Hier presenteren we een methodologie die een voorbijgaande activering van endogene ROS-productie in vivo in muizenhuid met fysiologische effecten mogelijk maakt. De methodologie is gebaseerd op een fotodynamische procedure om een gecontroleerde en lokale stimulatie van de endogene fotosensitizer PpIX te induceren (figuur 1B). Deze experimentele benadering is een interessant hulpmiddel om ROS-biologie te bestuderen in in vivo experimentele systemen die een aanzienlijke vooruitgang betekenen t...

Reactieve zuurstofsoorten (ROS) zijn het resultaat van de chemische reductie van moleculaire zuurstof tot water, en omvatten singletzuurstof, superoxide-anion, waterstofperoxide en het hydroxylradicaal 1,2,3. ROS hebben een zeer korte levensduur vanwege hun extreem chemisch reactieve karakter. In aerobe organismen wordt ROS incidenteel gevormd in de cellen als een belangrijk lekkend bijproduct van aerobe ademhaling (elektronentransportketen) in de mitochondriën. Voorbijgaande accumulatie van hoge niveaus van ROS in de cel resulteert in een oxidatieve stressconditie die de onomkeerbare inactivatie van eiwitten, lipiden en suikers en de introductie van mutaties in het DNA-molecuul 2,3,4,5 kan veroorzaken. De geleidelijke accumulatie van oxidatieve schade in cellen, weefsels en hele organismen neemt gestaag toe met de tijd en is in verband gebracht met de inductie van celdoodprogramma's, verschillende pathologieën en het verouderingsproces 2,3,4,6.
Aërobe organismen hebben gestaag efficiënte moleculaire mechanismen ontwikkeld om overtollige ROS-accumulatie in cellen en weefsels aan te pakken. Deze mechanismen omvatten leden van de superoxide dismutase (SOD) eiwitfamilie, die superoxide radicale desmutatie in moleculaire zuurstof en waterstofperoxide katalyseren, evenals verschillende catalasen en peroxidasen die de antioxidantpool (glutathion, NADPH, peroxiredoxine, thioredoxine 7,8) gebruiken om de daaropvolgende omzetting van waterstofperoxide in water en moleculaire zuurstof te katalyseren.
Verschillende rapporten ondersteunen echter de rol van ROS als belangrijke componenten van moleculaire circuits die kritieke celfuncties reguleren, waaronder proliferatie, differentiatie en mobiliteit 2,3,4. Dit concept wordt verder ondersteund door de initiële identificatie en karakterisering van speciale ROS-producerende mechanismen in aërobe organismen, waaronder lipoxygenases cyclooxygenases en NADPH-oxidasen 9,10. In die zin vertoont ROS een actieve rol tijdens de ontwikkeling van gewervelde embryo's 11,12,13 en sleutelrollen voor deze moleculen in de regulatie van specifieke in vivo fysiologische functies zijn gemeld in verschillende experimentele systemen, waaronder het differentiatieprogramma van hematopoietische voorlopers in Drosophila14, genezingsinductie bij zebravissen of staartregeneratie bij Xenopus-kikkervisjes 15. Bij zoogdieren is ROS betrokken geweest bij het zelfvernieuwings-/differentiatiepotentieel van neurale stamcellen in een neurosfeermodel16 en bij de deregulatie van de darmstamcelfunctie tijdens de initiatie van colorectale kanker17. In de huid is ROS-signalering in verband gebracht met epidermale differentiatie en de regulatie van de huidstamcelniche en de haarfollikelgroeicyclus18,19.
In dit perspectief is een belangrijke experimentele beperking om de fysiologische rollen van ROS in biologische systemen te bepalen, zowel in normale als pathologische omstandigheden, het gebrek aan adequate experimentele hulpmiddelen om gecontroleerde productie van deze moleculen in cellen en weefsels te induceren, die nauwkeurig lijken op hun fysiologische productie als tweede signaalboodschappers. Op dit moment omvatten de meeste experimentele benaderingen de toediening van exogene ROS, meestal in de vorm van waterstofperoxide. We hebben onlangs een experimentele aanpak geïmplementeerd om een voorbijgaande, niet-dodelijke in vivo productie van endogene ROS in de muizenhuid in te schakelen, gebaseerd op de toediening van precursoren van de endogene fotosensitizer protoporfyrine IX (PpIX; bijv. aminolaepilinezuur of het methylderivaat methylaminolevulinaat daarvan) en verdere bestraling van het monster met rood licht om de in situ vorming van ROS uit intracellulaire moleculaire zuurstof te induceren (figuur 1). Deze fotodynamische procedure kan efficiënt worden gebruikt om residente stamcelniches te stimuleren, waardoor de regeneratieve programma's van het weefselworden geactiveerd 19,20 en de weg wordt geopend voor nieuwe therapeutische modaliteiten in de huidregeneratieve geneeskunde. Hier presenteren we een gedetailleerde beschrijving van het protocol, met representatieve voorbeelden van stimulatie van stamcelniches, gemeten als een toename van het aantal langdurige 5-broom-2'-deoxyuridine (BrdU) labelbehoudende cellen (LRC's) in het uitstulpende gebied van de haarfollikel19,21, en daaropvolgende activering van regeneratieprogramma's (versnelling van haargroei en brandwondgenezingsprocessen) geïnduceerd door voorbijgaande, niet-dodelijke ROS-productie in de huid van de C57Bl6-muizenstam.

Hier beschrijven we een protocol om schakelbare in vivo fotogeneratie van endogene reactieve zuurstofsoorten (ROS) in de huid van muizen te induceren. Deze voorbijgaande productie van ROS in situ activeert efficiënt celproliferatie in stamcelniches en stimuleert weefselregeneratie zoals sterk gemanifesteerd door de versnelling van brandwondengenezing en haarfollikelgroeiprocessen. Het protocol is gebaseerd op een gereguleerde fotodynamische behandeling die het weefsel behandelt met voorlopers van de endogene fotosensitizer protoporfyrine IX en het weefsel verder bestraalt met rood licht onder streng gecontroleerde fysisch-chemische parameters. Over het algemeen vormt dit protocol een interessant experimenteel hulpmiddel om de ROS-biologie te analyseren.

De topische toediening van de mALA-voorloper in de rug- en staarthuid van de muis resulteert in een significante accumulatie van PpIX in het hele weefsel en, merkbaar, in de haarfollikel, zoals aangetoond door de roodroze fluorescentie van deze verbinding onder excitatie van blauw licht (407 nm) (figuur 2A,C). Latere bestraling van behandeld weefsel met rood licht (636 nm) bij een fluence van 2,5−4 J/cm 2 bevordert de voorbijgaande productie van ROS in het weefs...
