現在利用可能なさまざまな方法の中で、凍結脳腫瘍組織のレーザー捕捉マイクロ解剖(LMD)は、腫瘍組織からの離散解剖学的領域または特異的細胞集団の単離を可能にする費用対効果の高い、信頼性の高い技術であり、その分子プロファイル22,33を研究する。LMDは、選択した単一細胞または多細胞構造のmRNA遺伝子発現プロファイルの解析を可能にする4,5。54LMDは、腫瘍進行中に起こる分子事象に関する深い機械的な知識を得るために利用することができる。腫瘍組織の処理の改善は、組織形態およびRNA品質6の最適な光学分解能を得るために必要である。パラホルムアルデヒド固定は形態学的解析に最適な選択肢ですが、これらの条件下でRNAの品質が影響を受けて分解されるため、RNA-seq解析のRNA品質が低下します。凍結組織切片の使用は、細胞膜を破壊し、細胞内に穴を生成する可能性のある氷の結晶形成を回避し、RNA-Seq分析7のための最良の選択肢であり続ける。
ここでは、LMD用の凍結マウス脳腫瘍組織を処理するための最適化されたセクション固定および染色方法について述べている。氷の結晶が組織に形成されるのを防ぐために、我々は30%スクロースの溶液をマウスに浸透させた。この溶液は、極性水分子間の相互作用を破壊し、氷の結晶の形成を防ぎ、組織形態を維持します。組織染色は、腫瘍内の細胞または解剖学的に異なる領域の特定の集団を分化し、得るために必要である。RNAの完全性を維持するために、無害な染料で組織を固定し、染色することが不可欠です。以前に示されているヘマトキシリン/エオシン(H&E)を用いた組織の染色は、RNAの完全性を低下させる 8.エタノール、クレシルバイオレット4%、エオシンY 0.5%溶液で目的の組織を固定し、染色しました。クレシルバイオレットは、細胞核を濃い青色で染色する酸性の色素です。エオシンYは、細胞の基本的な成分を染色する好塩基性染料であり、細胞質と他の細胞構造8との区別を提供する。どちらの色素もエタノールに溶け、RNAの品質を低下させない。組織の損傷を避け、細胞構造の高い光学解像度を維持するために、我々はLMD9の前に組織切片を取り付けた。

このプロトコルは、レーザーマイクロディションとRNAシーケンシングを使用して、間葉細胞の蓄積領域や腫瘍浸潤領域などの腫瘍内不均一性のmRNA発現を分析します。この方法は、高品質のレーザーマイクロディスセクションサンプルの取得を容易にするために、良質の組織組織組織学とRNA完全性を維持するために最適化されています。この方法は、感度が高く、非常に再現性があります。様々な腫瘍モデルにおける腫瘍形態および不均一性を研究するために利用することができる。凍結した脳腫瘍組織のレーザー顕微鏡切除は、細胞集団の離散的な解剖学的領域を腫瘍組織から分離して分子プロファイルを研究するための費用対効果の高い、信頼性の高い技術である。LMDは、腫瘍進行中に起こる分子事象に関する深い機械的知識を得るために利用することができ、新しい治療標的を明らかにすることができる。マウス腫瘍の負担の生体発光は、1回10〜6回、7番目の光子の10倍の間のシグナルに達する。肝臓と肺が完全に血液を取り除くまで、安楽死腫瘍を持つマウスの循環系を通して酸素化されたタイロード溶液を洗い流す。さらに15分間、チロード溶液に溶解した30%スクロース溶液を動物に浸透させ続ける。灌流の成功を評価するために、首、尾、脚が循環の終わりに硬くなっていることを確認します。その後、標準的なプロトコルに従って脳を収穫し、新鮮な30%スクロース溶液で一晩脳を保存します。凍結保存の準備のために、冷たいイソペンタン-2-メチルブタンで瓶を満たし、液体窒素で満たされた容器に瓶を入れます。溶剤が冷えている間に、脳をろ過用紙の上で乾かし、脳をラベル付きのクリオマールルドに入れる。クライオモルドの中心に最適な温度切断媒体の約5ミリリットルで慎重に、所望の向きで型に脳を置きます。きれいな鉗子を使用して、すぐに30〜40秒間冷やしたイソペンタン-2-メチルブタンにクリオマールルドを置きます。切断媒体が固まったら、クライオモリをドライアイスに移し、クリオマールを包み、凍結した脳組織をアルミニウム箔で覆い、マイナス80°Cの貯蔵を行います。凍結した脳腫瘍組織切片を取得するには、まず、マイナス20度とマイナス24度の間のクライオスタットの温度を設定し、30〜60分間クライオスタットチャンバーにサンプルブロックを入れる。サンプルが平衡している間、100%エタノールでチャンバーとナイフホルダーを洗浄し、RNaseクリーニング溶液でセクションブラシをスプレーします。サンプルの準備ができたら、金型を取り出し、新しい切断媒体を使用して凍結した組織ブロックをクライオスタットの検体ディスクに取り付けます。ブロックをディスクホルダーに入れ、ブロックをナイフブレードに合わせます。Rnaseフリーブラシの1つを使用して脳の10マイクロメートルのセクションを取得し、組織片を切断面に慎重に平らにしてアンカールします。脳組織の切片をスライドに取り付けるには、ラベル付きRNaseフリーのPENガラススライドの正に帯電した側をセクションに滑らかに押し込み、スライドをチャンバー内の収納ボックスに入れます。すべてのスライドが収集されたら、保管ボックスをマイナス80°Cに置きます。レーザーマイクロ解剖前に切断媒体を溶解するには、スライドを30秒連続的に降水エタノール浸漬し、その後に0.5%eosin Y溶液で20秒間、5秒間の結晶紫色溶液染色を行います。エオシンYインキュベーションの終わりに、フィルターペーパーを使用してスライドを乾燥させてブロックし、順次上昇エタノール浸漬でスライドを脱水します。60秒のエタノール浸漬後、スライドをキシレンの容器に3分間リンスします。次に、RNaseフリー水で調製した取り付け媒体にサンプルを取り付ける前に、RNaseフリー表面のスライドを室温で10秒間乾燥させます。10~20秒後、顕微鏡顕微鏡の微細解剖プラットフォームにスライドを移します。レーザーキャプチャマイクロディシセクションの場合は、レーザーをオンにする前に電源を入れます。次に、コンピュータの顕微鏡コントローラをオンにして、レーザーキャプチャマイクロ解剖ソフトウェアを起動します。顕微鏡コントロールパネルで、10倍の倍率を選択します。レーザー制御の下で、組織解剖のためのレーザーパラメータを設定し、120ヘルツのレーザー周波数と100%のレーザー電流を選択して正確なレーザーマイクロ解剖を行い、速度を10に設定し、絞りを10マイクロメートルに設定します。次に、電源を 53 に設定します。セクションの後に組織をキャプチャする組織コレクターをロードし、2番目のアンロードボタンをクリックします。空のコレクターを、チューブあたり30マイクロリットルのライシスバッファーを含むDNaseおよびRnaseフリーの0.5ミリリットルPCRフラットヘッドチューブに交換してください。コレクターをマシンに戻し、続行をクリックして続行します。加工した標本を顕微鏡に積み込み、レーザーマイクロ解剖ソフトウェアで「アンロード」をクリックします。サンプルをスライドホルダーの上に上げ、スライドホルダーをステージに置きます。[続行] をクリックして続行し、[シェイプの切り取り] ウィンドウで [切り取り] を選択します。顕微鏡コントロールを使用して対象領域を見つけ、対象領域の周囲に輪郭を描きます。次に、デスティネーションコレクタチューブを選択し、[切り取り開始]をクリックして組織の微細解剖を進めます。関心のある領域がすべて解剖されたら、コレクタチューブをホルダーからドライアイスに移し、下流処理になるまで下流にします。優れた形態およびRNA完全性を有する組織を獲得するために、様々な灌流アプローチが評価された。関心のある領域を解剖するには、RNAの無害な染料で組織を染色する必要があります。その後、タイロード溶液と30%スクロースを有する腫瘍を持つマウスの灌流は、30%スクロース中で一晩のインキュベーションを行うと、マウス組織の形態およびRNA完全性の保存をもたらす。パラホルムアルデヒド組織の固定は、高品質の組織形態をもたらすが、RNAの完全性は悪影響を受けます。タイロード溶液インキュベーション、またはタイロード溶液+30%スクロース溶液インキュベーションを使用するなどの他のアプローチは、RNAの品質に影響を与えませんが、組織形態の分解能が低下することが観察されます。組織が実装媒体と一緒に取り付けられていない場合、セクションは脱水状態になり、水に溶解した15%の実装媒体を用いた組織サンプルの取り付けが高品質の組織形態を維持しながら形態が悪化する。レーザーキャプチャマイクロディスセクション顕微鏡イメージングは、解剖のための腫瘍組織の選択を可能にする。この関心領域選択により、各組織サンプル内の対象領域をRNA完全性解析に解剖することができます。高品質の腫瘍組織形態とRNAの完全性を維持することは非常に重要です。タイローデ溶液および30%スクロースの一晩保存と30%スクロース灌流は、LMDの組織形態を有意に改善する。迅速なライオマ固定、ガン溶液による染色および取り付けは、RNA品質を維持し、組織内での亀裂の形成を防ぐための重要なステップです。RNAの品質管理と転写分析の両方に適切な量のRNAを得るために、組織面積当たりの腫瘍全体を少なくとも2.5倍から6平方マイクロメートルに切除することをお勧めします。LMDは、神経膠腫の不均一性と浸潤を調節する分子シグナル伝達経路の分析を可能にし、前臨床グリオーマモデルにおける将来の翻訳発達のための新たな潜在的標的を明らかにすることができる。
