该协议使用激光显微解剖和RNA测序来分析肿瘤内异质性的差分mRNA表达，如间质细胞的积累区域或肿瘤入侵区域。该方法经过优化，可保持高质量的组织组织学和RNA完整性，从而促进高质量激光微分液样品的采集。此方法是敏感和高度可重复的。可用于研究各种肿瘤模型中的肿瘤形态和异质性。冷冻脑肿瘤组织的激光显微解剖是一种经济高效、可靠的技术，用于分离肿瘤组织细胞群离散解剖区域，以研究其分子特征。LMD可用于获得关于肿瘤进展过程中发生的分子事件的深入机械知识，并可以揭示新的治疗靶点。小鼠肿瘤负担的体内生物发光达到一个信号，在1倍10到6和1倍10到第7光子之间。通过安乐死肿瘤小鼠的循环系统冲洗含氧的酪氨酸溶液，直到肝脏和肺部完全清除血液。继续用溶解在酪蛋白溶液中的 30% 蔗糖溶液对动物进行进一步 15 分钟。要评估灌注的成功，请确认颈部、尾部和腿部在循环结束时是刚性的。然后按照标准方案收获大脑，将大脑储存在新鲜30%蔗糖溶液中过夜。为了准备低温保存，请将一个罐子装满冷异丙烷-2-甲基丁烷，然后将罐子放入装满液氮的容器中。当溶剂冷却时，将大脑印在一张滤纸上，将大脑放入标有 Cryomold 的纸张中。小心地在大约五毫升的最佳温度切割介质到 Cryomold 的中心，并放置大脑到模具中所需的方向。使用干净的钳子，快速将 Cryomold 放入冷冻异丙烷-2-甲基丁烷中 30 至 40 秒。一旦切割介质凝固，将 Cryomold 转移到干冰上，将 Cryomold 包裹在铝箔中，将冷冻脑组织包裹在零下 80 摄氏度的存储中。为了获得冷冻脑肿瘤组织部分，首先将低温恒温设定在零下20至零下24摄氏度之间，然后将样品块放入低温热室30至60分钟。当样品进行平衡时，用 100% 乙醇清洁腔室和刀架，然后用 RNase 清洁溶液喷洒截面刷。样品准备就绪后，取出模具并使用新鲜的切割介质将冷冻组织块连接到低温恒温器的试样盘上。将块放在圆盘支架中，然后将块与刀刃对齐。使用无 Rnase 的刷子之一获取大脑的 10 微米部分，小心地将组织片段压平并解开到切割表面上。要将脑组织部分安装到幻灯片上，请将标有无 RNase 的 PEN 玻璃幻灯片的带正向侧按到滑轨上，然后将幻灯片放入腔内的存储盒中。收集所有幻灯片后，将存储盒放在零下 80 摄氏度。要在激光微解剖之前溶解切割介质，将幻灯片浸入30秒的连续降乙醇浸入，然后将水晶紫罗兰溶液染色20秒，在0.5%eosin Y溶液中浸渍5秒。在 eosin Y 孵育结束时，使用滤纸将幻灯片挡干，并在连续上升乙醇浸入中脱水幻灯片。60 秒乙醇浸入后，在二甲苯容器中冲洗幻灯片三分钟。接下来，在室温下将无 RNase 表面的幻灯片干燥 10 秒，然后将样品安装到使用无 RNase 水准备的安装介质中。10 到 20 秒后，将幻灯片传输到显微镜显微切除平台上。对于激光捕获微切除，在打开激光之前打开电源。接下来，打开计算机中的显微镜控制器，启动激光采集微切除软件。在"显微镜控制"面板中，选择 10 倍的放大倍率。在激光控制下，设置组织解剖的激光参数，选择120赫兹的激光频率和100%的激光电流进行精确的激光微解剖，将速度设置为10，将孔径设置为10微米。然后将电源设置为 53。加载将捕获分区后的组织收集器，然后单击第二个卸载按钮。用无 0.5 毫升 PCR 平头管更换空收集器，每管含有 30 微升的解液缓冲液。将收集器返回到机器，然后单击继续。将加工过的试样加载到显微镜上，然后单击激光显微切除软件中的"卸载"。将样品上到滑动支架上，将滑动支架放在舞台上。单击"继续"，并在"剪切形状"窗口中选择"绘制剪切"窗口。使用显微镜控件查找感兴趣区域，并绘制有关感兴趣区域的轮廓。然后选择目标收集器管，然后单击"开始切割"以进行组织微切除。当所有感兴趣的区域都已解剖后，将收集管从支架转移到干冰上，直到它们进行下游处理。为了获得具有优越形态和RNA完整性的组织，对各种灌注方法进行了评估。要解剖感兴趣的区域，有必要用RNA无害染料染色组织。随后，用酪氨酸溶液和30%蔗糖灌注肿瘤小鼠，随后在30%蔗糖中隔夜孵育，可保存小鼠组织的形态和RNA完整性。虽然半成甲醛组织固定导致高质量的组织形态，但RNA的完整性受到负面影响。其他方法，如使用酪氨酸溶液孵育，或酪棒溶液加上30%蔗糖溶液孵育不会影响RNA质量，但观察到组织形态分辨率降低。如果组织没有安装介质，部分变得脱水，形态恶化，而安装组织样品与15%的安装介质溶解在水中保持高品质的组织形态。激光捕获显微解显微镜成像允许选择肿瘤组织进行解剖。然后，这个感兴趣区域选择允许解剖每个组织样本中感兴趣的区域，以进行RNA完整性分析。保持高质量的肿瘤组织形态和RNA完整性至关重要。泰洛德溶液和30%蔗糖灌注，在30%蔗糖中隔夜保存显著改善LMD的组织形态。使用 GAN 溶液快速固定、染色和安装是保持RNA质量和防止组织内形成裂纹的关键步骤。我们建议将每个组织面积的总肿瘤分段至少 2.5 倍至第 6 平方千分尺，以获得适当数量的 RNA 用于 RNA 质量控制和转录分析。LMD 能够分析调节胶质瘤异质性和入侵的分子信号通路，从而揭示临床前胶质瘤模型中未来转化发展的新的潜在目标。
