッファーに移し、一晩透析します。共役反応を設定するには、抗体の1ミリグラムあたり0.03マイクログラムのIRR色素800 CWUを合計容量500マイクロリットルで添加します。混合物を摂氏20度で2時間インキュベートし、PBSに対する広範な透析によって標識コンジュゲートを精製する。色素780ナノメートルの吸光度を測定し、タンパク質の吸光度を280ナノメートルで測定して標識の程度を推定する。マウスの異種を開発するには、動物の皮膚をそっと持ち上げ、下層の筋肉層から分離します。26ゲージ針で皮膚の下に100マイクロリットルの細胞懸濁液をゆっくりと注入する。ニードルを取り出す前に数秒待って、地下マトリックス培地が皮膚下の細胞と一緒に半固体ゲル状の構造を知らせ、注射部位から混合物が出てくるのを防ぎます。インビボイメージングを行うために、尾静脈を介して色素標識抗体を注入する。マウスを麻酔した後、ペダル反射に対する応答の欠如を確認し、温水を適用して静脈を拡張します。26ゲージ針の1ccインスリン注射器を使用して、100マイクロリットルの体積で標識抗体の25マイクログラムを注入する。陰性対照として、癌細胞を標的としない非特異的IgGアイソタイプ抗体を標識して注入する。インビボイメージングを行う 8、24、48 抗体注射後。イメージング ソフトウェアで、[初期化] をクリックします。ステージ温度が摂氏37度であることを確認します。コントロールパネルで、イメージングウィザードを使用して蛍光イメージングを設定し、励起を773ナノメートルに設定し、放出を792ナノメートルに設定します。麻酔マウスを撮像室に移します。ノーズコーンを使用してイメージングフィールドに配置します。準備ができたら、イメージ取得用のコントロールパネルの取得をクリックし、[自動公開]をクリックします。生成された画像は、光蛍光と濃度がカウントまたは光子の単位で表示された写真画像上の蛍光のオーバーレイです。可変重および可変光ドメインを有する陽性cDNAクローンはコロニーPCRで確認された。非還元および還元SDSページでの精製されたファルレツズマブランの代表的な結果をここに示す。重鎖および軽鎖は、還元後に50キロと25キロダルトンバンドを生産した。細胞表面における天然タンパク質に対する抗体の結合も確認された。蛍光標識抗体は、尾静脈を注射し、腫瘍を発現する完全なR1で移植されたマウスに注射した。動物は、IVISを使用して複数の時点で生きた画像であった。このデータは、腫瘍に完全なR1およびBaCa抗体の選択的濃縮を打ち明ける。このプロトコルを試みるとき、CHO細胞の維持は抗体の収量にとって重要であることを覚えておいてください。そして、競合性腫瘍局在化のために同様の程度の蛍光色素結合を有することが重要である。この手順に従って、組織免疫組織化学や組織または腫瘍特異的ELISAなどの他の方法を、生きたマウスのイメージングデータを支持するように行うことができる。
