Synthetische cellen zijn kunstmatige celachtige deeltjes, die een of meerdere functies van een levende cel nabootsen, zoals het vermogen om te verdelen, membraaninteracties te vormen en eiwitten te synthetiseren op basis van een genetische code1,2,3. Synthetische cellen die celvrije eiwitsynthese (CFPS) systemen omsluiten bezitten een hoge modulariteit vanwege hun vermogen om verschillende eiwitten en RNA sequenties te produceren na veranderingen in de DNA-sjabloon. CFPS-systemen zijn gebaseerd op cellysaat, gezuiverde componenten of synthetische componenten en bieden een aantrekkelijk alternatief voor de huidige benaderingen van eiwitproductie en omvatten alle transcriptie- en vertaalmechanismen die nodig zijn voor eiwitsynthese, zoals ribosomen, RNA-polymerase, aminozuren en energiebronnen (bijvoorbeeld 3-fosfhoglyceren en adenine trifosfaat)4,5,6,7,8,9. De inkapseling van een CFPS-systeem in lipideblaasjes maakt de eenvoudige en efficiënte productie van eiwitten mogelijk zonder afhankelijk te zijn van een levende cel10. Bovendien, dit platform maakt synthese van peptiden die kunnen degraderen in natuurlijke cellen, de productie van eiwitten die giftig zijn voor levende cellen, en eiwitten te wijzigen met niet-natuurlijke aminozuren11,12. Synthetische cellen zijn gebruikt als een model voor onderzoeksdoeleinden het onderzoeken van de minimale celcomponenten die nodig zijn om cellulair leven mogelijk te maken vanuit een evolutionair perspectief1,13. Synthetische cellen zijn ook gebruikt voor het bouwen en implementeren van genetische circuits en als modellen voor gerichte evolutie14,15,16. Andere studies hebben zich gericht op het vermogen van synthetische cellen om de biologische activiteit van natuurlijke cellen na te bootsen, gericht op het vervangen van beschadigde natuurlijke cellen, zoals bètacellen bij patiënten met diabetes17. Bovendien illustreert het vermogen van deze CFPS om synthetische cellen in te kapselen om een verscheidenheid aan therapeutische eiwitten te produceren, het potentieel ervan om in klinisch gebruik te worden opgenomen18.
Hier beschrijven we een bottom-up lab-schaal protocol (Figuur 1) voor de productie van RNA en eiwitproducerende synthetische cellen op basis van een CFPS-systeem ingekapseld in een lipide blaasje. Dit toont het mogelijke gebruik van synthetische celplatforms als nieuwe systemen voor de levering van geneesmiddelen voor de on-site productie van een therapeutisch eiwitmedicijn in vivo19. Eerdere studies hebben de optimalisatie van de CFPS-reactie en de cellysaatvoorbereidingsprocessen4,8,20onderzocht. Bovendien zijn verschillende technieken toegepast voor liposoompreparaat ter grootte van celformaat, zoals microfluïdische en polymeergebaseerde druppelstabilisatiemethoden21,22,23, die ook verschillen in de lipidesamenstelling van liposomen24,25,26. In het gepresenteerde protocol worden synthetische cellen geproduceerd met behulp van een water-in-olie emulsieoverdrachtsmethode en wordt het inkapselingsproces uitgevoerd bij lage temperaturen (<4 °C)5,10,24,27,28. Deze milde omstandigheden bleken gunstig te zijn voor het behoud van de bio-functionele integriteit van de moleculaire machines, namelijk ribosomen en eiwitten27,29,30. De lipidesamenstelling van de deeltjes bestaat uit zowel cholesterol als 1-palmitoyl-2-oleoyl-sn-glycero-3-fosfocholine (POPC). De eerste is te vinden in alle zoogdiercelmembranen en is essentieel voor de stabiliteit, stijfheid en doorlaatbaarheid vermindering van het membraan, en de laatste bootst zoogdier fosfolipide samenstelling11,13. De cellulaire transcriptie en vertaling moleculaire machines worden gewonnen uit de BL21 (DE3) Escherichia coli (E. coli) stam, die wordt getransformeerd met pAR1219 plasmid overexpressing T7 RNA polymerase om CFPS potentie en eiwitsynthese te verhogen. Dit systeem is gebruikt voor de productie van diagnostische en therapeutische eiwitten, met moleculaire gewichten tot 66 kDa in vitro en in vivo19,31. De bottom-up assemblage benadering voor de bouw van synthetische cellen is een effectief instrument voor het isoleren en onderzoeken van cellulaire processen in een cel nabootsen omgeving. Bovendien heeft de ontwikkeling van celvrije expressiesystemen aangetoond dat het vermogen is om de eiwitproductie-, transcriptie- en vertaalprocessen (DNA→RNA→eiwit) op een gecontroleerde manier te reconstrueren, waarbij synthetische biologie wordt gebruikt. Hier beschrijven we een protocol voor de voorbereiding van een cel-vrije expressie systeem, met inbegrip van de productie van een krachtige bacteriële lysaat en inkapselen dit lysaat binnen cholesterol-rijke lipide-gebaseerde gigantische unilamellar vesicles (GUVs) (dat wil zeggen, stabiele liposomen), om synthetische cellen te vormen. Het protocol beschrijft de methoden voor de voorbereiding van de componenten van de synthetische cellen, met inbegrip van de productie van actieve bacteriële lysaten, gevolgd door een gedetailleerde stapsgewijze voorbereiding van de synthetische cellen op basis van een water-in-olie emulsie overdracht methode. Preparing Protein Producing Synthetic Cells using Cell Free Bacterial Extracts, Liposomes and Emulsion Transfer

Eiwitproducerende synthetische cellen bieden een veelzijdig platform voor het bestuderen van de oorsprong van het leven en synthetische celgebaseerde therapieën voor geavanceerde drugslevering. Deze methode is eenvoudig en betaalbaar voor RNA en eiwitexpressie en kan worden toegepast voor on-site therapeutische eiwitproductie direct in het lichaam. De systemen kunnen worden gebruikt voor de behandeling van een breed scala van ziekten van metabole ziekten en eiwitvervangers tot kanker en eventueel diabetes.Dit protocol in meerdere stappen heeft een gedefinieerd stoppunt. Bij het uitvoeren van het voor de eerste keer, is het raadzaam om het werk te verdelen over meerdere dagen. Het protocol voor synthetische celproductie begint met de voorbereiding van de celvrije S30-T7 lysaat.Vervolgens zijn de membraanlipiden en de cellulaire voedingsoplossing voorbereid om de innerlijke omgeving te simuleren en de eiwitproductie te ondersteunen. De laatste stap is de bereiding van de synthetische cellen zelf. Bereid dubbele starter culturen in 100 milliliter Erlenmeyer kolven.Voeg aan elke kolf vijf milliliter LB-media toe, aangevuld met 50 microgram per milliliter ampicilline en inentule met één E.coli-kolonie. Kweek de cultuur 's nachts in een vloer incubator shaker bij 250 rpm en 37 graden Celsius. Bereid vervolgens twee liter verbijsterde Erlenmeyer-kolven met 500 milliliter tb-media, aangevuld met 50 microgram per milliliter ampicilline.Inenting elke twee liter kolf met een vijf milliliter starter cultuur. Plaats de kolven in een shaker bij 250 tpm en 37 graden Celsius. Monitor de culturen periodiek met behulp van een spectrofotometer en laat de culturen groeien tot OD600 tussen 0,8 en 1.Om T7 RNA polymerase expressie te induceren, voeg aan elke kolf drie milliliter van 100 millimolar IPTG toe om 0,6 millimolar te bereiken. Resuspend elke pellet in 250 milliliter koude S30 lysate buffer met behulp van een roerder indien gewenst.Centrifuge op 7.000 keer g gedurende 10 minuten bij vier graden Celsius. Gooi de supernatant weg en verleg alle pellets samen in 15 milliliter koude S30 lysate buffer. Filter de vering met gaaspads.Voordat u doorgaat naar de volgende stap, u de tips vooraf koelen in 1,5 milliliter flesjes die nodig zijn voor het opslaan van het lysaat. Voer de stap snel uit om de lysate-activiteit te behouden.Plaats 200 microliter aliquots van supernatant in voorgekoelde flesjes en onmiddellijk snap bevriezen met vloeibare stikstof. Bewaar de flesjes op negatieve 80 graden Celsius voor toekomstig gebruik. Los POPC en cholesterol in chloroform afzonderlijk op tot een uiteindelijke concentratie van 100 milligram per milliliter.Vortex elk flesje afzonderlijk. Combineer de componenten in een twee milliliter glazen flacon. Voeg 50 microliter van de cholesterol-chloroform oplossing, 50 microliter van de POPC-chloroform oplossing, en 500 microliters van minerale olie.Vortex de flacon, dan om de chloroform verdampen, verwarm het voor ongeveer een uur op 80 graden Celsius in een chemische kap. Na het uitbroeden van de resulterende emulsie gedurende 10 minuten op gemalen ijs, langzaam voeg de emulsie op de top van de oliefase in de 15 milliliter buis. Centrifugeren de buis gedurende 10 minuten op 100 keer g en vier graden Celsius.Dan centrifugeren gedurende 10 minuten op 400 keer g en vier graden Celsius. Tegen het einde van de centrifugatie moet een pellet aan de onderkant van de buis waarneembaar zijn. Om de pellet te extraheren, verwijder eerst de overtollige olielaag.Laad vervolgens een bijgesneden pipetpunt met ongeveer 400 microliters buitenoplossing en gebruik de pipetpunt om de pellet te verzamelen, waardoor de buitenste oplossing vrijkomt als de pipetpunt door de oliefase gaat. Veeg de pipetpunt af en breng de pellet over op een schone buis van 1,5 milliliter. Centrifugeren de pellet gedurende 10 minuten op 1,000 keer g en vier graden Celsius.Verwijder de supernatant en resuspend de pellet in 100 microliter van voeding oplossing. Voor eiwitexpressie, incubeer de suspensie gedurende twee uur bij 37 graden Celsius zonder te schudden. Plasmids die het model eiwit sfGFP en Renilla luciferase uitdrukken werden geïntroduceerd in celvrije eiwitsynthese, CFPS bulkreacties en synthetische cellen.De eiwitproductie werd geëvalueerd met behulp van verschillende methoden. Westerse vlek analyse gedetecteerd sfGFP His6 productie in zowel CFPS bulk reacties en in synthetische cellen. Gezuiverde sfGFP His6 werd gebruikt als een positieve controle.Een fluorescerende microscoop met een Brightfield en een GFP filter tonen synthetische cellen die sfGFP produceren. Flow cytometrie werd gebruikt om sfGFP producerende synthetische cellen te analyseren. De berekening van de actieve synthetische celpopulatie was gebaseerd op de sfGFP-fluorescentieintensiteitsdrempel gedefinieerd door het negatieve DNA-monster dat wordt vertegenwoordigd door het rode histogram.Gemiddelde fluorescentie intensiteit van sfGFP produceren synthetische cellen werd berekend van de actieve synthetische cel populatie en genormaliseerd tot de negatieve DNA-monster. De diameterverdelingen van de actieve synthetische cellen werden berekend op basis van het sfGFP-signaal. Renilla luciferase activiteit werd gekwantificeerd met luminescentie metingen.Deze methode is zeer veelzijdig en kan worden geoptimaliseerd voor verschillende doeleiwitten door het veranderen van lysate oorsprong, lipide samenstelling en innerlijke oplossing concentraties. FACS-analyse van synthetische cellen die een fluorescerend markereiwit produceren, kan worden uitgevoerd om kwantitatieve waarde te leveren van de fractie van actieve synthetische cellen. Westerse vlekanalyse zou de eiwitproductie meten.
