Het is aangetoond dat vaccins op basis van capsules effectief zijn tegen talrijke bacteriële pathogenen, en velen zijn gelicentieerd voor gebruik bij mensen25,26,27. Deze vaccins werken door antilichamen te genereren die gericht zijn op de capsule en veel van deze studies gebruiken de OPKA om de opsono-fagocytische functies van de antilichamen13,14,<sup class...

Herkenning, therapietrouw, internalisering en afbraak van een ziekteverwekker zijn integraal onderdeel van fagocytose1, een saillant pad in de gastheer aangeboren immuunrespons die voor het eerst werd beschreven door Ilya Metchnikoff in 18832,3. Fagocytische leukocyten, evenals andere cellen van het immuunsysteem, zijn zeer discriminerend in hun selectie van doelen; zij zijn in staat onderscheid te maken tussen "infectieuze niet-zelf" en "niet-infectieuze zelf" door middel van pathogene geassocieerde moleculaire patronen door hun repertoire van patroonherkenningsreceptoren (PRR's)4,5. Gastheerherkenning van een ziekteverwekker kan ook optreden met de binding van gastheeropsonines, zoals complement en antilichamen6. Dit proces, opsonisatie genaamd, bedekt de ziekteverwekker met deze moleculen en verbetert de internalisatie bij binding aan opsonische receptoren (bijv. complement- en Fc-receptoren) op fagocytische cellen6. Om een ziekteverwekker zich aan een fagocyten te laten hechten, is collectieve binding van meerdere receptoren met hun cognate liganden noodzakelijk. Alleen dan kan aanhankelijkheid cascades in de hostcel activeren en ondersteunen om internalisatie te starten6.
Vanwege het belang van fagocytose bij het opruimen van pathogenen en het voorkomen van infectie, hebben extracellulaire pathogenen tal van manieren ontwikkeld om dit proces te ondermijnen om hun overleving te verlengen. Een strategie van belang is de productie van een anionische polymere (bijv. polysacharide of polyaminozuur) capsule die anti-fagocytisch is vanwege zijn lading, slecht immunogeen is en moleculen op de bacteriële envelop afschermt van PRR 's6,7. Pathogenen zoals Cryptococcus neoformans en Streptococcus pneumoniae hebben capsules die zijn samengesteld uit saccharidepolymeren, terwijl Staphylococcus epidermidis en sommige Bacillus-soorten poly-ɣ-glutaminezuur (PGGA)7,8produceren . Maar andere ziekteverwekkers produceren capsules die lijken op het niet-infectieuze zelf. Streptococcus pyogenes en een pathogene stam van B. cereus hebben bijvoorbeeld een hyaluronzuurcapsule die niet alleen anti-fagocytisch is, maar die ook niet als vreemd kan worden herkend door het immuunsysteem9,10.
Vervoeging van capsule naar dragereiwitten zet ze om van arme, T-onafhankelijke antigenen in zeer immunogene T-afhankelijke antigenen die hoge serumanticapsuleantilichaamtiters kunnen induceren11,12. Deze strategie wordt gebruikt voor gelicentieerde vaccins tegen S. pneumoniae, Haemophilus influenzaeen Neisseria meningitides11. De opsonische activiteiten van anticapsuleantilichamen zijn gewoonlijk geëvalueerd door opsono-fagocytische moordtests (OPKA)13,14,15,16. Deze tests testen of functionele antilichamen fagocytose en doden kunnen veroorzaken14. Het gebruik van OPKA met infectieuze pathogenen, zoals Tier 1 Biological Select Agents and Toxins (BSAT), waaronder B. anthracis17,is echter gevaarlijk en brengt veiligheidsrisico's met zich mee; deze tests vereisen een uitgebreide behandeling van een selecte agent. Het hanteren van bepaalde agenten kan alleen worden uitgevoerd in beperkte bioveiligheidsniveau 3 (BSL-3) laboratoria; het werk in deze gebieden vereist langdurige operationele procedures vanwege de vele veiligheidsmaatregelen die moeten worden gevolgd. BSL-3 laboratoria zijn ook meestal niet uitgerust met de gespecialiseerde apparatuur die wordt gebruikt voor OPKA-werk, zoals microscopen en cytometers. Zo ontwikkelden we een alternatieve test op basis van het gebruik van geïnactiveerde bacteriën18,19. We noemen dit een opsono-adherence assay (OAA) die niet afhankelijk is van internalisatie en doden als testoutputs; in plaats daarvan wordt therapietrouw van geïnactiveerde geïnactiveerde pathogenen gebruikt als een index van fagocytose. Mechanistisch gezien is OAA een geschikt substituut omdat therapietrouw a priori optreedt en nauw verweven is met internalisatie en intracellulair doden. Vanuit bioveiligheidsperspectief heeft OAA de voorkeur omdat het een minimale behandeling van een infectieus agens vereist, experimenteel van kortere duur is dan OPKA en kan worden uitgevoerd in BSL-2-laboratoria nadat een voorraad van de geïnactiveerde ziekteverwekker is geproduceerd en overgedragen.
We demonstreren het gebruik van OAA om de opsonische functie van anticapsuleantilichamen te onderzoeken die worden aangetroffen in sera van niet-menselijke primaten (NHP's) gevaccineerd met een capsuleconjugaat [d.w.z. PGGA van B. anthracis geconjugeerd aan het buitenste membraaneiwitcomplex (OMPC) van Neisseria meningitides]20. Serum opsonized bacillen werden geïncubeerd met een aanhankelijkheid muis macrofaag cellijn, RAW 264.7. Na fixatie werden de celmonolaag en aanhankelijke bacillen in beeld gesteld door fluorescentiemicroscopie. Bacteriële hechting nam toe toen de bacillen werden geïncubeerd met serum van NHP's gevaccineerd met de capsuleconjugaat in vergelijking met controleserum20. Therapietrouw gecorreleerd met survival of the anthrax challenge20,21. Zo kenmerkte het gebruik van OAA de functie van anticapsuleantilichamen en vergemakkelijkte het testen van onze vaccinkandidaat aanzienlijk.

opsono-adherence assay to evaluate functional antibodies in vaccine development against bacillus anthracis and other encapsulated pathogens

De opsono-therapietrouwtest is een functionele test die de aanhechting van bacteriële pathogenen aan professionele fagocyten opsomt. Omdat therapietrouw noodzakelijk is voor fagocytose en doden, is de test een alternatieve methode voor opsono-fagocytische moordtests. Een voordeel van de opsono-adherence-test is de optie om geïnactiveerde pathogenen en zoogdiercellijnen te gebruiken, waardoor standaardisatie in meerdere experimenten mogelijk is. Het gebruik van een geïnactiveerde ziekteverwekker in de test vergemakkelijkt ook het werken met bioveiligheidsniveau 3 infectieuze agentia en andere virulente pathogenen. In ons werk werd de opsono-therapietrouwtest gebruikt om het functionele vermogen van antilichamen te beoordelen, van sera van dieren die zijn geïmmuniseerd met een vaccin op basis van miltvuurcapsules, om hechting van vaste Bacillus-antracis aan een muismacrofaagcellijn, RAW 264.7, te induceren. Geautomatiseerde fluorescentiemicroscopie werd gebruikt om beelden vast te leggen van bacillen die zich aan macrofagen hechten. Verhoogde therapietrouw was gecorreleerd met de aanwezigheid van anti-capsule antilichamen in het serum. Niet-menselijke primaten die hoge serum anti-capsule antilichaamconcentraties vertoonden, werden beschermd tegen miltvuuruitdaging. Zo kan de opsono-therapietrouwtest worden gebruikt om de biologische functies van antigeenspecifieke antilichamen in sera op te helderen, om de werkzaamheid van vaccinkandidaten en andere therapeutische middelen te evalueren en om te dienen als een mogelijke correlaat van immuniteit.

Deze sectie toont representatieve micrografen verzameld tijdens een OAA-experiment, samen met resultaten waaruit blijkt dat de OAA kan worden gebruikt om de biologische functie van antilichamen te onderzoeken. Hier werd de test met succes gebruikt om de werkzaamheid van een miltvuurvaccinkandidaat te evalueren.
