1. 制备装有细胞的SF-GelMA

三维（3D）生物印刷是生物制造和再生医学11、2、32,3中原生组织复杂建筑和功能复制品的一种有前途的方法。生物印刷的常见策略，包括喷墨、挤出和立体光刻印刷，从不同角度有优缺点。在这些技术中，挤出过程因其成本效益而最为常见。生物油墨在挤出生物印刷的工艺稳定性中起着关键作用。理想的细胞载生物油墨不仅应具有生物相容性，而且应适用于机械特性5。粘度低的生物油墨通常呈现为液体状状态。这些生物油墨易于快速沉积，避免在挤出过程中因高剪切应力引起的细胞膜损伤。然而，在需要长期印刷期的复杂情况下，低粘度往往导致生物油墨储液罐中封装细胞不可避免的沉淀，通常由重力驱动，导致生物油墨66、77中的不均匀细胞分散。因此，具有不均匀细胞分散的生物油墨阻碍了功能组织结构的体外生物打印。
最近几项以生物油墨为重点的研究都报道了封装细胞的同质分散性。一种基于双级交联的改性藻酸盐生物油墨用于挤出生物印刷8。在这项研究中，一种藻酸盐聚合物被用肽和蛋白质进行了改性。由于肽和蛋白质提供的附着位点，细胞在这种改良的藻酸盐中呈现比常用的藻酸盐更均匀分布。另外，混合生物油墨也被用来解决生物油墨中细胞的沉淀问题。另一项研究9中使用了含有聚乙烯乙二醇（PEG）和明胶或明胶二甲酰胺醇（GelMA）的混合生物油墨，具有更好的机械鲁棒性。封装的细胞呈现均匀分布，主要是因为混合生物油墨的粘度得到了提高。一般来说，影响生物油墨中封装细胞分散性的因素有若干，如生物油墨的粘度、细胞的重力、细胞的密度和工作周期。在这些因素中，细胞的引力在促进沉积中起着至关重要的作用。粘稠生物油墨提供的浮力和摩擦力已作为对抗重力的主要力量，迄今已于10日调查。
在这里，我们开发了一种新的策略，通过操纵生物油墨储液罐中的多个液体接口，促进生物油墨中封装细胞的均匀分散。生物油墨的多层修饰产生的这些液体界面不仅可以提供界面保留，延缓细胞的沉淀，还能保持生物油墨适宜的生物相容性和流变行为。在实践中，我们用多层方式修改了与丝纤维素（SF）的水性GelMA溶液（5%，w/v），纵向产生四个界面，在混合生物油墨中提供界面张力。因此，由于相邻细胞层的沉淀较少，细胞上的重力负荷被人为的界面张力所抵消，生物油墨中封装的细胞几乎均匀地分散。迄今为止，还没有类似的协议通过操纵液体生物油墨中的界面保留来减缓封装细胞的沉淀。我们在这里介绍我们的协议，以演示一种解决生物印刷中细胞沉淀的新方法。
