Diese Studien zeigen, dass Tiermodelle entscheidend sind, um unser Verständnis von FASD und seinen zugrunde liegenden Mechanismen zu fördern.
Der Zebrafisch hat sich als ein leistungsfähiges Modellsystem entwickelt, um viele Aspekte der Ethanolteratogenese22,23zu untersuchen. Aufgrund ihrer externen Befruchtung, hohen Fruchtbarkeit, genetischen Traktionsfähigkeit und Live-Bildgebung sind Zebrafische ideal geeignet, um Faktoren wie Timing, Dosierung und Genetik der Ethanol-Teratogenese zu untersuchen. Ethanol kann präzise inszenierten Embryonen verabreicht werden und die Embryonen können dann abgebildet werden, um die direkte Wirkung von Ethanol während der Entwicklungsprozesse zu untersuchen. Diese Arbeit kann direkt mit dem Menschen in Verbindung gebracht werden, da die genetischen Entwicklungsprogramme zwischen Zebrafischen und Menschen stark konserviert sind und daher helfen können, FASD-Humanstudien zu leiten24. Während Zebrafische verwendet wurden, um Ethanol-Teratogenese zu untersuchen, macht ein Mangel an Konsens bei der Meldung embryonaler Ethanolkonzentrationen den Vergleich mit Menschen schwierig25. In Säugetiersystemen korrelieren Blutalkoholwerte direkt mit dem Ethanolspiegel des Gewebes26. Viele der Zebrafisch-Studien behandeln Embryonen vor der vollständigen Bildung ihres Kreislaufsystems. Da keine mütterliche Probe zu untersuchen ist, ist ein Verfahren zur Bewertung der Ethanolkonzentrationen erforderlich, um den Ethanolgehalt im Embryo zu quantifizieren. Hier beschreiben wir ein Verfahren zur Quantifizierung der Ethanolkonzentrationen in einem sich entwickelnden Zebrafisch-Embryo mittels Kopfraumgaschromatographie.

quantification of ethanol levels in zebrafish embryos using head space gas chromatography

Fetale Alkoholspektrumsstörungen (FASD) beschreiben ein hochvariables Kontinuum von Ethanol-induzierten Entwicklungsfehlern, einschließlich Gesichtsdysmorphologien und neurologischen Beeinträchtigungen. Bei einer komplexen Pathologie betrifft FASD etwa 1 von 100 Kindern, die jedes Jahr in den Vereinigten Staaten geboren werden. Zebrafische produzieren eine große Anzahl von extern befruchteten, genetisch befruchtbaren, durchscheinenden Embryonen. Dies ermöglicht es Forschern, Timing und Dosierung der Ethanol-Exposition in mehreren genetischen Kontexten präzise zu steuern und die Auswirkungen der embryonalen Ethanol-Exposition durch Live-Bildgebungstechniken zu quantifizieren. Zusammen mit dem hohen Grad an Erhaltung sowohl der Genetik als auch der Entwicklung beim Menschen hat sich der Zebrafisch als ein starkes Modell erwiesen, um die mechanistische Grundlage der Ethanol-Teratogenität zu untersuchen. Die Ethanolexpositionsschemas unterscheiden sich jedoch zwischen verschiedenen Zebrafischstudien, was die Interpretation von Zebrafischdaten in diesen Studien verwirrt hat. Hier ist ein Protokoll zur Quantifizierung der Ethanolkonzentrationen in Zebrafischembryonen mittels Kopfraumgaschromatographie.

Der Ethanolgehalt im Blut kann bei frühen embryonalen Zebrafischen nicht bestimmt werden, da es an einem voll ausgebildeten Kreislaufsystem fehlt. Um die Ethanolkonzentration in den Zebrafischembryonen zu bestimmen, werden die Ethanolwerte direkt aus homogenisiertem embryonalem Gewebe gemessen. Diese Methode kann leicht auf andere Modellsysteme sowie andere Umweltfaktoren angewendet werden, die eine ordnungsgemäße Technik und Ausrüstungseinrichtung voraussetzt.Ich würde erwarten, dass Personen, die dieses Protokoll zum ersten Mal durchführen, mit der Geschwindigkeit kämpfen, die notwendig ist, um die Verdunstung von Ethanol während der Embryoverarbeitung zu verhindern. Beginnen Sie mit der Übertragung von 10 Embryonen mit extra embryonaler Flüssigkeit in 1,5 Milliliter Mikrozentrifugenröhren, die mit einer Linie bei einem Volumen von 250 Mikrolitern markiert sind. Bedecken, aber nicht versiegeln Sie die Petri-Gerichte und legen Sie sie in einem 28,5 Grad Celsius Inkubator für 18 Stunden oder bis sie 24 Stunden nach der Düngung erreichen. Verwenden Sie zwei P200-Mikropipetten, die auf 50 Mikroliter eingestellt sind, und aspirieren Sie die Protease-Cocktail-Lösung in eine und gießen Sie sie in die zweite.Dann legen Sie schnell 10 Embryonen in ein 1,5 Milliliter Mikrozentrifugenrohr mit einer Glaspipette und schließen Sie die Kappe. Wenn Sie mit jeweils einer Probe arbeiten, öffnen Sie schnell die Kappe und aspirieren Sie alle verbleibenden Embryomedien mit einer P200-Mikropipette, die auf 200 Mikroliter eingestellt ist.Schnell aber schonend die 50 Mikroliter Wasser hinzufügen und entfernen, dann die 50 Mikroliter Protease-Cocktail hinzufügen und die Rohrkappe schließen. Lassen Sie die Probe für 10 Minuten bei Raumtemperatur sitzen, dann schnell 450 Mikroliter von fünf Mol-Natriumchlorid hinzufügen und die Rohrkappe schließen. Fügen Sie die Gaschromatographie-Fläschchen in der Reihenfolge, in der die Proben eingegeben wurden, zum Autosampler hinzu und lassen Sie sie 10 bis 15 Minuten erwärmen. Erhalten Sie den Ethanolwert für jede Probe, indem Sie die Spitzenhöhe der Probe vom y-Abfang der Standards subtrahieren und diesen Wert durch die Steigung der Standards dividieren. Um die Ethanolkonzentration in den Embryonen zu berechnen, berechnen Sie den Verdünnungsfaktor für jede Probe und multiplizieren Sie den Ethanolwert der Probe mit diesem Verdünnungsfaktor.Berechnen Sie schließlich Medienreferenzproben, indem Sie den Ethanolwert der Medien mit einem Verdünnungsfaktor von 10 multiplizieren. Um die embryonalen Ethanolkonzentrationen richtig zu berechnen, wurden die Mengen der Embryonen sowohl in ihrem Chorion als auch in ihrem Chorion gemessen. In dieser Analyse machte der Embryo 56% des Gesamtvolumens innerhalb der Chorion aus.Basierend auf zuvor veröffentlichten Arbeiten wurden 73,6 % als embryonaler Wassergehalt für alle vorgestellten Analysen verwendet. Daraus resultierte Wasser mit 0,82 Mikrolitern der 1,1 Mikroliter embryonalen Volumens. Vom gesamten Wasservolumen sind 49 % im Embryo und 51 % in der extraembryonalen Flüssigkeit enthalten.Für die Gaschromatographie-Analyse wurden zwei Gruppen von 15 Proben und die Medien, mit denen sie behandelt wurden, fünfmal pro Gruppe gemessen. Der Ethanolgehalt der Medien war 143,6 Millimolar für Gruppe eins und 133,6 Millimolar für Gruppe zwei. Die Ethanolkonzentrationen der Embryonen betrugen durchschnittlich 63,5 Millimolar bzw. 53,1 Millimolar für die Gruppen eins und zwei.Für jede Probe wurde ein Verhältnis der embryonalen Ethanolkonzentration zu den Mediaspiegeln von Ethanol berechnet. Gruppe eins hatte 44% der Media-Ethanol-Werte, während Gruppe 40% der Medien-Ethanol-Werte hatte. Unbehandelte Kontrollembryonen wurden mit einer Ethanolkonzentration von null Millimolaren Medien gemessen.Das Wichtigste, was man beim Versuch dieses Verfahrens beachten sollte, ist, dass die richtige Verarbeitung der Embryonen für die GC-Analyse entscheidend ist, da Ethanol flüchtig ist und schnell verdampft. Der beste Weg, um diesen Schritt durchzuführen, wird beschrieben, aber es muss geübt werden, um richtige Muskelgedächtnis zu entwickeln. Dieses Verfahren wird dazu beitragen, Ethanol-Behandlungssysteme in Zebrafischen zu standardisieren und zukünftige Arbeiten zu ermöglichen, um die genetischen und zellulären Mechanismen der ethanalen Teratogenität zu sezieren.
