Bir patojenin tanınması, yapışması, içselleştirilmesi ve bozulması fagositozun ayrılmaz bir parçasıdır 1 , ilk olarakIlyaMetchnikoff tarafından 1883'te tanımlanan konakçı doğuştan gelen immün yanıtta belirgin bir yol2,3. Fagositik lökositler ve bağışıklık sisteminin diğer hücreleri, hedef seçimlerinde son derece ayrımcıdır; patojen ilişkili moleküler desenler aracılığıyla "enfeksiyöz olmayan benlik" ile "enfeksiyöz olmayan benlik" arasında, örüntü tanıma reseptörleri (PRR) repertuarları (PRR)4,5ile ayırt edebiliyorlar. Bir patojenin konak tanıması, tamamlayıcı ve antikorlar gibi konak opsoninlerin bağlanmasıyla da ortaya çıkabilir6. Opsonizasyon adı verilen bu işlem, patojeni bu moleküllerle kaplar, fagositik hücreler üzerindeki opsonik reseptörlere (örneğin, kompleman ve Fc reseptörleri) bağlanması üzerine içselleştirmeyi artırır6. Patojenlerin temizlenmesinde ve enfeksiyonun önlenmesinde fagositozun önemi nedeniyle, hücre dışı patojenler hayatta kalmalarını uzatmak için bu süreci altüst etmenin birçok yolunu geliştirmiştir. opsono-adherence assay to evaluate functional antibodies in vaccine development against bacillus anthracis and other encapsulated pathogens

Opsono-yapışma tahlili, bakteriyel patojenlerin profesyonel fagositlere bağlanmasını numaralayan işlevsel bir testtir. Fagositoz ve öldürme için bağlılık gerekli olduğundan, tahlil opsono-fagositik öldürme testlerine alternatif bir yöntemdir. Opsono-yapışma testinin bir avantajı, birden fazla deneyde standardizasyona izin veren inaktive patojenleri ve memeli hücre hatlarını kullanma seçeneğidir. Testte inaktive bir patojenin kullanılması, biyogüvenlik seviyesi 3 enfeksiyöz ajanlar ve diğer virülan patojenlerle çalışmayı da kolaylaştırır. Çalışmamızda, opsono-yapışmış test, şarbon kapsülü bazlı bir aşı ile aşılanmış hayvanların serasından, sabit Bacillus anthracis'in fare makrofaj hücresi hattı RAW 264.7'ye yapışmasını teşvik etmek için antikorların fonksiyonel yeteneğini değerlendirmek için kullanılmıştır. Makrofajlara bağlı basil görüntülerini yakalamak için otomatik floresan mikroskopisi kullanıldı. Serumda anti-kapsül antikorlarının varlığı ile artan yapışlık ilişkiliydi. Yüksek serum anti-kapsül antikor konsantrasyonları sergileyen insan olmayan primatlar şarbon zorluğundan korundu. Bu nedenle, opsono-yapışıklık testi, seradaki antijen spesifik antikorların biyolojik işlevlerini aydınlatmak, aşı adaylarının ve diğer terapötiklerin etkinliğini değerlendirmek ve bağışıklığın olası bir korelasyon olarak hizmet etmek için kullanılabilir.
