재생 의학은 나이, 질병 또는 외상으로 손상된 조직과 기관을 치유하거나 대체하고 선천적 결함을 교정하는 것을 목표로 합니다. 자가 요법에서는 세포나 조직을 환자에게서 빼내어 확장하거나 변형한 다음 기증자에게 다시 도입합니다. 이러한 형태의 치료제는 피부과 분야에서 광범위한 잠재력을 가지고 있다1. 자가 섬유아세포 치료에서는 환자의 섬유아세포를 배양하고 재주입하여 주름, 백합 또는 여드름 흉터를 치료합니다. 섬유아세포는 진피의 주요 기능세포이기 때문에 자가 섬유아세포 주사는 안면 회춘에 다른 치료법보다 더 유익할 수 있다2.
피부에서 섬유아세포는 세포외 단백질(즉, 콜라겐, 엘라스틴, 히알루론산 및 글리코사미노글리칸)의 합성 및 분비를 담당합니다. 또한 정상적인 피부 항상성 및 상처 치유에서 세포 기능, 이동 및 세포-기질/세포-세포 상호작용을 조절하는 성장 인자를 방출합니다3. 피부 섬유아세포는 피부 상처 치유를 위한 잠재적인 임상 세포 치료제로 이미 소개되었다.4, 조직 재생5 또는 심미와 성형 수술에서 피부 필러6. 일부 연구에서는 재생 의학의 맥락에서 섬유아세포가 중간엽 줄기세포보다 더 실용적이고 효과적인 세포 치료제가 될 수 있다고 제안하기도한다 7.
임상 적용을 위해 충분한 수의 섬유아세포를 얻으려면 일반적으로 세포 확장이 필수입니다. 생체 외/체외 세포 배양에는 세포 부착 및 증식을 지원하기 위해 성장 인자, 단백질 및 효소가 보충된 기본 배지가 필요합니다. 태아 소 혈청(FBS)은 태아 혈액이 1) 성인 혈액에 비해 성장 인자가 풍부하고 2)항체 함량이 낮기 때문에 세포 배양 배지의 일반적인 보충제입니다8. 세포 치료가 진행됨에 따라 FBS가 배양 배지에 첨가되는 고전적인 세포 배양 조건의 안전성에 대한 우려가 있습니다. 더욱이 FBS를 대안으로 대체하는 경향이 있습니다9. 여러 FBS 대체품이 유망한 결과를 보여주었습니다10.
혈소판 풍부 혈장(PRP) 대체품이 여기에서 선택되었으며, 당사는 CuteCell-PRP라는 PRP의 표준화된 제제를 생산하기 위한 의료 기기를 개발했습니다. 이 장치의 의도된 용도는 GMP 조건에서 자가 세포의 시험관 내 확장을 위한 배양 배지 보충제로 사용할 자가 PRP의 준비입니다.
