ISH, met behulp van een gelabelde nucleïnezuursonde om de specifieke DNA- of RNA-sequentie van belang te detecteren, is een nuttige methode om het ruimtelijke expressiepatroon van doelgenen in morfologisch bewaarde weefsels te beschrijven1,2,3. Normaal gesproken wordt de doelsequentie gegenereerd door polymerase kettingreactie (PCR) en vervolgens gebruikt als sjabloon om de antisense/sense RNA-sonde te synthetiseren met digoxigenin uridine-5'-trifosfaat. Monsters worden vastgemaakt en permeabilized voor incubatie met riboprobe. Na het afwassen van de overtollige sonde, hybridisatie wordt gevisualiseerd door immunohistochemie met behulp van een anti-digoxygenin antilichaam, dat is alkalische fosfatase-geconjugeerd3,4,5,6.
Sipunculus nudus (Phylum Sipuncula; order Sipunculida: Sipunculidae) is een niet-gesegmenteerde, coelomaat en bilateraal symmetrische mariene worm7,8. Sipunculus nudus is een kosmopolitische soort op grote schaal verspreid in tropische en gematigde kustwateren. Het is ook een belangrijke mariene visbestanden in het zuiden van China vanwege de hoge voedingswaarden en geneeskrachtige waarden9,10. Echter, Sipunculus nudus in moleculaire biologie staat nog in de kinderschoenen. Om de biologische rol van genen volledig te begrijpen, is het onderzoek naar genenexpressiepatronen bij een cellulaire resolutie van groot belang. In het niet-modelorganisme Sipunculus nudusis de ISH-methode, een ideale methode om de genenexpressiepatronen te detecteren, nog niet vastgesteld. De coelomic vloeistof bevat verschillende celtypes, waaronder granulocyten, urncellen, vesiculaire cellen, kiemcellen, erythrocyten,enz. De dubbele seks /mab-3 gerelateerde transcriptie factor-1 (dmrt1), gebruikt als een representatief gen in deze methode, is een sterk geconserveerde transcriptie regulator van geslachtsbepaling en differentiatie in de meeste soorten, variërend van ongewervelde dieren tot zoogdieren12,13. In een reeks soorten (d.w.z. zwarte porgy, enz.) 14, dmrt1 werd uitgedrukt in de Sertoli cellen, rond de kiemcellen, waarvan de functie is vergelijkbaar met trofoblast cellen van Sipunculus nudus. Daarom veronderstelden we dat dmrt1 van Sipunculus nudus wordt uitgedrukt in trofoblastcellen van spermatozeugmata, en het resultaat van de ISH-methode bevestigde de hypothese duidelijk.
Dit protocol beschrijft voor het eerst ISH, met digoxigenin-gelabelde antisense/sense mRNA-sondes, om mRNA-expressiepatronen in zijn coelomic vloeistofuitstrijkje te bepalen. De optimale reactieomstandigheden worden geleverd, die een zeer gevoelige visualisatie van mRNA-expressie bij hoge resolutie mogelijk maken. De ontwikkelde ISH-methode kan mogelijk worden toegepast in meer Sipunculida-soorten anders dan Sipunculus nudus.

In situ hybridisatie (ISH) is een zeer informatieve techniek om cellulaire distributiepatronen van specifieke genen (bijvoorbeeld mRNA en ncRNA) in weefsels te presenteren. De sipunculid worm Sipunculus nudus is een cruciale visbestanden omdat het hoge voedingswaarden en medicinale waarden heeft. Momenteel staat het onderzoek naar de moleculaire biologie van Sipunculus nudus nog in de kinderschoenen. Het doel van dit artikel is het ontwikkelen van een gevoelige methode voor het lokaliseren van specifieke mRNA in Sipunculus nudus coelomic vloeistof. Het protocol omvat gedetailleerde stappen van ISH, met inbegrip van digoxigenin-gelabeld antisense en zin riboprobe voorbereiding, coelomic vloeistof collectie en sectie voorbereiding, specifieke riboprobe hybridisatie, antilichaam incubatie, kleuring en post-coloration behandelingen. De representatieve resultaten verkregen uit een succesvol experiment met behulp van deze methode worden aangetoond. Het protocol moet ook van toepassing zijn op andere Sipuncula-soorten.

Een samenvatting van de stappen die betrokken zijn bij ISH wordt geïllustreerd in figuur 1. Antisense en overeenkomstige zin riboprobes voor dmrt1 werden gesynthetiseerd uit PCR-producten versterkt uit de coelomic vloeistof cDNO's. In situ hybridisatie is een zeer informatieve techniek om mRNA en IncRNA expressie patronen van specifieke genen in weefsels te presenteren. Ons doel in dit artikel is het ontwikkelen van een gevoelige methode voor het opsporen van de specifieke mRNA lokalisatie in coelomic vloeistof van Sipunculus nudus, dat is een cruciale visbron. We hebben de representatieve resultaten van onze succesvolle experimenten gepresenteerd met behulp van deze methode.Het is te hopen dat onze methode zal worden toegepast op andere Sipuncula soorten voor het identificeren van de expressiepatronen van de specifieke mRNA en IncRNA in coelomic vloeistof. Bevestig de Sipunculus nudus op de dissectie tafel. Open het lichaam van de Sipunculus nudus met kleine autoclaved schaar.Verzamel en breng vervolgens de coelomic vloeistof met pipet over naar poly-D-lysine behandelde microscoopdia's, die zich mild verspreiden. Luchtdrogen de glijbanen voor een uur op 37 graden Celsius. Was de glijbanen met DEPC-PBS drie keer, vijf minuten per wasbeurt met zachte agitatie.Giet de PBS af, voeg de proteinase K-oplossing vijf minuten toe op de glijbanen bij kamertemperatuur. Giet de proteinase K-oplossing af. Voeg de PFA-oplossing 20 minuten toe op de glijbanen bij kamertemperatuur.Giet de PFA-oplossing af. Was de glijbanen met DEPC-PBS drie keer, vijf minuten per wasbeurt met zachte agitatie. Giet de PBS af.Voeg 50 microliter riboprobe toe. Voeg een coverlip toe. Doe de glijbanen in de natte doos en sluit goed af met paraffinefilm.Hybridiseren 's nachts op 60 graden Celsius. Dompel de glijbanen onder in de wasbuffer en laat deze staan totdat de coverslip automatisch afglijdt. Was de glijbanen met de voorverwarmde wasbuffer twee keer bij 65 graden Celsius, 30 minuten per wasbeurt met zachte agitatie.Was de glijbanen met de voorverwarmde SSC-oplossing twee keer bij 65 graden Celsius, 30 minuten per wasbeurt met zachte agitatie. Was de glijbanen met de MABT-oplossing twee keer bij kamertemperatuur. 30 minuten per wasbeurt met zachte agitatie.Giet de MABT af. Voeg de blokkeringsbuffer toe. Broed de glijbanen drie tot vier uur op kamertemperatuur.Laat de blokkeringsbuffer leeglopen. Voeg het antilichaam toe, broed de dia's 's nachts bij vier graden Celsius uit. Giet de antilichaamoplossing af.Was de glijbanen met de MABT-oplossing vier keer, 25 minuten per wasbeurt met zachte agitatie. Giet de MABT af. Voeg de alkalische fosfatase buffer voor drie keer, vijf minuten per incubatie.Verwijder de alkalische fosfatase buffer. Voeg de BCIP-NBT kleuroplossing toe. Houd de dia's in het donker.De kleuringstijd varieert van minuten tot uren, zelfs 's nachts, wat afhankelijk is van de signaalintensiteit. De kleuringsstatus moet aan het begin van de kleuring vaker worden gecontroleerd. Bijvoorbeeld met een interval van enkele minuten, en enkele uren ten laatste van de kleuring.De controletijd moet zo kort mogelijk zijn om te voorkomen dat er een hoge achtergrondsignalering wordt ingevoerd als gevolg van de blootstelling aan licht. De representatieve signalen van in situ hybridisatie worden getoond. In situ hybridisatie van Sipunculus nudus coelomic vloeistof met anti-sense riboprobe dat richt zich op dmrt1 geeft paarse vlekken geconcentreerd in trofoblast cellen van de spermatozeugmata, figuur 2A en B.Sense riboprobe voor dmrt1 niet detecteren een hybridisatie signaal, figuur 2C.Na het bekijken van deze video, zou je een zeer goed begrip van hoe de expressie patronen van doel mRNA of IncRNA visualiseren in de coelomic vloeistof van Sipunculus nudus.
