C.エレガンスは、開発を含む様々な研究のためのモデル生物として一般的に使用されています。この動物の単純な解剖学は、生殖腺、腸、透明キューティクルを含むいくつかの器官で構成されており、ワームの発達の分析を容易にする。生殖腺に存在する生殖細胞は、生殖細胞幹細胞が胚に成長し、最終的には次世代の子孫に成長するジテ5に成熟する方法を研究するのに最適なツールです。生殖細胞系列の遠位先端領域には、自己更新幹細胞のプールが含まれています(図1)。幹細胞がニッチを離れると、それらは大乳頭蓋に進行し、最終的には若年成人期に卵母細胞に発展する(図1)。生殖細胞系列におけるこの開発プログラムは、RNA結合タンパク質(RRP)6によって促進される転写後調節ネットワークを含む異なるメカニズムを介して厳しく調節される。6PPPは、RPPが他の補因子と関連付けて機能を発揮するので、この規制活動にとって重要です。

PPIのインシチュ検出における代替アプローチは、共免疫染色、フェルスター共鳴エネルギー伝達(FRET)、および二分子蛍光相補(BiFC)である。コ免疫染色は、固定ワーム組織での目的とする2つのタンパク質の同時検出と、シグナルの共局在化の程度の測定に依存します。標準的な顕微鏡検査7よりも詳細な超解像顕微鏡の使用は、200-300nm8の回折限界障壁を越えてタンパク質の共局在化をより厳格にテストするのに役立ちます。しかし、従来の顕微鏡と超解像顕微鏡の両方を使用した共同免疫染色は、明確に定義されたローカリゼーションパターンを持つタンパク質に最適です。対照的に、拡散分散型の相互作用パートナーにとってはあまり有益ではありません。重複に基づいてシグナルの共局在化を測定しても、タンパク質が互いに複雑であるかどうかについては正確な情報を提供しません9,,10.
さらに、タンパク質複合体の共免疫沈降と共免疫染色は定量的ではなく、そのような相互作用が有意であるかどうかを判断することは困難です。FRETとBiFCは、いずれも蛍光ベースの技術です。FRETは、あるFP(ドナー)からのエネルギーが別のFP(アクセプタ)11に伝達されるスペクトル重複を有する蛍光タンパク質(FP)と関心のあるタンパク質をタグ付けすることに依存する。このエネルギーの非放射移動は、それぞれの発光波長で検出することができるアクセクサFPの蛍光をもたらす。BiFCは、インビボの蛍光タンパク質の再構成に基づいています。これは、ヘリックス1-10とヘリックス1112などの2つの相補的な断片にGFPを分割し、その後、対象となる2つのタンパク質に融合することを必要とします。これら2つのタンパク質が相互作用する場合、GFPの相補的な断片は、折り畳み式と組み立てに十分近く、GFPフルオロフォアを再構成するほど近くなります。再構成されたGFPは、蛍光として直接観察され、PPIが発生した場所を示す。
そのため、FRETとBiFCの両方が、タグ付きタンパク質の機能を破壊する可能性のある大きな蛍光タグに依存しています。さらに、FRETとBiFCは、正確なデータを得るためにタグ付きタンパク質の豊富で同等の発現を必要とします。FRETは、一方のパートナーが他方を超えている実験には適しておらず、高い背景13につながる可能性がある。BiFC実験における過剰発現は、背景の増加をもたらす非特異的アセンブリ14を誘発する可能性もあるので、避けるべきである。どちらの手法でも、タグ付きタンパク質の発現条件と画像化条件の最適化が必要であり、実験の完了に必要な時間が長くなる可能性があります。
近接ライゲーションアッセイ(PLA)は、上述の技術の限界に対処できる代替アプローチである。PLAは、目的のタンパク質(またはそのタグ)を認識する一次抗体を利用します。これらの一次抗体は、40nm(または短い)距離15内で互いにハイブリダイズすることができるオリゴヌクレオチドプローブを含む二次抗体によって結合される。得られたハイブリダイズされたDNAはPCR反応を通じて増幅され、DNAを補うプローブによって検出される。これは顕微鏡によって視覚化される病巣で起因する。この技術は、複雑な組織(すなわち、ワーム生殖腺)においてPPIを検出することができ、これは、開発および分化の様々な段階で細胞を含む組立ラインとして組織される。PLAを使用すると、PPIは固定ワーム生殖腺で直接視覚化することができ、開発の特定の段階でPPIが発生するかどうかを調査する上で有利です。PLAは、正確な測定に最適な共ローカリゼーションベースのアッセイとは対照的に、PPIの解像度を高めます。使用する場合、超解像顕微鏡は、細胞内のPLA病巣の位置についてのより細かい詳細を提供する可能性を有する。もう1つの利点は、PLA反応に起因する病巣をImageJベースの解析ワークフローで数えることができることであり、この技術を定量化する。
