Sinds de jaren 1980, de middelste cerebrale slagader occlusie (MCAO) model geïnduceerd door siliconen filamenten is op grote schaal gebruikt in de fundamentele slag onderzoek1. Er zijn ook andere methoden gebruikt (d.w.z. het hechten van één tak van de MCA2 en fotochemisch geïnduceerde focale infarcten). Deze modellen zijn genoemd MCA-gebaseerde slagmodellen en hebben sterk bijgedragen aan onderzoeken van de pathofysiologische mechanismen die ten grondslag liggen aan beroerte en potentiële therapieën. Hoewel er beperkingen van deze experimentele modellen3,4, deze methoden zijn gebruikt veel laboratoria5,6. Op MCA gebaseerde slagmodellen vertegenwoordigen een beroerte in de voorste circulatie; echter, weinig rapporten hebben onderzocht modellen nabootsen beroerte in de achterste circulatie7.
Er zijn significante verschillen tussen de etiologie, mechanismen, klinische manifestatie en prognose tussen voorste en achterste circulatieslagen8. Daarom kunnen de resultaten die zijn afgeleid van voorste circulatieslagmodellen niet worden toegepast op een circulatieslag achteraf. Zo is het reperfusietijdvenster voor voorste circulatie uitgebreid tot 6 uur, met een klein deel van de studies die zich uitstrekken tot 24 uur op basis van beeldvormingsbevindingen9. Echter, het tijdvenster voor achterste circulatie kan langer zijn dan 24 uur, volgens eerdere rapporten10 en onze eigen klinische ervaringen. Dit langwerpige reperfusietijdvenster moet verder worden bestudeerd en bevestigd in experimentele modellen.
Wat de achterste circulatieslagen betreft, is het pontijninfarct het meest voorkomende subtype, goed voor 7% van alle ischemische beroertegevallen11,12. Volgens infarcttopografie, pontine infarcten zijn verdeeld in geïsoleerde en niet-geïsoleerde pontine infarcten13. Geïsoleerde pontijn infarcten zijn gecategoriseerd in drie soorten op basis van de onderliggende mechanismen: grote slagaderziekte (LAD), basilarslagader tak ziekte (BABD), en kleine slagaderziekte (SAD). Kennis van de mechanismen, manifestatie en prognose van pontine infarct is afgeleid van klinische onderzoeken van gevallen14. Echter, een knaagdier model nabootsen pontine infarct is minder onderzocht.
In eerdere studies, diffuse hersenstam tegmentum letsel waarbij de pons is onderzocht7. Een groep probeerde een pontine infarct model te creëren via ligatie van de basilaire slagader (BA)15. Een andere groep gebruikte een 10-0 nylon monofilament hechting om selectief te ligate vier punten van de proximale BA selectief16. Dit model bootst LAD na, maar de meeste pontine infarcten zijn het gevolg van BABD en SAD. Bovendien is selectieve ligatie van de BA een ingewikkelde operatie en heeft een hoog sterftecijfer.
Hier is een gedetailleerd protocol voor een gemakkelijk uit te voeren, gemakkelijk gereproduceerd, en succesvolle rat model van acute pontine infarct door elektrische stimulatie.

Pontine infarct is de meest voorkomende slag subtype in de achterste circulatie, terwijl er ontbreekt een knaagdier model nabootsen pontine infarct. Mits hier is een protocol voor het succesvol vaststellen van een rat model van acute pontine infarct. Ratten met een gewicht van ongeveer 250 g worden gebruikt, en een sonde met een geïsoleerde schede wordt geïnjecteerd in de pons met behulp van een stereotaxic apparaat. Een laesie wordt veroorzaakt door de elektrische stimulatie met een enkele puls. De Longa score, Berderson score, en balk balans test worden gebruikt om neurologische tekorten te beoordelen. Daarnaast wordt de kleefverwijdering somatosensorische test gebruikt om de sensorimotorische functie te bepalen, en de ledemaat plaatsing test wordt gebruikt om proprioceptie te evalueren. MRI-scans worden vervolgens gebruikt om het infarct in vivo te beoordelen, en TTC-kleuring wordt gebruikt om het infarct in vitro te bevestigen. Hier wordt een succesvol infarct geïdentificeerd dat zich in de anterolaterale basis van de rostralpons bevindt. Tot slot wordt een nieuwe methode beschreven om een acuut pontïneinfarct rat model vast te stellen.

Wij bieden een protocol om een infarct vast te stellen in een pons van rat. Dit pontine infarct rat model werd gemakkelijk uitgevoerd, gemakkelijk gereproduceerd, en zeer succesvol model. De elektrische stimulatie kan worden gebruikt om een infarct te maken in een ander hersengebied zoals thalamus.Mensen die dit model willen uitvoeren, moeten de pontine autonomie leren voor het experiment. Bedek de craniotomie met botcement en laat het cement drogen voordat u 4.0 polyamide hechtingsfilamenten gebruikt om de wond te sluiten.Na drie of vier hechtingen, stropdas 2-1-1 standaard chirurgische knopen. Injecteer de rat vervolgens intraperioonally met 250 microliters penicilline en controleer de rat elke 15 minuten tot ze volledig wakker zijn voordat het dier terugkeert naar zijn kooi. In deze analyse werden zes dieren onderworpen aan de operatie zoals aangetoond en zes dieren werden behandeld als controles.De hersensegmenten in deze beelden zijn afgeleid van één rat per groep. MRI-scan bleek dat het infarct was gelegen in de basis van de pons nabootsen anterolaterale infarcten bij menselijke patiënten. Omdat een geïsoleerde schede werd gebruikt, werd het infarct niet waargenomen in de cortex, cerebellum of midbrain.Diffusie gewogen beelden bleek ook de acute pontine infarct. TTC kleuring werd gebruikt om het infarct te bevestigen 24 uur na de operatie. Zoals verwacht was het infarctvolume aanzienlijk hoger in de operatiegroep in vergelijking met de controlegroep.Gedragsscores werden gemeten voor en na de operatie. Vergeleken met de controlegroep, ratten met een pontine infarct omcirkeld aan de linkerkant. Significante verschillen werden waargenomen in de Longa, Berderson, ledemaat plaatsing, bundel balans, en lijm verwijdering somatosensorische test scores tussen rants met pontine infarct en controle groep ratten.Elk specifiek hersengebied zal vernietigd worden door de elektrische stimulatie.
