인간의 피부에서 유래된 인간 1차 각질세포는 표피1,2,3,4의생물학을 연구하기 위해 세포 모델로 자주 사용된다. 표피의 계층화는 2D 침수 단층 패션 또는 3D 공기 리프트 organotypic 모델2,3,5,6,7에서각질 세포 분화에 의해 모델링 될 수 있습니다. 3D 모델은 표피 구조와 기능을 평가하는 것이 점점 더 중요해지고 있지만, 2D 분화 모델은 여전히 널리 사용되고 있습니다.

이러한 방법 중 하나는 배양 배지8에서성장 인자의 각질 세포 접촉 억제 및 고갈을 포함한다. 세포가 전체 밀도에 도달할 때 각질 세포가 자발적으로 분화할 수 있음을 보여주었습니다.  배양 배지의 성장 요인을 제외하면 차별화를 더욱 향상시킬 수 있습니다. 접촉 억제 및 고갈성장 인자를 결합하는 방법은 여러 표피 마커3을사용할 때 정상 계층화 표피와 유사한 유전자 발현 패턴과 분화된 각질 세포체를 생성하는 것으로 나타났으며, 이 모델은 정상 각질 세포 분화를 연구하는 데 적합하다는 것을 시사한다. 최근에는 본 모델을 이용한 각질 세포 분화의 2개의 포괄적인 유전자 발현 분석이9,10으로보고되었다. 연구원은 분자 수준에서 이 모형을 확인하고 정상및 병은 각질 세포 분화를 공부하는 것을 이용될 수 있다는 것을 보여주었습니다.
이 프로토콜은 RNA-seq를 사용하여 분화 된 세포의 체외 분화 방법 및 분자 분석에 대한 절차를 설명합니다. 또한 분화일 0(증식 단계), 2일차, 4일, 7일(각각 조기, 중, 후기 분화)에 대한 세포의 전사의 특성화를 보여 준다. 분화 된 각질 세포가 표피 계층화 중에 세포를 크게 닮은 유전자 발현 패턴을 표시하는 것으로 나타났습니다. 이 방법이 피부 병리학을 연구하는 데 사용될 수 있는지 여부를 조사하기 위해 동일한 실험 및 분석 파이프 라인을 적용하여 방황, 이장 물 분비증 및 갈라진 입술 / 미각 증후군 (EEC) 증후군11,12를가진 환자로부터 파생된 전사 인자 p63의 돌연변이를 운반하는 각질 세포를 조사했습니다. 이 프로토콜은 각질 세포의 체외 분화뿐만 아니라 RNA-seq의 후속 생물 정보 분석에 초점을 맞추고 있습니다.
