Als ontwikkelingsmodelsysteem zijn zebravissen bij uitstek geschikt om de impact van omgevingsfactoren op de ontwikkeling te bestuderen. Foetale alcoholspectrumstoornissen (FASD) beschrijven een breed scala aan neurologische stoornissen en craniofaciale dysmorfologieën geassocieerd met blootstelling aan embryonale ethanol1. Meerdere factoren, waaronder timing en dosering van blootstelling aan ethanol en genetische achtergrond, dragen bij aan de variatie van FASD2,3. Bij mensen maakt de complexe relatie van deze variabelen het bestuderen en begrijpen van de etiologie van FASD uitdagend. Diermodellen zijn cruciaal gebleken bij het ontwikkelen van ons begrip van de mechanistische basis van ethanolteratogeniteit. Een breed scala van dier model systemen is gebruikt om meerdere aspecten van FASD studie en de resultaten zijn opmerkelijk consistent met wat wordt gevonden in de blootstelling bij de mens4. Knaagdiermodelsystemen worden gebruikt om vele aspecten van de FASD te onderzoeken, waarbij muizen de meest voorkomende5,6,7zijn. Het merendeel van dit werk is gericht op ontwikkelingsfouten aan vroege blootstelling aan ethanol8, hoewel later blootstelling aan ethanol is aangetoond dat ontwikkelingsafwijkingen veroorzaken en9. Bovendien hebben de genetische vermogens van muizen sterk geholpen in ons vermogen om de genetische onderbouwing van FASD10,11te onderzoeken. Deze studies bij muizen suggereren sterk dat er gen-ethanol interacties met de sonische egel route, retinoïne zuur signalering, Superoxide dismutase, stikstofmonoxide synthase I, Aldh2 en Fancd28,10,11,12,13,14,15,16,17,18, 19,20,21. Deze studies tonen aan dat diermodellen van cruciaal belang zijn voor het bevorderen van ons begrip van De FASD en de onderliggende mechanismen ervan.
De zebravis is ontstaan als een krachtig modelsysteem om vele aspecten van ethanol teratogenese te onderzoeken22,23. Vanwege hun externe bevruchting, hoge vruchtbaarheid, genetische traktaaten en levende beeldvormingsmogelijkheden, zijn zebravis bij uitstek geschikt om factoren zoals timing, dosering en genetica van ethanolteratogenese te bestuderen. Ethanol kan worden toegediend aan precies geënsceneerde embryo's en de embryo's kunnen vervolgens worden afgebeeld om de directe impact van ethanol tijdens ontwikkelingsprocessen te onderzoeken. Dit werk kan rechtstreeks aan mensen worden verwant, omdat de genetische programma's van ontwikkeling hoogst behouden tussen zebravissen en mensen zijn en daarom kunnen helpen fasd menselijke studies24begeleiden. Terwijl zebravis zijn gebruikt om ethanol te onderzoeken, een gebrek aan consensus in de rapportage van embryonale ethanol concentraties maakt vergelijking met de mens moeilijk25. In zoogdiersystemen correleren de bloedalcoholniveaus rechtstreeks met weefselethanolniveaus26. Veel van de zebravisstudies behandelen embryo's vóór volledige vorming van hun bloedsomloop. Zonder moedermonster om te onderzoeken, is een proces om ethanolconcentraties te beoordelen nodig om ethanolniveaus in het embryo te kwantificeren. Hier beschrijven we een proces om ethanolconcentraties te kwantificeren in een zich ontwikkelend zebravisembryo met behulp van hoofdruimtegaschromatografie.

quantification of ethanol levels in zebrafish embryos using head space gas chromatography

Foetale Alcohol Spectrum Disorders (FASD) beschrijven een zeer variabel continuüm van ethanol-geïnduceerde ontwikkelingsstoornissen, met inbegrip van gezichtsdysmorfologieën en neurologische stoornissen. Met een complexe pathologie treft FASD jaarlijks ongeveer 1 op de 100 kinderen die in de Verenigde Staten worden geboren. Door het zeer variabele karakter van FASD zijn diermodellen kritisch gebleken in ons huidige mechanistische begrip van door ethanol veroorzaakte ontwikkelingsfouten. Steeds meer laboratoria richten zich op het gebruik van zebravissen om door ethanol veroorzaakte ontwikkelingsfouten te onderzoeken. Zebravissen produceren grote aantallen extern bevruchte, genetisch hardnekkige, doorschijnende embryo's. Dit stelt onderzoekers in staat om de timing en dosering van blootstelling aan ethanol in meerdere genetische contexten nauwkeurig te controleren en de impact van blootstelling aan embryonale ethanol te kwantificeren door middel van live imaging technieken. Dit, gecombineerd met de hoge mate van behoud van zowel genetica als ontwikkeling met de mens, heeft bewezen dat zebravis een krachtig model is om de mechanistische basis van ethanolteratogeniteit te bestuderen. Echter, ethanol blootstelling regimes hebben gevarieerd tussen verschillende zebravissen studies, die de interpretatie van zebravissen gegevens over deze studies heeft verward. Hier is een protocol om ethanolconcentraties in zebravissenembryo's te kwantificeren met behulp van hoofdruimtegaschromatografie.

Bloedethanol niveaus kunnen niet worden bepaald in de vroege embryonale zebravissen, omdat ze niet over een volledig gevormde bloedsomloop. Om het ethanolgehalte in de zebravisembryo's te bepalen, worden de ethanolniveaus rechtstreeks gemeten aan de andere dag van gehomogeniseerd embryonaal weefsel. Dit protocol is belangrijk omdat het helpt bij het begrijpen van de farmacokinetiek van ethanol bij het ontwikkelen van zebravisembryo's en dit zal helpen bij het standaardiseren van ethanolblootstellingsregimes in zebravisstudies. Het belangrijkste voordeel van onze techniek is dat onderzoekers hierdoor snel en reproduceerbaar ethanolniveaus kunnen meten tijdens de embryonale ontwikkeling, met name in embryo's waar geen bloedtoevoer beschikbaar is. Deze methode kan gemakkelijk worden toegepast op andere modelsystemen, evenals andere omgevingsfactoren uitgaande van de juiste techniek en apparatuur setup.Ik verwacht dat individuen die dit protocol voor het eerst uitvoeren, worstelen met de snelheid die nodig is om verdamping van ethanol tijdens embryoverwerking te voorkomen. Begin met het overbrengen van 10 embryo's met extra embryonale vloeistof in 1,5 milliliter microcentrifugebuizen gemarkeerd met een lijn met een volume van 250 microliter. Voeg water toe aan de vullijn.Bereid extra microcentrifugebuizen voor gedechorioneerde monsters. Om het chorion te verwijderen, incubeer de embryo's in een 100 millimeter petrischaal met twee milligram per milliliter proteasecocktail in embryomedia gedurende 10 minuten. Draai de schotel voorzichtig om de paar minuten om de chorion te breken.Zodra alle embryo's vrij zijn van het chorion, verwijder ze uit de protease cocktail en was ze twee keer met verse media. Breng de embryo's vervolgens over naar een verse schaal van 100 millimeter. Wanneer alle monsters klaar zijn voor volumemeting, gebruikt u een P200-micropipet met de kleinst mogelijke punt om alle vloeistof voorzichtig uit de embryo's te verwijderen.Weeg het water met een schaal met minder dan 0,1 milligram precisie, dan bepalen het volume van de 10 embryo's door het gewicht van het verwijderde water af te trekken van 250 microliter. Om het volume van een enkel embryo te bepalen, verdeelt u het verschil door 10. Verzamel de embryo's uit de paringstanks en leg ze in een standaard petrischaal van 100 millimeter en broed de schaal vervolgens uit op 28,5 graden Celsius.Breng na zes uur bevruchting tot 100 embryo's over naar een nieuw gerecht met embryomedia en 1% ethanol of media alleen. Bedek maar om de petrischaaltjes niet te verzegelen en plaats ze 18 uur in een couveuse van 28,5 graden Celsius of tot ze 24 uur na de bevruchting zijn. Gebruik twee P200 micropipetten ingesteld op 50 microliters en aspirate de protease cocktail oplossing in een en water in de tweede.Plaats dan snel 10 embryo's in een 1,5 milliliter microcentrifuge buis met behulp van een glazen pipet en sluit de dop. Herhaal dit proces voor alle monsters worden getest, controles en ethanol behandelde embryo's. Werken met een monster tegelijk, snel open de dop en aspirate alle resterende embryo media met een P200 micropipet ingesteld op 200 microliters.Voeg snel maar voorzichtig de 50 microliter water toe en verwijder ze, voeg vervolgens de 50 microliter proteasecocktail toe en sluit de dop van de buis. Laat het monster 10 minuten op kamertemperatuur zitten en voeg dan snel 450 microliters van vijf molaire natriumchloride toe en sluit de dop van de buis. Homogeniseer het monster door 10 minuten te vortexen en breng twee microliter supernatant over naar een gaschromatografie flacon.Verzegel de flacon met een polytetrafluorethyleendop. Na het voltooien van de opstartmethode, laad 436 huidige SPME ethanol twee tot vijf minuten RG run. meth uit het methodenmenu in de analysesoftware voor elk voorbeeld op de voorbeeldlijst.Vul de monsterlijst in die begint met de lucht, het water en vijf molaire natriumchloride protease cocktail blanks. Voer dan in de ethanol normen in volgorde van 0,3125 tot 40 millimolar. Volg de normen met de tweede ronde van losse flodders.Voer alle gehomogeniseerde embryo-supernatantmonsters in die moeten worden getest van de laagste tot hoogste voorspelde ethanolconcentratie en eindig door een derde ronde losse flodders in te voeren. Voeg de gaschromatografie flacons toe aan de autosampler in de volgorde waarin de monsters zijn ingevoerd en laat ze 10 tot 15 minuten opwarmen. Start vervolgens het monster wordt uitgevoerd met behulp van de software.Nadat alle runs zijn voltooid, activeert u de afsluitmethode door een definitief voorbeeld toe te voegen aan de voorbeeldlijst en standby.meth uit te voeren. Een back-up maken van alle gegevens die tijdens de samplerun zijn verkregen. Zodra de afsluitmethode is voltooid, klikt u op het geopende chromatogram en opent u de map met de resultaten.Monsters worden automatisch geïntegreerd in dit programma. Zorg er in de resultaten voor dat de juiste pieken zijn geïntegreerd. Zodra alle monsters zijn bevestigd, drukt of exporteert u de resultaten.Plot de piekhoogte van de ethanolnormen op een grafiek en bereken vervolgens de hellings-, y-intercept- en r-kwadraatwaarden. Verkrijg de ethanolwaarde voor elk monster door de piekhoogte van het monster af te trekken van de y-intercept van de normen en deze waarde te delen door de helling van de normen. Om de ethanolconcentratie in de embryo's te berekenen, berekent u de verdunningsfactor voor elk monster en vermenigvuldigt u de waarde van de monster ethanol met deze verdunningsfactor.Bereken ten slotte referentiemonsters in de media door de media-ethanolwaarde te vermenigvuldigen met een verdunningsfactor van 10. Om de embryonale ethanolconcentraties goed te berekenen, werden de hoeveelheden embryo's in hun chorion en verwijderd uit hun chorion gemeten. In deze analyse bestond het embryo uit 56% van het totale volume in het chorion.Op basis van eerder gepubliceerd werk werd 73,6% gebruikt als embryonaal watergehalte voor alle gepresenteerde analyses. Dit resulteerde in water bestaande uit 0,82 microliter van de 1,1 microliter van embryonale volume. Van het totale watervolume zit 49% in het embryo en 51% in de extraembryonische vloeistof.Voor de gaschromatografieanalyse werden twee groepen van 15 monsters en de media waarmee ze werden behandeld vijf keer per groep geanalyseerd. Media-ethanolniveaus waren 143,6 millimolar voor groep één en 133,6 millimolar voor groep twee. De ethanolconcentraties van de embryo's bedroegen gemiddeld 63,5 millimolar en 53,1 millimolar voor respectievelijk de groepen één en twee.Voor elk monster werd een verhouding berekend tussen de concentratie embryonale ethanol en de medianiveaus ethanol. Groep één had 44% van de media-ethanolniveaus, terwijl de groep 40% van de ethanolniveaus in de media had. Onbehandelde controle embryo's werden gemeten bij nul millimolar media ethanol concentratie.Het belangrijkste om te onthouden bij het proberen van deze procedure is dat de verwerking van de embryo's voor GC-analyse goed is van cruciaal belang als ethanol is vluchtig en zal snel verdampen. De beste manier om deze stap uit te voeren wordt beschreven, maar het moet worden beoefend om een goede spier geheugen te ontwikkelen. Deze procedure zal helpen standaardiseren ethanol behandeling regimes in zebravissen en laat toekomstig werk om de genetische en cellulaire mechanismen van ethanal terratogeneiteit ontleden.
