성숙한 PDX 종양 조직은 이질적인 인간 암세포 및 마우스 기질 (섬유아세포, 내피 세포 등)의 가변 혼합물을 포함한다. 체외에서 종양 진행에 대한 세포 형 특이적 기여를 연구하기 위해 종양을 해리하고 세포 집단을 분리하며 세포 간 통신의 경로를 해부하는 조직된 방식으로 실험적으로 통합하는 것이 유리할 수 있습니다. 조직 발굴내의 혼합 세포 인구는 특정 배양 조건과의 차동 적 호환성을 갖는다. 예를 들어, 종양 관련 섬유아세포 생존가능성은 인테그린 리간드로 기능화된 표면 준수 또는 3D 행렬을 필요로 하며, 상피 유래 PDX 세포는 전형적으로 이러한 요구 사항을 갖지 않고 세포 세포 상호 작용을 선호합니다. 이러한 차이는 마우스 기질 세포를 오염에서 PDX 세포의 효과적인 분리를 달성하기 위해 악용 될 수있다. 조직 발굴의 회전 배양은 세포-세포 접착이 24-48h에서 상체체에 다세포 클러스터를 형성하기 위해 회전 배양 표면 위에 떠 있는 PDX 세포를 구동하는 동안 조직 배양 표면에 기질 세포 부착을 허용합니다. 이러한 클러스터의 특정 특성은 PDX(예: 크고, 단단하고, 구형군이 많거나, 포도 무리를 닮은 더 작고, 느슨한 골재)와 다르지만, 일반적으로 생물학적으로 관련된 크기(50-250 μm 직경)에 따라 세포 간 접촉에 의존하는 세포 상호 작용을 평가하기에 충분합니다.

3D 배양에서 PDX 유래 클러스터의 연구를 강화하기 위해, 우리는 384 웰 마이크로티터 플레이트에 최대 96 개의 개별, 3D 배양의 동시 배양을 허용하는 높은 처리량 오르간 온 - 칩 플랫폼인 미세 유체 기반 관류 배양7플랫폼인OrganoPlate(그림1)를활용했습니다. 2차선 미세유체 플레이트에서 단일 티슈 칩은 2개의 미세유체채널(도 1B,젤 채널: 빨강, 관류 채널: 블루)에 의해 연결됩니다. 두 개의 미세 유체 채널은 한 채널의 인접 한 채널의 오버플로를 방지하는 Phaseguide라는 짧은 플라스틱 능선으로 분리되며 동시에 젤과 관류 채널9의내용 사이의 막 없는 인터페이스를 허용합니다. 미세 유체 판의 바닥은 현미경 등급 유리로 구성되기 때문에, 배양은 표준 또는 자동화 된 현미경으로 플레이트의 바닥을 통해 관찰 창에서 볼 수 있습니다. 관류는 프로그래밍 가능한 로커가 있는 미세유체 판에 설치되어 중력을 사용하여 미세유체 채널을 통해 미디어를 구동하며, 저수지우물(도 1C)을통해 미디어를 구동한다. 관류 흐름 모방은 정적 배양보다 종양 미세 환경을 더 밀접하게 재구성하여 전단 응력의 통합과 가스와 영양소의 향상된 분포를 허용합니다. 미세유체판에서 관융합된 암 세포 배양을 유지하는 이점은 이전에 동일한 세포의 정적 3D 배양에 비해 최적의 생존가능성을 나타내는 것으로 설명되어 있다7.
본 보고서는 라이브 다세포 PDX 클러스터를 격리하기 위한 적응된 밀도 그라데이션 원심분리 방법을 설명하고, 구불구형 미세 유체 플레이트 내에서 3D PDX 배양을 확립하는 데 있어 유용성을 입증합니다. PDX 사용을 용이하게 하는 방법을 모색하는 연구 실험실의 수가 늘어나고 있기 때문에 여기에 제시된 프로토콜이 즉각적인 유용성일 것으로 예상됩니다.
