Dit werk biedt een gedetailleerd experimenteel protocol om de neurotoxiciteit van milieuverontreinigende stoffen met behulp van zebravissenlarven te evalueren. Zebravissen gaan tijdens de blootstellingsperiode door het proces van embryo's tot larven, wat betekent dat goede verzorging van de embryo's en larven essentieel is. Alles wat de ontwikkeling van de embryo's en larven beïnvloedt, kan het eindresultaat beïnvloeden. Hier worden de cultuuromgeving, het blootstellingsproces en de experimentele omstandigheden besprok...

In de afgelopen jaren zijn steeds meer milieuverontreinigende stoffen neurotoxisch gebleken1,2,3,4. De beoordeling van neurotoxiciteit in vivo na blootstelling aan milieuverontreinigende stoffen is echter niet zo eenvoudig als die van hormoonontregeling of ontwikkelingstoxiciteit. Bovendien heeft vroegtijdige blootstelling aan verontreinigende stoffen, met name bij milieurelevante doses, steeds meer aandacht getrokken in toxiciteitsstudies5,6,7,8.
Zebravissen wordt opgericht als een dier model geschikt voor neurotoxiciteit studies tijdens de vroege ontwikkeling na blootstelling aan milieuverontreinigende stoffen. Zebravissen zijn gewervelde dieren die zich na bevruchting sneller ontwikkelen dan andere soorten. De larven hoeven niet gevoed te worden omdat de voedingsstoffen in het chorion voldoende zijn om ze 7 dagen na bevruchting (dpf) te ondersteunen9. Larven komen uit de chorion op ~ 2 dpf en ontwikkelen gedrag zoals zwemmen en draaien dat kan worden waargenomen, bijgehouden, gekwantificeerd, en geanalyseerd automatisch met behulp van gedragsinstrumenten10,11,12,13 vanaf 3-4 dpf14,15,16,17,18. Daarnaast kunnen high-throughput tests ook worden gerealiseerd door gedragsinstrumenten. Zo zijn zebravissenlarven een uitstekend model voor de neurogedragsstudie van milieuverontreinigende stoffen19. Hier wordt een protocol aangeboden met behulp van hoge doorvoer monitoring om de neurogedragstoxiciteit van milieuverontreinigende stoffen op zebravissen larven onder lichte stimuli te bestuderen.
Ons lab heeft de neurogedragstoxiciteit bestudeerd van 2,2',4,4'-tetrabromodiphenylether (BDE-47)20,21, 6'-Hydroxy/Methoxy-2,2',4'-tetrabromodiphenylether (6-OH/MeO-BDE-47)22, deca-broomdineerde diphenylether (BDE-209 lood, en commerciële gechloreerde paraffine23 volgens het gepresenteerde protocol. Veel laboratoria gebruiken ook het protocol om de neurogedragseffecten van andere verontreinigende stoffen op larven of volwassen vissen te bestuderen24,25,26,27. Dit neurogedragsprotocol werd gebruikt om mechanistische ondersteuning te bieden waaruit blijkt dat blootstelling met lage dosis aan bisfenol A en vervanging bisfenol S voortijdige hypothalamische neurogenese bij embryonale zebravissen27veroorzaakte. Daarnaast optimaliseerden sommige onderzoekers het protocol om overeenkomstige studies uit te voeren. Een recente studie elimineerde de toxiciteit van amyloïde bèta (Aβ) in een eenvoudig, high-throughput zebravis model met caseïne-gecoate gouden nanodeeltjes (βCas AuNPs). Het toonde aan dat βCas AuNPs in systemische circulatie over de bloed-hersenbarrière van zebravissen larven en sekwestreerde intracerebrale Aβ42, het uitlokken van toxiciteit in een niet-specifieke, chaperonne-achtige manier, die werd ondersteund door gedragspathologie28.
Bewegingsvrijheid, padhoek en sociale activiteit zijn drie neurogedragsindicatoren die worden gebruikt om de neurotoxiciteitseffecten van zebravissenlarven te bestuderen na blootstelling aan verontreinigende stoffen in het gepresenteerde protocol. Bewegingsbeweging wordt gemeten door de zwemafstand van larven en kan worden beschadigd na blootstelling aan verontreinigende stoffen. Padhoek en sociale activiteit zijn nauwer verwant aan de functie van de hersenen en het centrale zenuwstelsel29. De padhoek verwijst naar de hoek van het pad van de dierlijke beweging ten opzichte van de zwemrichting30. Acht hoekklassen van ~-180°-~+180° zijn in het systeem ingesteld. Om de vergelijking te vereenvoudigen, worden zes klassen in het eindresultaat gedefinieerd als routinewendingen (-10° ~0°, 0° ~+10°), gemiddelde bochten (-10° ~-90°, +10° ~+90°), en responsieve bochten (-180° ~-90°, +90° ~+180°) volgens onze eerdere studies21,22. Sociale activiteit van twee vissen is fundamenteel van groepsshoalinggedrag; hier wordt een afstand van < 0,5 cm tussen twee larven geldig gedefinieerd als sociaal contact.
Het hier gepresenteerde protocol toont een duidelijk proces aan voor het bestuderen van neurogedragseffecten op zebravissenlarven en biedt een manier om de neurotoxiciteitseffecten van verschillende stoffen of verontreinigende stoffen te onthullen. Het protocol zal ten goede komen aan onderzoekers die geïnteresseerd zijn in het bestuderen van de neurotoxiciteit van milieuverontreinigende stoffen.

De laatste jaren zijn steeds meer milieuverontreinigende stoffen neurotoxisch gebleken, vooral in de vroege ontwikkelingsstadia van organismen. Zebravislarven zijn een bijuitstek model voor de neurobehavioral studie van milieuverontreinigende stoffen. Hier wordt een gedetailleerd experimenteel protocol verstrekt voor de evaluatie van de neurotoxiciteit van milieuverontreinigende stoffen met behulp van zebravissenlarven, waaronder het verzamelen van de embryo's, het blootstellingsproces, neurogedragsindicatoren, het testproces en data-analyse. Ook de cultuuromgeving, het blootstellingsproces en de experimentele omstandigheden worden besproken om het succes van de test te garanderen. Het protocol is gebruikt bij de ontwikkeling van psychopathische geneesmiddelen, onderzoek naar milieuneurotoxische verontreinigende stoffen, en kan worden geoptimaliseerd om overeenkomstige studies te maken of nuttig te zijn voor mechanistische studies. Ons protocol biedt een duidelijk proces voor het bestuderen van de neurogedragseffecten van de stoffen en de verontreinigende stoffen die van belang zijn voor zebravissenlarven en voor het onthullen van de potentiële neurotoxiciteit van deze stoffen. De techniek kan worden uitgevoerd met behulp van hoge doorvoer testen van neurobehaviors in de zebravis model. Deze methode kan worden toegepast om alleel-neurobehavioral effecten van verontreinigende stoffen te evalueren om potentiële neurotoxiciteit op ons mensen aan te tonen, aangezien zebravissen genoom een hoge homologie met mensen heeft, zodat het aanwijzingen voor chemisch beheer en de volksgezondheidsproblemen kan verstrekken die door verontreinigende stoffen worden veroorzaakt.Aangezien zebravissen het voordeel hebben van een hoge doorvoerscreening, geeft deze methode inzicht in zowel de ontwikkeling van psychotrope geneesmiddelen als onderzoek naar milieutoxicologie. Laat je niet intimideren over het bestuderen van het eerste experiment. Als je het protocol volgt, heb je een succesvol resultaat.Stel ten minste twee uur voor het uitvoeren van een experiment de kamertemperatuur in op 28 graden Celsius en voeg 800 microliter blootstellingsoplossing toe aan elke put van een 48-bron microplaat. Gebruik vervolgens een pipet van één milliliter met een gewijzigde tip om één larve in 200 microliter blootstellingsoplossing over te brengen op elke put van de microplaat ter voorbereiding op een locomotion- en padhoekexperiment. Om een zes-goed microplaat voor sociale gedragsexperimenten voor te bereiden, voeg vier milliliter blootstellingsoplossing aan elke put van de zes goedmicroplate toe en breng twee larven in 200 microliters van blootstellingsoplossing aan elke put over.Voordat u met de test begint, opent u het softwareprogramma voor hoge doorvoerbewaking en selecteert u de module spoorpadhoeken voor het bijhouden van rotaties. Breng de 48-well microplate naar het opnameplatform en trek de cover naar beneden. Klik op Bestand en Protocol genereren om te beginnen met het genereren van een nieuw protocol en voer 48 in de positie Locatietelling in.Klik op Parameters, Protocolparameters en Tijd, stel de duur van het experiment in op een uur en 10 minuten en de integratieperiode op 60 seconden. Teken met het elliptische vormgereedschap een cirkel linksboven goed. Selecteer de cirkel en klik op Kopiëren, Rechtsboven markeren, Plakken en Selecteren.Gebruik de muis om de gekopieerde cirkel goed naar rechtsboven te slepen en klik nogmaals op Kopiëren, Markering rechtsboven, Plakken en Selecteren. Sleep vervolgens de gekopieerde cirkel rechtsonder goed en klik op Markeringen maken en wissen. Het systeem zal automatisch een cirkel over elke andere put van de plaat tekenen.Klik vervolgens op Schaal tekenen en teken een kalibratielijn op het scherm. Voer de lengte van de regel in, stel het apparaat in en klik op Toepassen op groep. Stel de dierkleur op zwart en stel de detectiedrempel in op 16 tot 18 om detectie van de dieren mogelijk te maken, stel de inactieve snelheid op 0,5 centimeter per seconde en de kleine tot grote snelheid op 2,5 centimeter per seconde.Stel de padhoekklassen in van min 180 naar plus 180 zoals aangegeven. Als u de lichtomstandigheden wilt instellen, klikt u op Parameters, Licht rijden, gebruikt u een van de 3 triggeringmethoden hieronder en verbeterde stimuli om de lichtomstandigheden in te stellen. Klik op Rand en stel een donkere periode van 10 minuten in, gevolgd door drie cycli van afwisselende licht- en donkere perioden van 10 minuten.Sla vervolgens het protocol op. Wanneer alle parameters zijn ingesteld, schakelt u de verlichting in de testruimte uit en laat u de larven gedurende 10 minuten in het systeem acclimatiseren. Klik aan het einde van de acclimatisatieperiode op Experimenteren en Uitvoeren en maak de map waarin de experimentele bestanden moeten worden opgeslagen.Voer vervolgens de resultaatnaam in en klik op Achtergrond en Start. Klik aan het einde van het experiment op Experimenteren en Stoppen. Breng de 48-bron microplaat terug naar de lichtincubator voor latere experimenten.Omdat elke groep 16 dieren kan bevatten, kan het systeem worden gebruikt om hoge doorvoertests van de bewegings- en padhoek uit te voeren onder verschillende behandelingsomstandigheden in één microplaat van 48 put. De sociale activiteit van de larven kan worden getest met behulp van zes-put platen. In dit representatieve experiment vertoonde de hoogste concentratiegroep van BDE-47 een uitgesproken hypoactiviteit tijdens de donkere periode, terwijl er geen veranderingen als gevolg van BDE-47-blootstelling werden waargenomen tijdens de lichtperiodes.Verder voerde de groep met hoge concentratie van 6-hydroxy-BDE-47 minder routine- en gemiddelde bochten uit na vijf dagen na de bevruchting. Echter, meer responsieve bochten werden geïnduceerd in de 6-methoxy-BDE47 blootstelling groepen. De sociale activiteit van de zebravis werd gestimuleerd door gechloreerde paraffine-70 en de korte keten gechloreerde paraffine-52b, terwijl de lange keten gechloreerde paraffine-52a de duur voor contact van de larven verkortte.Het is belangrijk om ervoor te zorgen dat de overgedragen larven voor de test een normaal zwemvermogen en geen misvormingen moeten hebben. Andere variabelen, zoals kleurvoorkeur, licht-donkere voorkeur, alle duisternis, staartbewegingen van zebravissen, kunnen worden opgenomen om aanvullende vragen over neurobehaviorale toxiciteit van milieuverontreinigende stoffen te beantwoorden. De BDE-47 blootstellingsoplossingen zijn neurotoxisch en hebben hormoonontregelingspotentieel, dus zorg ervoor dat u direct contact met de huid vermijdt.
