生理学的条件下での豚の心臓のスライスと培養

このプロトコルは、急性心臓毒性検査に使用できる生理学的条件下で6日間の心臓組織切片をスライスして培養し、心不全療法の有効性を検査するためのプロトコルです。この培養システムは、臨床前と臨床検査の結果のギャップを埋める急性心毒性試験の現場モデルにおいて、強力な予測ヒトになる可能性を秘めています。手順をデモンストレーションするのが、当研究室のラボマネージャーである青輝Quです。豚の心臓組織のスライスを得るために、まずビブラートメの組織浴冷却ジャケットに氷を加え、タイローデ溶液を浴に加える。1リットルのプラスチック瓶を使用して、組織バス冷却ジャケットから溶けた氷の流出を収集し、バイオセーフティキャビネットの新鮮な冷たい心肢溶液の1リットルを含むトレイに豚の心臓を移します。引き込み可能な無菌メスを使用して心臓を解剖して左心室を分離し、長方形のカミソリブレードを使用して左心室を1〜2立方センチメートルのブロックに切断します。スライスするための1つの組織ブロックを脇に置き、残りの組織ブロック片を氷の上の冷たいタイロード溶液の50ミリリットルチューブに入れる。予約した組織ブロックを穏やかにマッサージし、ビブラートメの金属サンプルホルダーにティッシュ接着剤を1~2滴加えます。4%寒天ブロックの一部を約1平方センチメートルの表面積を接着剤に貼り付け、寒天にティッシュ接着剤を1~2滴加えます。寒天心心気道側に心臓ブロックを持ち込み、組織を可能な限りホルダーに対して平らにし、心臓ブロック付きの組織ホルダーをビブラートメのスライス浴に入れます。心臓心気道側が組織の接着剤の上を下に向いている寒天に心臓ブロックを貼り付け、組織が可能な限り平らであることを確認することが重要です。次に、酸素チューブをスライス浴に取り付け、タイロード溶液で満たされた金属トレイを金属トレイに置き、40マイクロメートルの細胞ストレーナーを金属トレイに置き、スライスされた心臓組織の切片を収集します。ビブラートメの操作ソフトウェアを使用して、ブレードが組織の上部に近いが、乳頭の筋肉の下に、液体が組織とブレードを覆っているように、ブレードとサンプルの高さを調整します。組織のスライスを開始する場所を調整する進入を選択します。スライスを押して速度を上げ、ノブを使用してブレードをティッシュの端に向かって動かします。スライスをもう一度押すと、プロセスが停止して先に進み、プロセスが無効になります。次に、示されているように切断パラメータを調整し、スライスを押して組織のスライスを開始します。ブレードが組織の端に達したが、それが標本ホルダーの端に当たる前に、スライスをもう一度押して停止し、戻って開始位置に戻ります。スライスが完全な長さに達し、良質であるように見える場合は、冷たいタイロード溶液で満たされたプラスチック製のパスツールピペットを使用して、鉗子とスプリングハサミを使用してお風呂から組織を穏やかに収集し、必要に応じて心臓からスライスを外します。採取した各スライスを酸素化されたタイローデ浴中の単一の細胞ストレーナーに入れ、ピペットの溶液を使用して細胞ストレーナー表面の組織を押します。その後、金属ワッシャーを組織の上部に置き、それを押さえます。酸素化されたタイローデ溶液で少なくとも1時間後、各スライスの外側をトリミングして不均一なエッジを取り除き、各スライスの端を金属ワイヤーが埋め込まれた6ミリメートル幅の滅菌ポリウレタンプリンタタイミングベルトに接着します。サポートされている心臓スライスを井戸あたり6ミリリットルの培養培地を含む6ウェルプレートに入れ、滅菌鉗子を使用して組織が井戸の中央にあることを確認します。プレートカバーがティッシュに触れないようにし、プレートカバーの湾曲した側がプレートの斜めの側と一致し、正方形の角が並ぶように、プレートを培養プレートに置くように注意してください。スライスを摂氏37度と5%の二酸化炭素インキュベーターに入れる。培養上清を6ミリリットルの新鮮な酸素化培養培地に1日3回交換します。カバーを細胞培養用電気刺激装置に接続し、刺激装置を調整して、10ボルトの電気と1.2ヘルツ周波数を心臓スライスに継続的に供給します。刺激を差し込んだ後、心臓のスライスが拍動し始めます。毎日、正午の培地交換時に、培養皿からの刺激プレートカバーを交換して、有毒な黒鉛粒子が培養培地に放出されるのを防ぐ。使用されたカバーが電気回路への水の損傷を防ぐために細胞培養電気刺激装置のためのケーブルに接続する白い泡プラグを挿入し、2X抗生物質/抗ミコティックで補われたオートクレーブ水の浴にプレートカバーを置きます。翌日、プレートカバーを70%エタノール浴で5〜15分間除染してから、カバーを2倍の抗生物質/抗ミコティックを3分間補充した新鮮なオートクレーブウォーターバスに移します。プレートカバーをすすった後、エタノールスプレーのきれいな糸くずのないワイプを使用して、プラスチック部品から残った水を取り除き、白い泡プラグを取り除きます。カバーは、培養プレートに戻す準備が整いました。この新しいバイオミメティック培養のセットアップを実証した方法で、MTTアッセイによって評価された豚の心臓スライスの生存率を6日間維持した。最初の6日間で、新鮮な心臓スライスに対して観察された応答と同様に、豚の心臓スライス心筋細胞内に自発的なカルシウム過渡症はありません。心筋細胞は、外部の電気およびβ-アドレナリン刺激に反応した。さらに、収縮力とイソプロテレノールへの応答は、新鮮な心臓スライスで観察されたのと同様の最大6日間、心臓スライス培養で維持された。MTTの生存率アッセイ、免疫染色、電子顕微鏡、カルシウム過渡測定、収縮機能評価、代謝評価、分析など、心臓スライスに対して、いくつかの構造評価と機能評価を行うことができます。現在、この培養システムを使用して、豚やヒトの心臓スライスに関心のある薬物および遺伝子治療の急性心臓毒性と有効性をテストしています。
