原発性患者癌またはヒト癌細胞株をモデル生物に異種移植することは、生体内での腫瘍の進行および行動、薬物治療に対する腫瘍応答、および微小環境との癌細胞相互作用を研究するために広く使用されている技術である。従来、細胞は免疫不全マウスに異種移植され、これは分野の標準のままです。しかし、このモデルシステムには、高コスト、低複製数、生体内での腫瘍負担を正確に定量化する難さ、腫瘍の生着や薬物検査にかかる時間の延長など、いくつかの制限があります。近年、ゼブラフィッシュは代替異種移植モデルとして登場し、2005年に最初に報告され、緑色蛍光タンパク質(GFP)標識ヒト黒色腫細胞株がブラツラ期胚11,22に移植された。さらに最近では、2日間のポスト受精(dpf)ゼブラフィッシュ幼虫が異種移植片のレシピエントとして使用され、周囲の微小環境33,44との腫瘍相互作用の生体内イメージングにおける解像の高分解能の制御を可能にしている。

ゼブラフィッシュ異種移植薬スクリーニングワークフローは、ヒトがん細胞の自動イメージングと定量化、薬物治療への反応を可能にします。これは、迅速に既知の薬物に忍耐の癌細胞の応答をテストしたり、新しい抗癌化合物を同定するために使用することができます.ゼブラフィッシュゼノグラフモデルと薬物スクリーニングを使用すると、以前はインビトロシステムでのみ得られたInvivaレプリカント番号が可能になります。また、化合物の大規模なライブラリを介してスクリーニングする際にマウスモデルよりもはるかに費用対効果が高いです。このワークフローは、ゼブラフィッシュにおけるがん細胞の挿入から、視覚的なデモンストレーションによって最もよく学ばれる薬物応答のイメージングまで、いくつかのステップを有する。始めに、500回gでPBSの5ミリメートルで5ミリメートルの6番目の細胞に最低20倍10〜5分間遠心し、上清を吸引する。5ミリメートルのdii染色溶液で細胞パレットを再中断します。20分間光から保護された37°Cで細胞をインキュベートします。その後、穏やかに渦を、光から保護された15分間氷上の細胞をインキュベートします。その後、細胞を200回gで5分間遠心し、上清を吸引する。PBSを5ミリリットル加え、200倍gで5分間遠心し、上清を吸引して細胞を洗浄します。さらに1回洗い、洗い流します。25万個の生細胞につき1マイクロリットルのPBSで細胞を再懸濁し、1.5ミリメートルマイクロ遠心チューブに移します。再懸濁した細胞を暗闇の中で氷の上に置いておきます。細胞を染色して注入する前に、1X TBEに3%のアガロースの25ミリリアをペトリ皿に注ぎ、固化させることで注入するためのアガロースプレートを作ります。プレートを摂氏4度で2週間保存します。また、細胞を染色して注入する前に、解剖顕微鏡の下で鉗子を使用して2日後の受精後シマウマ魚をデクリニングする。シマウマの魚の保護絨毛の反対側の端から、絨毛が裂け、シマウマの魚が包み込まれなくなるまで鉗子で引っ張ります。マイクロインジェクション中の細胞の凝集を防ぐために、4度または氷上で非糸状ホウケイ酸ガラス針を冷やしてください。細胞を染色した直後に、マイクロローダーピペットチップを使用して、5マイクロリットルの染色された細胞を冷蔵針にロードします。マイクロインジェクターアームに針をロードします。滅菌カミソリの刃を使用して針先をレベル付けします。ステージマイクロメーターを使用して、鉱物油中の液滴サイズを測定します。液滴の体積は直径約1.5ミリメートルの2ナノリットルで一貫して保ちます。麻酔の1分後、麻酔した幼虫を3%のアガロースで調製した平らな表面注入プレートに移し、所望の注射部位に染色された細胞のポンプ1つで幼虫を注入する。すべての注射は、針の凝集を防ぐために細胞を染色した後、1〜3時間以内に完了する必要があります。注射プレートからメチレンブルーを入れた10センチメートルのシャーレに幼虫を洗い、摂氏28度で1時間の回収期間インキュベートします。注射した幼虫の皿を摂氏34度の保育器に移します。棚と幼虫のシャーレの間に置かれた空のシャーレで、緩衝液として機能します。48時間の注射後、蛍光腫瘍細胞の生着および健康のためのゼブラ魚の幼虫をスクリーニングする。死んだ魚や奇形のシマウマの魚を取り除き、同様の生着を持つシマウマの魚を選択します。未移植シマウマの魚を取り除きます。黄身注入魚の場合は、生着した細胞塊の周りにヨークの境界が見えない魚を取り除きます。心膜注入魚の場合は、注入された細胞塊が黄身袋に侵入する魚を取り除きます。選択した魚を96ウェルプレートに移すために、最初に200マイクロリットルのピペットチップから先端を切り落とし、4日間の受精後シマウマ魚が収まるのに十分な大きさです。P200ピペットを使用してプレートから1シマウマの魚とE3培地の150マイクロリットルを吸引し、平らな底96ウェルプレートの空の井戸に追加します。シマウマの魚の幼虫を含む各井戸に150マイクロリットルの2X希釈薬物溶液を加え、井戸あたり1X薬液で300マイクロリットルの最終体積に達する。プレートを摂氏34度でインキュベートします。毎日死んだシマウマの魚をチェックしてください。2日後、必要に応じて、各ウェルから200マイクロリットルの液体をE3培地中の1X薬物溶液またはDMSOに置き換えることによって薬物をリフレッシュする。Zen blueソフトウェアでは、イメージングソフトウェア内の不要な蛍光チャネルをすべて取り外し、Diiチャンネルを追加します。希望の蛍光チャネルを確認します。同じウィンドウで、画像の撮影方法、Zスタック、オートメーション、ループ、シリーズ、エトセトラを選択します。薬物処理魚の前にDMSOコントロール魚を画像化します。そして、撮像ソフトウェアに適切な露光時間を設定する。蛍光を定量化するために、オープン imagej ソフトウェア。ファイルに移動し、開いて、目的の CZI ファイルを選択します。ソフトウェアは、インポートオプションウィンドウを表示します。スタック表示の場合は、ハイパースタックを選択し、開いているファイルを個別にチェックし、自動スケールを確認し、分割チャンネルを確認します。色のオプションで、[色付き] を選択します。次に、プラグイン、マクロ、レコードをクリックします。画像をクリックし、調整し、しきい値を調整します。しきい値ウィンドウの右側にあるドロップダウンメニューで、画像の種類を赤として選択します。ソフトウェアが蛍光を持つ領域のみをハイライト表示するまで最小しきい値を調整し、[適用] をクリックします。ソフトウェアは、黒で選択した領域と白黒に写真を変換します。[分析と測定] をクリックします。ソフトウェアは、その画像の蛍光領域を含む結果ウィンドウを引き上げる。マクロ記録ウィンドウで作成をクリックします。マクロのコードを含む新しいウィンドウが開きます。解析に必要なすべての画像をハイライト表示し、スタック表示またはカラーオプションに従って開きます。マクロを使用してウィンドウで実行を選択します。結果ウィンドウに、各画像の領域が表示されます。このプロトコルでは、注射後48時間で、異種移植された魚を蛍光標識腫瘍細胞についてスクリーニングし、化学療法またはDMSOで治療した。全体として、ビンクリスチンで処理された異種移植魚は、DMSO処理魚と比較して、異種移植細胞量の最大かつ最も一貫した減少を示した。デキサメタゾン治療魚はビンクリスチンと比較して腫瘍領域の約半分の減少を示した。しかし、DMSOと比較して腫瘍領域の減少を示した。この手順は、各患者の腫瘍に関する洞察を与える異なる薬物または特定の標的療法への応答のために、任意の癌細胞株または患者サンプルをスクリーニングするために使用することができる。
