Desuden har de genetiske evner mus i høj grad hjulpet i vores evne til at sonde den genetiske fundament af FASD10,11. Zebrafisken har vist sig som et kraftfuldt modelsystem til at undersøge mange aspekter af ethanol teratorene22,23. På grund af deres eksterne befrugtning, høj frugtbarhed, genetisk tractability, og levende billeddannelse kapaciteter, zebrafisk er velegnet til at studere faktorer såsom timing, dosering, og genetik af ethanol teratogenese. Ethanol kan administreres til præcist iscenesatte embryoner, og embryonerne kan derefter afbildes for at undersøge ethanolens direkte virkning under udviklingsprocesser. Dette arbejde kan relateres direkte til mennesker, fordi de genetiske programmer for udvikling er stærkt bevaret mellem zebrafisk og mennesker og kan derfor hjælpe guide FASD menneskelige undersøgelser24. Mens zebrafisk er blevet brugt til at undersøge ethanol terateseene, en mangel på konsensus i rapportering embryonale ethanol koncentrationer gør sammenligning med mennesker vanskeligt25. I pattedyrsystemer, blod-alkohol niveauer korrelerer direkte til væv ethanol niveauer26. Mange af zebrafiskundersøgelserne behandler embryoner før fuldstændig dannelse af deres kredsløbssystem. Uden nogen moderprøve at undersøge, en proces til vurdering af ethanol koncentrationer er nødvendig for at kvantificere ethanol niveauer i fosteret. quantification of ethanol levels in zebrafish embryos using head space gas chromatography

Føtale alkoholspektrumforstyrrelser (FASD) beskriver et meget varierende kontinuum af ethanol-inducerede udviklingsdefekter, herunder ansigtsdysmorfoster og neurologiske funktionsnedsættelser. Med en kompleks patologi påvirker FASD ca. 1 ud af 100 børn født i USA hvert år. På grund af FASD's meget variable karakter har dyremodeller vist sig at være kritiske i vores nuværende mekanistiske forståelse af ethanolinducerede udviklingsfejl. Et stigende antal laboratorier har fokuseret på at bruge zebrafisk til at undersøge ethanol-inducerede udviklingsfejl. Zebrafisk producerer et stort antal eksternt befrugtede, genetisk tractable, gennemskinnelige embryoner. Dette gør det muligt for forskere præcist at kontrollere timingen og doseringen af ethanoleksponering i flere genetiske sammenhænge og kvantificere virkningen af embryonal ethanoleksponering gennem levende billeddannelsesteknikker. Denne protokol er betydelig, fordi det hjælper med at forstå farmakokinetik af ethanol i udviklingen af zebrafisk embryoner, og dette vil hjælpe standardisere ethanol eksponering regimer i zebrafisk undersøgelser. Den største fordel ved vores teknik er, at den giver forskerne mulighed for hurtigt og reproducerbart at måle ethanolniveauer under embryonal udvikling, især i embryoner, hvor blodforsyningen ikke er tilgængelig. Begynd med at overføre 10 embryoner med ekstra embryonal væske til 1,5 milliliter mikrocentrifugerør markeret med en linje i et volumen på 250 mikroliter. Ethanolkoncentrationen i embryonerne beregnes ved at beregne fortyndingsfaktoren for hver prøve og multiplicere prøvenedhanetværdien med denne fortyndingsfaktor.Endelig beregnes mediereferenceprøver ved at gange medieethanolværdien med en fortyndingsfaktor på 10. For korrekt at beregne de embryonale ethanolkoncentrationer blev mængderne af embryoner både i deres chorion og fjernet fra deres chorion målt. I denne analyse udgjorde embryonet 56% af det samlede volumen inde i chorion.Baseret på tidligere offentliggjort arbejde blev 73,6% brugt som fostervandsindhold for alle de præsenterede analyser. Dette resulterede i vand bestående af 0,82 mikroliter af de 1,1 mikroliter af embryonale volumen. Af det samlede vandvolumen er 49% indeholdt i fosteret, og 51% er i den ekstraembryoniske væske.Til gaskromatografianalysen blev to grupper på 15 prøver og de medier, som de blev behandlet med, målt fem gange pr. gruppe analyseret. Medieethanolniveauerne var 143,6 millimolar for gruppe 1 og 133,6 millimolar for gruppe to. Ethanolkoncentrationerne af embryonerne var i gennemsnit 63,5 millimolar og 53,1 millimolar for henholdsvis gruppe 1 og 2.Der blev beregnet et forhold mellem embryonal ethanolkoncentration og medieniveauer af ethanol for hver prøve. Gruppe 1 havde 44% af medierne ethanol niveauer, mens gruppen havde 40% af medierne ethanol niveauer. Ubehandlede kontrolembryoner blev målt ved koncentrationen af ethanol i nul millimolarmediethanol.Det vigtigste at huske, når du forsøger denne procedure er, at behandling af embryoner til GC analyse korrekt er kritisk som ethanol er flygtig og vil fordampe hurtigt. Den bedste måde at udføre dette trin er beskrevet, men det skal praktiseres for at udvikle ordentlig muskel hukommelse. Denne procedure vil bidrage til at standardisere ethanol behandling regimer i zebrafisk og give det fremtidige arbejde for at dissekere de genetiske og cellulære mekanismer ethanal teratogenicitet.
