Dit protocol werd gebruikt om de chemische vorm van selenium en ijzer in biologische monsters te bestuderen door middel van röntgenabsorptiespectroscopie. Het richt zich op de cryo-voorbereiding en opslag van biologische monsters en referentieverbindingen, evenals op de HERFD-XAS-metingen.

De studie van de cellulaire biotransformaties van essentiële of toxische elementen vereist soortvormingstechnieken met een hoge gevoeligheid en moet monstervoorbereidingsstappen minimaliseren die vaak vatbaar zijn voor modificatie van chemische soorten.
Fysiologische elementen zoals selenium en ijzer staan erom bekend bijzonder moeilijk te specificeren zijn vanwege hun complexe chemie, verschillende stabiliteiten van de selenium- of ijzersoorten en hun lage concentratie in het ppm (mg / kg) of zelfs sub-ppm-bereik. De studie van de soortvorming van deze elementen door XAS kan dus uiterst uitdagend zijn. Synchrotron XAS en met name hoge energie resolutie fluorescentie gedetecteerde XAS (HERFD-XAS), die een zeer lage signaal-tot-achtergrondverhouding1 mogelijk maakt, zijn beschikbaar bij synchrotronbronnen om sterk verdunde elementen in complexe biologische matrices te speciëren 2,3. Conventionele fluorescentie-XAS-metingen kunnen worden uitgevoerd met behulp van een energy resolved solid state detector (SSD) met een energiebandbreedte ~ 150-250 eV, op de CRG-FAME-bundellijn bij de European Synchrotron Radiation Facility (ESRF)4, terwijl HERFD-XAS-metingen een kristalanalysatorspectrometer (CAS) nodig hebben, met een energiebandbreedte ~ 1-3 eV, op de CRG-FAME-UHD-bundellijn op de ESRF2 . Fluorescentiefotonen worden gediscrimineerd met betrekking tot hun energie met respectievelijk elektronische of optische processen.
De cryopreparaat van het monster is essentieel om structuren te behouden en de chemische integriteit van de samenstelling te behouden, waardoor analyse dicht bij de biologische inheemse toestand5 mogelijk is. Bovendien zorgen analyses uitgevoerd bij cryogene temperaturen zo laag als 10 K met behulp van vloeibare helium cryogene koeling (LN2), ervoor dat stralingsschade vertraagt en elementaire speciatie voor XAS behoudt. Hoewel sommige beoordelingen van XAS-technieken toegepast op biologische monsters de noodzaak melden om monsters in cryogene omstandigheden voor te bereiden en te analyseren (bijv. Sarret et al.6, Porcaro et al.7), beschrijft geen van hen duidelijk het gerelateerde gedetailleerde protocol. In deze publicatie wordt een methode beschreven voor cryo-voorbereiding van kankercellen en planktonmicro-organismen voor HERFD-XAS-speciatie van Se8 en Fe9 bij cryogene temperatuur.
Goede praktijken voor monstervoorbereiding en omgeving tijdens state-of-the-art XAS-spectroscopiemetingen vereisen 1) een opstelling; 2) een analyseprocedure die de effecten van stralingsschade zoveel mogelijk beperkt; en 3) een monster (of model samengestelde referentie) dat zo homogeen mogelijk is ten opzichte van de bundelgrootte van röntgenfotonen. Het eerste item wordt in aanmerking genomen door de acquisitie bij een lage temperatuur uit te voeren, met behulp van een vloeibare helium cryostaat. Het tweede item wordt behandeld door elke acquisitie uit te voeren op een vers deel van het monster door het ten opzichte van de balk te verplaatsen. Ten slotte worden, rekening houdend met de derde voorwaarde, monsters (pellets) en referenties (poeders) geconditioneerd in geperste bulkpellets om porositeiten en inhomogeniteiten zoveel mogelijk te beperken en ruwheid te voorkomen met betrekking tot de bundelgrootte op het oppervlak van het röntgensonde monster. We leggen uit hoe het protocol met al deze punten omgaat.
We gebruikten menselijke prostaatcellijn PC-3 (hoog gemetastaseerd potentieel) en ovariumcellijn OVCAR-3 (die goed is voor maximaal 70% van alle gevallen van eierstokkanker) om de antiproliferatieve eigenschappen ten opzichte van kankercellen van selenium nanodeeltjes (Se-NP's) en Phaeodactylum tricornutum diatomeeën te onderzoeken als een modelsoort om ijzervastlegging in fytoplankton te onderzoeken.

biological samples preparation for speciation at cryogenic temperature using high-resolution  x-ray absorption spectroscopy

De studie van elementen met röntgenabsorptiespectroscopie (XAS) is van bijzonder belang bij het bestuderen van de rol van metalen in biologische systemen. Monstervoorbereiding is een belangrijke en vaak complexe procedure, met name voor biologische monsters. Hoewel röntgenspeciatietechnieken veel worden gebruikt, is er nog geen gedetailleerd protocol verspreid voor gebruikers van de techniek. Verder is chemische toestandsmodificatie van belang en worden cryo-gebaseerde technieken aanbevolen om de biologische monsters in hun bijna-inheemse gehydrateerde toestand te analyseren om het maximale behoud van de chemische integriteit van de cellen of weefsels te bieden. Hier stellen we een cellulair voorbereidingsprotocol voor op basis van cryo-geconserveerde monsters. Het wordt aangetoond in een hoge energie resolutie fluorescentie gedetecteerdE röntgenabsorptie spectroscopie studie van selenium in kankercellen en een studie van ijzer in fytoplankton. Dit protocol kan worden gebruikt met andere biologische monsters en andere röntgentechnieken die door bestraling kunnen worden beschadigd.

De belangrijkste doelen van deze preparaten waren het onderzoeken van de interactie tussen selenium nanodeeltjes (Se-NP's) en kankercellen, en ijzerbinding en sekwestratie in fytoplankton.

Biological Samples Preparation for Speciation at Cryogenic Temperature using High-Resolution  X-Ray Absorption Spectroscopy

Dit protocol is gestandaardiseerd voor cellulaire monsters, waardoor experimenten die op verschillende synchrotronlocaties worden uitgevoerd, kunnen worden vergeleken. Het belangrijkste voordeel van deze techniek is een eenvoudige en ongecompliceerde workflow die een snelle en betrouwbare conservering van de chemische integratie van het monster mogelijk maakt. Personen die nieuw zijn in deze techniek kunnen worstelen met de pelletering van bevroren iteratieve cellen en het snel laden van de cellulaire pellet in de cryosamplevolgorde.Een nauwkeurige en snelle ontkoppeling van een monster bij cryogene temperatuur is van cruciaal belang. Als zodanig is een visuele demonstratie essentieel om te begrijpen hoe de techniek moet worden uitgevoerd. Om menselijke prostaat- en eierstokkanker cellijncelkorrels voor seleniumspeciatie te bereiden, zaait u het juiste aantal cellen van elke cellijn in drie T-75-kolven per toestand in het juiste celkweekmedium in een laminaire stroomkap en plaatst u de kolven in een laminaire stroomkap in een incubator voor celkweek van 37 graden Celsius en 5% kooldioxide totdat de cellen 80% confluent zijn.Om de kankercellen bloot te stellen aan seleniumbehandeling, eerst vers bereide nanodeeltjesvoorraadoplossingen in een ultrasoon waterbad gedurende 30 minuten bij kamertemperatuur te soniceren voordat de selenium nanodeeltjesoplossing in volledig celkweekmedium serieel wordt verdund tot de juiste werkconcentraties. Was de cellen in de laminaire stromingskap voorzichtig twee keer met vijf milliliter van 37 graden Celsius PBS per wasbeurt en gebruik een steriele pipet van 25 milliliter om voorzichtig 15 milliliter van de seleniumbehandeling van belang toe te voegen aan de bodem van de kolf. Sluit de deksels zonder de kolf volledig af te sluiten en plaats de kolven gedurende 24 uur horizontaal in de celkweekincubator.Om de celkorrels te bereiden na het wassen en aanvullen van het kweekmedium, gebruikt u één celschraper per kolf om de cellen voorzichtig los te maken. Gebruik medium om alle cellen die aan de kolf vastzitten in het supernatant te spoelen en verzamel de gedissocieerde cellen door centrifugering. Was de pellets twee keer in vijf milliliter PBS per buis per wasbeurt om alle resterende sporen van de behandeling te verwijderen en de cellen opnieuw op te suspenderen in één milliliter PBS.Breng de cellen over in een polypropyleenbuis van 1,5 milliliter en verzamel de cellen met een andere centrifugatie. Gebruik vervolgens een pipet van 200 microliter om voorzichtig al het supernatant te verwijderen en het onderste deel van elke buis van 1,5 milliliter in vloeibare stikstof te storten tot het niveau van de celkorrel. Breng de buizen onmiddellijk na het invriezen over in een vloeibare stikstofdewar voor langdurige opslag.Optimaliseer voor hoge resolutie röntgenabsorptiespectroscopie alle kristallen van de kristalanalysatorspectrometer in Bragg-omstandigheden met betrekking tot de fluorescentielijnenergie van belang en gebruik een referentie waarvan bekend is dat de energiepositie van de absorptierand de invallende monochromatische bundelenergie kalibreert. Breng de monsterhouder over naar een vloeibare helium cryostaat voor biologische monsters en stel de cryostaat in op 10 graden Kelvin, sluit vervolgens het experimentele hok volgens de synchrotronveiligheidsregels en begin met de analyse. Hier tonen representatieve hoge resolutie röntgenabsorptiespectroscopiespectroscopiespectra van het selenium in de begintoestand en in cellen geïncubeerd tot voedingsmedium aan dat het selenium in de initiële seleniumnanodeeltjes aanwezig was als zowel selenium nul als cellulitisachtige vormen.Na interacties met de PC3-cellen was het selenium echter voornamelijk aanwezig in de cellen als selenium nul, wat een verandering van seleniumsoorten in de cellen aantoont. Voor ijzer vertonen röntgenabsorptiespectroscopiereferentiespectra met hoge resolutie verschillende randposities, afhankelijk van de ijzeroxidatietoestand, waarbij verminderde ijzersoorten worden verschoven naar lage energiewaarden. Twee opeenvolgende spectra verzameld op dezelfde positie van een diatomeeënkorrel waren vergelijkbaar, wat aangeeft dat de bundelschade beperkt was tussen twee acquisities bij gebruik van een helium cryostaat op 10 Kelvin.Bovendien waren spectra van verschillende posities van de diatomeeënkorrel identiek, wat aantoont dat de monsterpellet homogeen was en dat de spectra konden worden gemiddeld om een beter signaal-ruisverhoudingsspectrum te verkrijgen. De meest kritische stappen zijn de bereiding van de nanodeeltjesdiltuion, pelleteren bij cryogene temperatuur en het prepareren van de standaard referentiemonsters. Deze procedure kan worden toegepast op andere bulkanalysetechnieken, zoals ICP-MS of HPLC-ICP-MS of andere niet-synchrotronspectroscopische technieken die kunnen worden uitgevoerd bij cryogene temperatuur.Andere wetenschappelijke vragen kunnen worden onderzocht met behulp van deze procedure, met name op het gebied van biogeologie, biochemie en milieuwetenschappen of toxicologisch onderzoek. Seleniumverbindingen en selenium nanodeeltjes kunnen net zo zwaar zijn voor uw gezondheid en moeten worden gemanipuleerd in een speciale chemische kap met handschoenen, een bril en een gezichtsmasker.
