LNT-netwerken zijn een nuttig hulpmiddel voor in vitro studies naar membraaneigenschappen en het transport van biomoleculen zoals transmembraaneiwitten. Bovendien maakt het gebruik van complexe lipideformuleringen om LNT-netwerken te fabriceren meer biologisch relevante studies mogelijk. De fabricage van lipide nanobuis (LNT) netwerken is van toenemend belang voor in vitro onderzoek van niet-evenwicht lipidestructuren 1,2,3. Cellen gebruiken lipide tubuli voor het diffusieve transport van eiwitten4 en nucleïnezuren5 en cel-naar-cel communicatie 6,7. Het endoplasmatisch reticulum en het Golgi-apparaat zijn bijzonder interessant, omdat deze membraangebonden organellen de primaire locaties zijn voor lipide- en eiwitsynthese en transport van deze integrale biomoleculen in het cytoplasma van een cel 8,9. De membranen van deze organellen bestaan uit meerdere lipidesoorten, waaronder sfingolipiden, cholesterol en fosfolipiden10 die uiteindelijk helpen bij het definiëren van hun functionaliteit. Dus, om deze organellen nauwkeuriger te repliceren en te bestuderen, moeten in vitro LNT's worden vervaardigd uit blaasjes met steeds complexere lipideformuleringen11.
Gigantische unilamellaire blaasjes (GUVs) worden alomtegenwoordig gebruikt voor het bestuderen van lipidemembraangedrag omdat ze betrouwbaar kunnen worden gesynthetiseerd met complexe formuleringen die cholesterol, fosfatidylcholine (PC), fosfatidylethanolamine (PE), fosfatidylserine (PS) en fosfatidylinositol (PI) bevatten 12,13. Hier wordt een methode beschreven om LNT's te fabriceren uit GUVs met verschillende lipideformuleringen met behulp van de gliding motility assay (GMA), waarbij LNT's worden geëxtrudeerd op basis van het werk dat wordt uitgevoerd door kinesinemotoren en microtubule-filamenten die op GUVs werken. In dit systeem stuwen kinesinemotoreiwitten geadsorbeerd aan een oppervlak gebiotinyleerde microtubuli aan, waardoor chemische energie van de hydrolyse van ATP wordt omgezet in nuttig werk (met name de extrusie van LNT's uit gebiotinyleerde blaasjes)11. Het resulterende LNT-netwerk biedt een modelplatform om effecten van de verschillen in lipidefasen op veranderingen in LNT-morfologie te bestuderen.
In het kort worden kinesinemotoreiwitten in een stroomkamer ingebracht in een oplossing die caseïne bevat, waardoor de motoren op het glazen oppervlak van de kamer kunnen worden geplaatst. Vervolgens stromen gebiotinyleerde microtubuli in een oplossing die ATP bevat door de kamer en mogen ze binden aan de kinesinemotoren en beginnen met motiliteit. Een streptavidin-oplossing wordt vervolgens in de kamer ingebracht en mag niet-covalent aan de microtubuli binden. Ten slotte worden GUVs met een gebiotinyleerd lipide in de kamer gebracht en binden ze aan de gestreepte microtubuli en extruderen ze vervolgens LNT's om grootschalige netwerken te vormen in de loop van 15-30 minuten. Deze methode produceert grote, vertakte LNT-netwerken met behulp van standaard laboratoriumapparatuur en reagentia tegen lage kosten11.

Lipide nanobuisjes (LNT) netwerken vertegenwoordigen een in vitro modelsysteem voor het bestuderen van moleculair transport en lipide biofysica met relevantie voor de alomtegenwoordige lipide tubuli in eukaryote cellen. In vivo LNT's zijn echter zeer niet-evenwichtsstructuren die chemische energie en moleculaire motoren vereisen om te worden geassembleerd, onderhouden en gereorganiseerd. Bovendien is de samenstelling van in vivo LNT's complex, bestaande uit meerdere verschillende lipidesoorten. Typische methoden om LNT's te extruderen zijn zowel tijd- als arbeidsintensief, en ze vereisen optische pincetten, microbeads en micropipettes om nanobuisjes met geweld uit gigantische lipideblaasjes te trekken. Hier wordt een protocol gepresenteerd voor de gliding motility assay (GMA), waarbij grootschalige LNT-netwerken snel worden gegenereerd uit gigantische unilamellaire blaasjes (GUVs) met behulp van kinesine-aangedreven microtubule motiliteit. Met behulp van deze methode worden LNT-netwerken gevormd uit een breed scala aan lipideformuleringen die de complexiteit van biologische LNT's nabootsen, waardoor ze steeds nuttiger worden voor in vitro studies van lipidebiofysica en membraangeassocieerd transport. Bovendien is deze methode in staat om in korte tijd (<30 min) op betrouwbare wijze LNT-netwerken te produceren met behulp van veelgebruikte laboratoriumapparatuur. LNT-netwerkkenmerken zoals lengte, breedte en lipidenverdeling zijn ook instelbaar door de lipidesamenstelling van de GUVs die worden gebruikt voor het fabriceren van de netwerken te veranderen.

LNT-netwerken (figuur 4) werden vervaardigd met behulp van het beschreven protocol, dat het werk gebruikt dat wordt uitgevoerd door kinesinetransport van microtubuli om LNT's uit GUVs te extruderen. Dit protocol maakt het mogelijk om eenvoudig grootschalige lipide nanobuisnetwerken te genereren waarin de individuele nanobuisjes fasescheidingsgedrag vertonen dat vergelijkbaar is met dat van het moederblaasje. Het belangrijkste voordeel van deze aanpak is dat we het werk van de moleculaire motoren coöpteren om de lipide nanobuisjes zelf op een zeer parallelle manier en zonder menselijke tussenkomst te assembleren. Om een glijdende motiliteitstest op te zetten, bevestigt u eerst drie sets dubbelzijdige tapestrips op een glazen schuif die vijf millimeter uit elkaar ligt.Druk voorzichtig op een afdekplaat op de tape en voeg 30 microliter kinesineoplossing van één micromolair toe aan de voorbereide stroomcel. Voeg na vijf minuten 30 microliter van een 10-microgram / milliliter microtubulioplossing toe aan de cel en druk zachtjes op een laboratoriumdoekje tegen het andere uiteinde van het stroomkanaal om de uitwisseling van de oplossing te vergemakkelijken. Was de cel na nog eens vijf minuten incubatie één tot drie keer met 50 microliter motiliteitsoplossing op kamertemperatuur per wasbeurt voordat u 30 microliter streptavidinoplossing van 10 microgram/milliliter aan de stroomcel toevoegt.Voeg na 10 minuten 30 microliter van een 10X GUV-oplossing toe voor een incubatie van 30 minuten bij kamertemperatuur. Voeg vervolgens twee microliter van een AMP-PNP-oplossing van 100 millimolair toe en sluit de kamer met kit. Breng de stroomkamer over naar een omgekeerde microscoop voor beeldvorming en selecteer de juiste filterset.Gebruik een olieobjectief van 100X om eerst het oppervlak van de afdekslip scherp te stellen voordat u een afbeelding van een interessant netwerk krijgt. Open vervolgens de afbeelding in ImageJ en klik op Afbeelding, Kleur en Samenstelling om de rode en groene kanalen te bedekken om een samengestelde afbeelding te maken. Open na de beeldvorming de gewenste afbeelding en klik op Analyseren en schaal instellen om de schaal voor de microscoop te kalibreren.Vul de pixels in op de conversiefactor van de micrometer en klik op OK. Gebruik het meerpuntslijngereedschap om de nanobuisjes te traceren vanaf de bovenliggende GUV en houd Control en M ingedrukt om de lengte te meten. De beeldverwerkingstool slaat elke nieuwe meting op in het resultatenvenster. Wanneer alle buizen zijn gemeten, selecteert u Afbeelding, Aanpassen en Drempel en klikt u op Toepassen om de drempel toe te passen.Teken vervolgens een rechthoek van bekende lengte over de gewenste buis. Als u de geïntegreerde dichtheid van het gebied wilt meten, klikt u op Analyseren en meten Om de lipideverdeling in de vloeibare nanobuisknooppunten te bepalen, gebruikt u het lijngereedschap om een lijn over het knooppunt van belang te tekenen en de knooppuntintensiteit in zowel het groene als het rode kanaal te meten. In deze representatieve analyse werden vloeibare nanobuisnetwerken gefabriceerd, waarvan is aangetoond dat ze vloeistof-ongeordende en geordende fasen genereren, evenals fase-gescheiden blaasjes.Blaasjes gesynthetiseerd met 45% verzadigd lipide en 55% onverzadigde lipide resulteert bijvoorbeeld in blaasjes die zich scheiden in naast elkaar bestaande vloeistof-ongeordende en vaste fasen. Als cholesterol echter wordt opgenomen, kan coëxistentie tussen vloeistof en vloeistof worden waargenomen, waardoor GUVs ontstaan die zich scheiden in naast elkaar bestaande vloeistofgeordende en vloeistof-ongeordende fasen. Bovendien kunnen knooppunten worden waargenomen binnen de fasegescheiden mengsels.Het belangrijkste om te onthouden is om de juiste belichtingstijd en ND-filterset te kiezen om de lipide nanobuisjes te kunnen visualiseren zonder de fluoroforen te fotobleken. Door de samenstelling van het fasegedrag van de lipide nanobuisjes aan te passen, kunnen we een minimalistisch modelsysteem gebruiken om de biofysica van de tunnelende nanobuisjes die cellen verbinden te bestuderen.
