RNA干扰（RNAi）是研究基因功能的一种强有力的研究工具，通过敲击一个特定的基因9。然而，长期稳定的基因敲解技术有一定的局限性，如探索对细胞生存至关重要的基因的功能的挑战。有条件的RNAi系统可用于通过诱导剂的管理，以时间和/或特殊控制的方式降低所需基因的调节。四环素（Tet）可应用系统是使用最广泛的条件RNAi系统10之一。Tet诱导系统可以通过在添加外源诱导剂（优待多氧环素，Dox）时控制shRNA的表达来诱导目标基因沉默。Tet-致失系统可分为两种类型：Tet-On 或 Tet-Off 系统。shRNA的表达可以在诱导器存在的情况下打开（Tet-On）或关闭（Tet-Off）。在没有诱导剂的 Tet-ON 系统中，构成表达的 Tet 抑制器 （TetR） 绑定到包含 Tet 响应 Pol III 依赖启动子的 Tet 响应元素 （TRE） 序列，从而抑制 shRNA 的表达。而加入 Dox 后，TetR 与 Tet 响应 Pol III 依赖的启动器隔离开来。这有利于 shRNA 的表达，并导致基因敲击。
