בהכלאה situ היא טכניקה אינפורמטיבית מאוד להציג mRNA ודפוסי ביטוי IncRNA של גנים ספציפיים ברקמות. מטרתנו במאמר זה היא לפתח שיטה רגישה לגילוי לוקליזציה mRNA ספציפי בנוזל הקולומי של עירום סיפונקולוס, שהוא משאב דיג חיוני. הצגנו את התוצאות הייצוגיות שהתקבלו מהניסויים המוצלחים שלנו באמצעות שיטה זו.יש לקוות כי השיטה שלנו תחול על מינים אחרים Sipuncula לזיהוי דפוסי הביטוי של mRNA ספציפי IncRNA בנוזל קוסמי. תקן את עירום סיפונקולוס על טבלת הניתוחים. פתח את גופו של עירום סיפונקולוס עם מספריים אוטוקלאבים קטנים.לאחר מכן לאסוף ולהעביר את הנוזל הקואלומי עם פיפטה לשקופיות מיקרוסקופ פולי-D-לינזין שטופלו, להתפשט במתינות. מייבשים את המגלשות למשך שעה ב-37 מעלות צלזיוס. לשטוף את השקופיות עם DEPC-PBS במשך שלוש פעמים, חמש דקות לכל לשטוף עם תסיסה עדינה.מסננים את ה-PBS, מוסיפים את תוסף החלבון K על השקופיות במשך חמש דקות בטמפרטורת החדר. מסננים את תסנון החלבון K. הוסף את פתרון PFA על השקופיות במשך 20 דקות בטמפרטורת החדר.רוקן את פתרון PFA. לשטוף את השקופיות עם DEPC-PBS במשך שלוש פעמים, חמש דקות לכל לשטוף עם תסיסה עדינה. לנקז את PBS.מוסיפים 50 ריבופרוב מיקרוליטר. הוסף כיסוי. שים את השקופיות לתוך הקופסה הרטובה וחותם היטב עם סרט פרפין.הכלאה לילה ב 60 מעלות צלזיוס. טבול את השקופיות במאגר הכביסה ותן לו לעמוד עד שהכיסוי יחליק באופן אוטומטי. לשטוף את השקופיות עם חיץ לשטוף שחומם מראש במשך פעמיים ב 65 מעלות צלזיוס, 30 דקות לכל לשטוף עם תסיסה עדינה.לשטוף את השקופיות עם פתרון SSC שחומם מראש במשך פעמיים ב 65 מעלות צלזיוס, 30 דקות לשטוף עם תסיסה עדינה. לשטוף את השקופיות עם פתרון MABT פעמיים בטמפרטורת החדר. 30 דקות לשטוף בתסיסה עדינה.לנקז MABT. הוסף את מאגר החסימה. דגירה המגלשות בטמפרטורת החדר במשך שלוש עד ארבע שעות.רוקן את מאגר החסימה. מוסיפים את הנוגדן, דגירה את המגלשות בארבע מעלות צלזיוס לילה. לנקז את פתרון הנוגדנים.לשטוף את השקופיות עם פתרון MABT במשך ארבע פעמים, 25 דקות לכל לשטוף עם תסיסה עדינה. לנקז MABT. מוסיפים את מאגר הפוספטזה אלקליין במשך שלוש פעמים, חמש דקות לכל דגירה.הסר את מאגר הפוספטזה אלקליין. הוסף את פתרון הכתמים של BCIP-NBT. שמור את השקופיות בחושך.זמן הכתמים משתנה בין דקות לשעות, אפילו במהלך הלילה, מה שתלוי בעוצמת האות. יש לבדוק את מצב הכתמים בתדירות גבוהה יותר בתחילת הכתם. לדוגמה, עם מרווח של מספר דקות, וכמה שעות לכל המאוחר של הכתם.זמן הבדיקה צריך להיות קצר ככל האפשר כדי למנוע החדרת איתות רקע גבוה עקב החשיפה לאור. האותות הייצוגיים של היברידיות במקום מוצגים. בהכלאה situ של נוזל קוסמי סיפונקולוס nudus עם ריבופרוב אנטי חושי כי מטרות dmrt1 מציין כתמים סגולים מרוכזים בתאי trophoblast של spermatozeugmata, איור 2A ו B.Sense ריבופרופ עבור dmrt1 לא לזהות כל אות הכלאה, איור 2C.לאחר צפייה בסרטון זה, תהיה לך הבנה טובה מאוד כיצד לדמיין את דפוסי הביטוי של mRNA היעד או IncRNA בנוזל הקואלומי של עירום סיפונקולוס.
