Erkännande, vidhäftning, internalisering och nedbrytning av en patogen är integrerade i phagocytos1, en framträdande väg i värd medfödda immunsvar som först beskrivs av Ilya Metchnikoff 18832,3. Fagocytiska leukocyter, liksom andra celler i immunsystemet, är mycket diskriminerande i sitt urval av mål. de kan skilja mellan "infektiöst icke-jag" och "icke-infektiöst jag" genom patogen associerade molekylära mönster genom sin repertoar av mönsterigenkänningsreceptorer (PRR)4,5. Värd erkännande av en patogen kan också uppstå med bindning av värd opsoniner, såsom komplement och antikroppar6. Denna process, kallad opsonisering, täcker patogenen med dessa molekyler, vilket förbättrar internaliseringen vid bindning till opsoniska receptorer (t.ex. komplement och Fc-receptorer) på fagocytiska celler6. För att en patogen ska hålla fast vid en faagocyt är kollektiv bindning av flera receptorer med deras cognate ligands nödvändig. Först då kan vidhäftningsutlösare och upprätthålla signalkaskader inuti värdcellen för att initiera internalisering6.
På grund av vikten av faagocytos vid clearance av patogener och förebyggande av infektion, har extracellulära patogener utvecklat många sätt att undergräva denna process för att förlänga deras överlevnad. En viktig strategi är produktionen av en anjonpolymerisk (t.ex. polysackarid eller polyaminosyra) kapsel som är anti-faagocytisk på grund av sin laddning, är dåligt immunogen och skyddar molekyler på bakteriehöljet från PRR6,7. Patogener som Cryptococcus neoformans och Streptococcus pneumoniae har kapslar bestående av sackaridpolymerer, medan Staphylococcus epidermidis och vissa Bacillus-arter producerar poly-ɣ-glutaminsyra (PGGA)7,8. Men andra patogener producerar kapslar som liknar det icke-infektiösa jaget. Till exempel har Streptococcus pyogener och en patogen stam av B. cereus en hyaluronsyrakapsel som inte bara är anti-fagocytisk utan som inte heller kan erkännas som främmande av immunsystemet9,10.
Konjugation av kapsel till bärarproteiner omvandlar dem från dåliga, T-oberoende antigener till mycket immunogena T-beroende antigener som kan inducera höga serum antikapselantikroppstitrar11,12. Denna strategi används för licensierade vacciner mot S. pneumoniae, Haemophilus influenzae, och Neisseria meningitides11. De opsoniska aktiviteterna hos antikapselantikroppar har ofta utvärderats med opsono-fagocytiska avlivningsanalyser (OPKA)13,14,15,16. Dessa analyser testar om funktionella antikroppar kan utlösa faagocytos och döda14. Användningen av OPKA med infektiösa patogener, såsom biologiska urvalsmedel och toxiner i tier 1 (BSAT), inklusive B. anthracis17,är dock farlig och utgör säkerhetsrisker. Dessa analyser kräver omfattande hantering av en utvald agent. Hantering av utvalda agenser kan endast ske i laboratorier med begränsad biosäkerhetsnivå 3 (BSL-3). arbete på dessa områden kräver utdragna driftsrutiner på grund av de många säkerhets- och säkerhetsåtgärder som måste följas. BSL-3-laboratorier är vanligtvis inte utrustade med den specialiserade utrustning som används för OPKA-arbete, såsom mikroskop och cytometrar. Således utvecklade vi en alternativ analys baserad på användningen av inaktiverade bakterier18,19. Vi hänvisar till detta som en opsono-adherence assay (OAA) som inte är beroende av internalisering och dödande analysutgångar; I stället används vidhäftning av opsoniserade inaktiverade patogener som ett index för faagocytos. Mekanistiskt är OAA en lämplig ersättning eftersom vidhäftning sker a priori och är intimt sammanflätad med internalisering och intracellulärt dödande. Ur ett biosäkerhetsperspektiv föredras OAA eftersom det kräver minimal hantering av ett smittämne, är experimentellt kortare än OPKA och kan utföras i BSL-2-laboratorier efter att ett lager av den inaktiverade patogenen har producerats och överförts.
Vi visar användningen av OAA för att undersöka den opsonic funktionen av anti-kapsel antikroppar som finns i serum av icke-mänskliga primater (NHPs) vaccineras med en kapsel konjugat [dvs PGGA från B. anthracis konjugeras till yttre membran protein komplexa (OMPC) av Neisseria meningitides]20. Serum opsonized bacilli inkuberades med en vidhäftande mus makrofag cellinje, RAW 264,7. Efter fixering, cell monolayer och vidhäftande bacilli var av fluorescens mikroskopi. Bakteriell vidhäftning ökade när bacilli inkuberades med serum från NHPs vaccinerade med kapselkonjugat jämfört med kontrollserum20. Vidhäftning korrelerad med överlevnad av myraxutmaningen20,21. Således karakteriserade användningen av OAA funktionen av antikroppar mot kapseln och underlättade kraftigt testning av vår vaccinkandidat.

opsono-adherence assay to evaluate functional antibodies in vaccine development against bacillus anthracis and other encapsulated pathogens

Opsono-adherence-analysen är en funktionell analys som räknar upp fastsättningen av bakteriella patogener till professionella fagocyter. Eftersom vidhäftning är nödvändiga för fagocytos och dödande, är analysen en alternativ metod till opsono-faagocytiska dödande analyser. En fördel med opsono-adherence-analysen är möjligheten att använda inaktiverade patogener och däggdjurscelllinjer, vilket möjliggör standardisering över flera experiment. Användningen av en inaktiverad patogen i analysen underlättar också arbetet med biosäkerhetsnivå 3 smittämnen och andra virulenta patogener. I vårt arbete användes opsono-adherence-analysen för att bedöma antikropparnas funktionella förmåga, från serum hos djur immuniserade med ett mjältbrandskapselbaserat vaccin, för att inducera vidhäftning av fast Bacillus anthracis till en musmakrofagcelllinje, RAW 264.7. Automatiserad fluorescensmikroskopi användes för att fånga bilder av bacilli som följer makrofager. Ökad vidhäftning korrelerades med förekomsten av antikroppar mot kapseln i serumet. Icke-mänskliga primater som uppvisade höga serum anti-kapsel antikropp koncentrationer skyddades från anthrax utmaning. Således kan opsono-adherence-analysen användas för att belysa de biologiska funktionerna hos antigenspecifika antikroppar i serum, för att utvärdera effekten av vaccinkandidater och andra terapier och för att fungera som en möjlig korrelering av immunitet.

Detta avsnitt visar representativa mikrografer som samlats in under ett OAA-experiment tillsammans med resultat som visar att OAA kan användas för att undersöka antikroppars biologiska funktion. Här användes analysen framgångsrikt för att utvärdera effekten av en anthraxvaccinkandidat. Det är viktigt att verifiera tillståndet för inkapsling på bacilli så lite eller ingen inkapsling orsakar dem att hålla sig till värdceller, vilket ger en hög bakgrund. Figur 1 är en bild ...
