この方法により、統合アミロイドタンパク質構造と、可溶性および蓄積されたタンパク質構造を明確に区別することができます。この技術は、毒性の副作用がほとんどまたはまったくない非侵襲的な方法で生体内で行われる。蛍光植物自体の蛍光特性に依存するので、時間の経過とともに非常に堅牢で再現性が高い。FLIMは癌診断の分野で利用されている。しかし、神経変性疾患に関するタンパク質凝集の基本的なメカニズムを研究するために採用しています。この方法論は、タンパク質凝集の分野で使用することができる。疾患関連タンパク質が蛍光プローブでタグ付けされている限り、それは時間の経過とともに追跡および監視することができる。凝集を促進または防止する化合物および戦略を、正常に研究することができます。光顕微鏡で可視化できる任意の生物学的システムは、例えば細胞培養、固定または透過サンプルおよび必要な任意の媒体において、in vivoまたはex vivoのいずれかに適しています。実験データを取得する前に線虫の中で選択した蛍光素の特性をテストし、測定を取得する前にセットアップが完全に機能することをテストしてください。まず、NGMプレート上で20°Cで線虫を成長させ、維持します。若い成人として人生の4日目まで線虫を老化させ、老人として人生の8日目を過ごします。イメージングの日に、イメージングスライドを準備します。アガロースと二重蒸留水を3%重量の濃度で置きます。電子レンジに入れて溶かし、少し冷まします。1ミリリットルのピペットチップの先端を切り、溶かしたアガロースの約200マイクロリットルを取ります。きれいなガラススライドにアガロースをピペットし、すぐに任意の気泡の形成を避けるために上に第二のものを置きます。スライドを乾燥させ、上のガラススライドをそっと取り除きます。結果は線虫が配置される均一のアガロース表面を有するガラススライドである。イメージングのセットアップが準備ができたら、ステレオ顕微鏡の下で働き、10マイクロリットルの麻酔化合物をアガロースパッドに置き、5~10個の線虫をそっと移します。まつげの先端を使用して線虫を分離します。それらを近くに保ちますが、画像取得中に線虫のローカライズを容易にするために触れないようにしてください。慎重にカバースリップで線虫をオーバーレイします。取り付け後1時間以内に、測定を行います。線虫が完全に不動であることを確認してください。FLIM 取得ソフトウェアを開きます。タブボタンを見つけて、出力を有効にします。ステージ上に取り付けられたC.elegansとスライドを配置し、伝送モードで10倍レンズを使用して、スライド上の線虫の位置を局所化します。スライドを取り外し、目的を63倍レンズに切り替え、必要な浸漬媒体を適用します。スライドをステージ上に置き、線虫を局地に入れる。サンプルのスキャンを開始します。対象の領域を選択し、最大投影面に焦点を当てます。FLIM ソフトウェアのインターフェイスで、検出されたフォトンの数をプレビューします。ADC 値は、10 ~ 4 番目の 10 から 5 番目の 10 倍の間にする必要があります。必要に応じて、別の平面に焦点を合わせるか、レーザーパワーを増やして、より多くの光子を収集します。フォトンの積み重ねを避けるが、適切な寿命フィットと測定を作成するために十分な量の光子を収集することを確認してください。メニューバーで、取得パラメータを設定するタブを選択します。スキャン同期を選択して、単一のフォトン検出を許可します。取得を固定時間またはフォトン数に設定します。取得を開始するには、[開始]を押します。ソフトウェアを開き、ファイルを介してFLIMデータファイルをインポートし、FLIMデータをロードします。異なるセッションで、異なる生物学的反復から得られた場合でも、1つの条件からすべてのサンプルをロードします。必要に応じて、セグメントは、セグメンテーション、セグメンテーションマネージャを介して多くのFLIM画像のための線虫を信号する。トリミング ツールを目的の領域の周囲にドラッグして、ハイライト表示します。完了したら、[OK] をクリックします。C.elegans の強度ベースのイメージが表示される小さな領域を選択します。その領域の減衰曲線は、インタフェースの右側にある大きな減衰ウィンドウに表示されます。有効期間を推定するには、[データ] タブで、40 ~ 300 の任意の統合最小値を設定して、薄暗すぎて適切なピクセルを除外します。FLIM 信号をこれらの値に制限するには、時間の最小値と時間の最大値を選択します。1 のプリセットカウントフォトンは変更しないでください。取得時に使用したレーザーのメガヘルツで繰り返し率を入力します。ゲートの最大値を入力して、すべての飽和ピクセルを除外します。[有効期間] タブで、使用するグローバル 継手を選択します。選択した蛍光減衰が多重指数であり、1 回の寿命を超える場合は、指数選択の数を除き、他のパラメーターを変更しないでください。IRF メニューから IRF をアップロードします。IRF シフトを見積もるためには、IRF を選択し、IRF シフトを見積もります。値のセットが自動的に IRF タブに表示されます。これが確立されたら、このタブの他のパラメータは変更しないでください。すべてのパラメーターを設定したら、fit データ・セットを押します。青い線でハイライト表示されたフィット感は、すべてのイベントと重なっている必要があります。すべてのイベントが適合に沿って揃えられると、適切な適合が得られます。ソフトウェアのインターフェースの右上のメニューにあるパラメータタブをクリックし、統計値、加重平均を選択し、カイ二乗値が可能な限り近い値であることを確認します。このように選択された画像の寿命値はタウ1として明らかにされる。ファイル、エクスポート強度画像、フィット結果表、画像、ヒストグラムを介して関心のある情報をエクスポートします。4日目または8日目の筋肉質Q40 RFPを表現するC.elegansの代表的な地図をここに示しています。IQ85のQストレッチが長いほど凝集しやすく、4日目のヒストグラムに蛍光寿命シフトが既に示された。実際、4日目には、IQ44にまだ存在しない間にIQ85の病巣形成が観察された。しかし、老化の際、IQ44は病巣形成を示し、したがって蛍光寿命を減少させた。最後に、NQ40CFP株における蛍光寿命の低下も検出されなかった。この株については、老化時の平均蛍光寿命にわずかな有意な変化しかありませんでした。線虫モデルが確立されると、収集される光子の量は、良好な寿命フィットを得るために最も重要です。この方法は、さらに、フォスター共鳴エネルギー伝達、FRET、光漂白後の蛍光回復、FRAP、または測定などの異なる技術と結合することができる。これらの適応はすべて、タンパク質研究の特性とその凝集形態に関する追加情報を提供します。タンパク質凝集およびタンパク質恒常性の分野では、この技術は、システムに課せられた摂動、異なるタンパク質種の分布、人身売買、および一般的に生物内の可溶性および不溶性タンパク質画分の違いと有意性を調査することを可能にした。麻酔薬のアジ化ナトリウムは有毒であり、手袋とゴーグルで処理し、ヒュームフードの下で希釈する必要があります。この技術は顕微鏡ベースであるため、レーザーの処理に関連するすべての警告に従うことが重要です。
