我们的斑马鱼异种移植药物筛选工作流程允许自动成像和定量人类癌细胞及其对药物治疗的反应。它可以用来快速测试耐心的癌细胞对已知药物的反应或识别新的抗癌化合物。使用斑马鱼异种图模型和药物筛选允许 Inviva 复制数字，以前只能通过体外系统获得。在通过大型化合物库筛选时，它也比小鼠模型更具成本效益。此工作流程有几个步骤，从斑马鱼癌细胞的注射到药物反应的成像，这些反应通过视觉演示获得最佳效果。首先，在5毫米PBS中将至少2倍10的离心机离到第六细胞，用200倍g进行5分钟，然后吸进上一代。将细胞调色板重新用五毫米的 dii 染色溶液中重新发送。在37摄氏度下孵育细胞，防止光线产生光，20分钟。然后轻轻涡流，在冰上孵化细胞15分钟，防止光线。然后将细胞以200倍g离心5分钟，吸进上一代。通过加入5毫升PBS，以200倍g离心5分钟，并吸升上一升，清洗细胞。再重复一次洗涤。每25万活细胞将细胞重新浓缩在PBS的微升中，并转移到1.5毫米微离心管中。在黑暗中将重新暂停的细胞放在冰上。在染色细胞和注射之前，通过将25毫莱的3%的阿加糖倒入培养皿中，使其凝固，使加糖板注射。将盘子存放在四摄氏度，最多两周。此外，在染色细胞和注射之前，在解剖显微镜下用钳子除氯两天受精后斑马鱼。用钳子从斑马鱼的保护巧克力的两端拉，直到夹层撕裂，斑马鱼变得未被保护。为了防止细胞在微注射过程中出现块状，在摄氏四度或冰上预冷却非丝状硅酸盐玻璃针。染色细胞后，立即使用微加载器移液器尖端将五微升染色细胞装入冷冻针头。将针头加载到微注射器臂中。使用无菌剃刀刀片将针尖平。使用级千分尺测量矿物油中的液滴大小。将液滴体积保持一致，在两纳米光，直径约1.5毫米。麻醉一分钟后，将麻醉幼虫转移到准备好的平面注射板中，用3%的阿加罗斯，并在所需的注射部位用一泵染色细胞注射幼虫。所有注射应在染色细胞后一到三个小时内完成，以防止针头的隆起。将幼虫从注射板中洗入含有E3培养基的10厘米培养皿中，不含甲基蓝，并在28摄氏度下孵育，恢复期为一小时。将注射的幼虫的培养皿放入34摄氏度的培养箱中。在架子和幼虫的培养皿之间放置一个空的培养皿，作为缓冲器。注射后48小时，筛选斑马鱼幼虫，用于荧光肿瘤细胞移植和健康。清除任何死斑马鱼或畸形斑马鱼，并选择具有类似镶体斑马鱼。去除未捕获的斑马鱼。对于蛋黄注射的鱼，去除在捕获的细胞质量周围看不到束缚边界的鱼。对于注射的鱼，去除注射细胞质量侵入蛋黄袋的鱼。要将选定的鱼转移到一个96井的盘子，首先切断一个200微升移液器尖端，只是足够大，四天后受精斑马鱼适合通过。使用 P200 移液器从盘子中吸出 150 微升 E3 介质，并将其添加到平底 96 井板的空井中。将 150 微升 2X 稀释药物溶液添加到每只含有斑马鱼幼虫的井中，达到每井 1X 药物溶液的最终体积 300 微升。在34摄氏度下孵育板。每天检查死斑马鱼。两天后，如果需要，在E3介质中用1X药物溶液或DMSO替换每井中200微升液体来刷新药物。在 Zen Blue 软件中，删除成像软件中所有不需要的荧光通道并添加 Dii 通道。检查所需的荧光通道。在同一窗口中，选择图像的拍摄、Z 堆栈、自动化、循环和序列等。在药物处理鱼之前，成像 DMSO 控制鱼。并在成像软件中设置适当的曝光时间。要量化荧光，请打开图像j软件。转到文件，打开并选择所需的 CZI 文件。该软件会显示一个导入选项窗口。要查看堆栈，请选择超堆栈，单独检查打开的文件，检查自动缩放，并检查拆分通道。对于颜色选项，选择着色。然后单击插件，宏，记录。点击图像，调整，阈值。在阈值窗口右侧的下拉菜单中选择红色图像类型。调整最小阈值，直到软件仅突出显示具有荧光的区域，然后单击"应用"。软件将照片转换为黑白，所选区域为黑色。单击分析和测量。软件拉起一个包含该图像荧光区域的结果窗口。单击"在宏记录器"窗口中创建。这将打开一个包含宏代码的新窗口。突出显示所有所需的图像进行分析，并根据堆叠查看或颜色选项打开。选择使用宏在窗口上运行。结果窗口现在包含每个图像的区域。在该协议中，在注射后48小时，异种鱼被筛查为荧光标记的肿瘤细胞，并经过化疗或DMSO治疗。总体而言，与DMSO处理的鱼相比，用Vincristine处理的异种移植鱼的异种移植细胞质量降幅最大、最一致。与Vincristine相比，经过德沙梅松治疗的鱼类的肿瘤面积减少了一半。但与DMSO相比，肿瘤面积仍有减少。此过程可用于筛查任何癌细胞系或患者样本，以了解不同药物或特定靶向疗法，从而深入了解每个患者的肿瘤。
