Erkennung, Adhärenz, Internalisierung und Abbau eines Erregers sind integraler Bestandteil der Phagozytose1, ein markanter Weg im Wirt angeborene Immunantwort zuerst beschrieben von Ilya Metchnikoff in 18832,3. Phagozytäre Leukozyten, sowie andere Zellen des Immunsystems, sind sehr diskriminierend in ihrer Auswahl von Zielen; sie sind in der Lage, zwischen "infektiösem Nicht-Selbst" und "nicht-infektiöses Selbst" durch pathogene assoziierte molekulare Muster durch ihr Repertoire an Mustererkennungsrezeptoren (PRRs)4,5zu unterscheiden. Die Wirterkennung eines Erregers kann auch mit der Bindung von Wirtsopsoninen, wie Komplement undAntikörpern 6auftreten. Dieser Prozess, der als Opsonisierung bezeichnet wird, beschichtet den Erreger mit diesen Molekülen und verbessert die Internalisierung bei Bindung an Opsonic-Rezeptoren (z. B. Komplement- und Fc-Rezeptoren) auf phagozytären Zellen6. Damit ein Erreger an einer Phagozyten haften kann, ist eine kollektive Bindung mehrerer Rezeptoren mit ihren Cognate-Ninentinanden erforderlich. Nur dann kann die Adhärenz Signalkaskaden innerhalb der Hostzelle auslösen und aufrechterhalten, um die Internalisierung zu initiieren6.
Aufgrund der Bedeutung der Phagozytose für die Clearance von Krankheitserregern und die Prävention von Infektionen haben extrazelluläre Krankheitserreger zahlreiche Wege entwickelt, diesen Prozess zu untergraben, um ihr Überleben zu verlängern. Eine Strategie von Bedeutung ist die Herstellung einer anionischen Polymerkapsel (z. B. Polysaccharid oder Polyaminosäure), die aufgrund ihrer Ladung antiphagozytisch ist, schlecht immunogen ist und Moleküle auf der bakteriellen Hülle aus PRRs6,7abschirmt. Pathogene wie Cryptococcus neoformans und Streptococcus pneumoniae haben Kapseln aus Saccharidpolymeren, während Staphylococcus epidermidis und einige Bacillus-Arten Poly-ɣ-Glutaminsäure (PGGA)7,8produzieren. Doch andere Krankheitserreger produzieren Kapseln, die dem nicht-infektiösen Selbst ähneln. Zum Beispiel haben Streptococcus pyogenes und ein pathogener Stamm von B. cereus eine Hyaluronsäurekapsel, die nicht nur antiphagozytisch ist, sondern auch vom Immunsystem nicht als fremd erkannt werden kann9,10.
Konjugation von Kapsel zu Trägerproteinen wandelt sie von armen, T-unabhängigen Antigenen in hochimmunogene T-abhängige Antigene um, die hohe Serum-Antikapsel-Antikörper-Titers induzieren können11,12. Diese Strategie wird für lizenzierte Impfstoffe gegen S. pneumoniae, Haemophilus influenzaeund Neisseria meningitides11eingesetzt. Die Opsonic-Aktivitäten von Anti-Kapsel-Antikörpern wurden häufig durch opsono-phagozytische Tötungstests (OPKA)13,14,15,16ausgewertet. Diese Assays testen, ob funktionelle Antikörper Phagozytose auslösen und14töten können. Die Verwendung von OPKA mit infektiösen Krankheitserregern wie Tier 1 Biological Select Agents and Toxins (BSAT), einschließlich B. anthracis17, ist jedoch gefährlich und birgt Sicherheitsrisiken; diese Assays erfordern eine umfassende Handhabung eines ausgewählten Agenten. BSL-3-Labore sind in der Regel auch nicht mit den speziellen Geräten für OPKA-Arbeiten wie Mikroskope und Zytometer ausgestattet. So entwickelten wir einen alternativen Assay auf Basis der Verwendung von inaktivierten Bakterien18,19. Wir bezeichnen dies als einen Opsono-Adherence-Assay (OAA), der nicht von Internalisierung und Tötung als Assay-Output sabhängt; Stattdessen wird die Adhärenz von opsonisierten inaktivierten Krankheitserregern als Index der Phagozytose verwendet. Mechanistisch ist OAA ein geeigneter Ersatz, da die Adhärenz a priori auftritt und eng mit Internalisierung und intrazellulärer Tötung verflochten ist. Aus Sicht der Biosicherheit wird OAA bevorzugt, da es einen minimalen Umgang mit einem Infektier erfordert, experimentell kürzer ist als OPKA und in BSL-2-Labors durchgeführt werden kann, nachdem ein Vorrat des inaktivierten Erregers produziert und übertragen wurde.
Wir zeigen die Verwendung von OAA zur Untersuchung der Opsonic-Funktion von Anti-Kapsel-Antikörpern, die in Seren von nichtmenschlichen Primaten (NHPs) gefunden wurden, die mit einem Kapselkonjugat geimpft sind [d.h. PGGA von B. anthracis konjugiert mit dem äußeren Membranproteinkomplex (OMPC) von Neisseria Meningitiden]20. Serum-opsonisierte Bazillen wurden mit einer anhaftenden Mausmakrophagenzelllinie, RAW 264.7, inkubiert. Nach der Fixierung wurden die Zellmonolayer und die anhaftenden Bazillen durch Fluoreszenzmikroskopie abgebildet. Die bakterielle Adhärenz nahm zu, wenn die Bazillen mit Serum von NHPs inkubiert wurden, die mit dem Kapselkonjugat geimpft wurden, verglichen mit Kontrollserum20. Die Einhaltung korrelierte mit dem Überleben der Milzbrand-Herausforderung20,21. So charakterisierte die Verwendung von OAA die Funktion von Anti-Kapsel-Antikörpern und erleichterte die Prüfung unseres Impfstoffkandidaten erheblich.

opsono-adherence assay to evaluate functional antibodies in vaccine development against bacillus anthracis and other encapsulated pathogens

Der Opsono-Adhärenz-Assay ist ein funktioneller Test, der die Bindung bakterieller Krankheitserreger an professionelle Phagozyten aufzählt. Da die Einhaltung der Phagozytose und Tötung erforderlich ist, ist der Assay eine alternative Methode zu opsono-phagozytischen Tötungstests. Ein Vorteil des Opsono-Adhärenz-Assays ist die Möglichkeit, inaktivierte Krankheitserreger und Säugetierzelllinien zu verwenden, was eine Standardisierung über mehrere Experimente hinweg ermöglicht. Die Verwendung eines inaktivierten Erregers im Test erleichtert auch die Arbeit mit Infektionserregern der Biosicherheitsstufe 3 und anderen virulenten Krankheitserregern. In unserer Arbeit wurde der Opsono-Adhärenz-Test verwendet, um die funktionelle Fähigkeit von Antikörpern von Tieren zu bewerten, die mit einem Impfstoff auf Milzbrandkapsel-Basiert immunisiert wurden, um die Einhaltung der festen Bacillus-Anthrakis an einer Maus-Makrophagenzelllinie, RAW 264.7, zu induzieren. Automatisierte Fluoreszenzmikroskopie wurde verwendet, um Bilder von Bazillen zu erfassen, die an Makrophagen haften. Erhöhte Adhärenz wurde mit dem Vorhandensein von Anti-Kapsel-Antikörpern im Serum korreliert. Nichtmenschliche Primaten, die hohe Serum-Antikapsel-Antikörperkonzentrationen aufwiesen, wurden vor Milzbrand-Herausforderungen geschützt. So kann der Opsono-Adhärenz-Test verwendet werden, um die biologischen Funktionen von antigenspezifischen Antikörpern in seren zu klären, die Wirksamkeit von Impfstoffkandidaten und anderen Therapeutika zu bewerten und als mögliches Korrelat der Immunität zu dienen.

Dieser Abschnitt zeigt repräsentative Mikrographen, die während eines OAA-Experiments gesammelt wurden, zusammen mit Ergebnissen, die zeigen, dass die OAA zur Untersuchung der biologischen Funktion von Antikörpern verwendet werden kann.
