Tilapia lake virus disease (TiLVD) is een virale ziekte in tilapia(Oreochromis spp.) die naar verluidt tilapia sterfgevallen veroorzaakt in veel regio's van de wereld, waaronder Azië1,,2, Afrika en Amerika. De ziekte werd voor het eerst erkend tijdens de massale sterfte van tilapia in 2009 in Israël, waar het aantal wilde tilapia in het Kinneretmeer drastisch kelderde van 257 tot 8 ton per jaar2. De ziekte wordt veroorzaakt door het tilapiameervirus (TiLV), dat aan de familie Amnoonviridae is toegewezen als een nieuw geslacht Tilapinevirus en een nieuwe soort Tilapia tilapinevirus3. Genetische karakterisering van TiLV toonde aan dat het virus is een nieuwe omhuld, negatief-zin, single-stranded RNA virus dat 10 segmenten codering 10 eiwitten1,2,4. Verschillende soorten tilapia in het geslacht Sarotherodon, Oreochromis, en Tilapine en andere warmwatervissen (bijvoorbeeld reuzengourami (Osphronemus goramie))zijn vatbaar gebleken voor TiLV2,5. Momenteel blijft dit virus zich wereldwijd verspreiden, mogelijk door de beweging van besmette levende vissen6,7, terwijl het risico van virale overdracht via bevroren tilapia of het product ervan beperkt is8. Aanzienlijke sterfte als gevolg van TiLV-infectie heeft het potentieel om een aanzienlijk schadelijk economisch effect op de tilapia-industrie hebben. Bijvoorbeeld, de economische impact van de zomer sterfte syndroom in Egypte in verband met TiLV infectie werd berekend op US $ 100 miljoen9. Daarom is het belangrijk om een snelle en goede diagnostische methode te ontwikkelen om de bestrijding van deze ziekte in viskwekerijen te vergemakkelijken.
Tot nu toe was de diagnose van TiLVD gebaseerd op moleculaire testen, virale isolatie en histopathologie. Voor TiLV diagnose10,11zijn verschillende PCR protocollen en primers ontwikkeld. Bijvoorbeeld, een SYBR groen-gebaseerde reverse transcription kwantitatieve PCR (RT-qPCR) methode met de gevoeligheid voor het detecteren van zo weinig als twee kopieën / μL van het virus is ontwikkeld en gevalideerd voor TiLV detectie10. Andere PCR-methoden voor TiLV-detectie zijn TaqMan quantitative PCR11,RT-PCR2,geneste RT-PCR12en semi-geneste RT-PCR13. Deze methoden vereisen echter geavanceerde laboratoriumapparatuur en relatief langere perioden om resultaten op te leveren vanwege de complexiteit van de reacties, waardoor ze ongeschikt zijn voor veldtoepassing.
De lus-gemedieerde isothermale versterking (LAMP) test is een snelle, eenvoudige en praktische for-field toepassing14,15. De techniek maakt gebruik van het principe van een bundelverplaatsingsreactie, terwijl de versterkingsreactie onder isothermale omstandigheden loopt zonder een geavanceerde en dure thermische cycler14,15. Bijgevolg worden versterkte LAMP-producten of RT-LAMP-producten geanalyseerd in ladderachtige banden met behulp van agarosegelelektroforese met een fluorescerende vlek voor ofwel de veilige visualisatie van DNA of RNA14 of observatie met het blote oog voor de aanwezigheid van troebelheid of een witteneerslaan16,17,18. Om deze redenen is deze techniek gebruikt voor de detectie ter plaatse van verschillende vispathogenen17,18,19,20,21,22,23,24,25,26,27. Het doel van deze studie was om een snelle, gevoelige en nauwkeurige RT-LAMP test voor TiLV detectie vast te stellen. De RT-LAMP test biedt screening voor TiLV in vismonsters binnen 30 min. De techniek kan worden toegepast voor de diagnose en bewaking van TiLVD.

Tilapia lake virus disease (TiLVD), een opkomende virale ziekte in tilapia veroorzaakt door het tilapia meer virus (TiLV), is een aanhoudende uitdaging in de aquacultuur-industrie die heeft geresulteerd in de massale morbiditeit en mortaliteit van tilapia in vele delen van de wereld. Een effectieve, snelle en nauwkeurige diagnostische test voor TiLV-infectie is daarom noodzakelijk om de eerste infectie op te sporen en de verspreiding van de ziekte in de aquacultuurlandbouw te voorkomen. In deze studie wordt een zeer gevoelige en praktische reverse transcription loop-gemedieerde isothermale versterking (RT-LAMP) methode gepresenteerd om tilapia lake virus in visweefsel op te sporen. Een vergelijking van de RT-qPCR en RT-LAMP testen van geïnfecteerde monsters bleek positieve resultaten in 63 (100%) en 51 (80,95%) respectievelijk monsters. Bovendien bleek uit een analyse van niet-geïnfecteerde monsters dat alle 63 niet-geïnfecteerde weefsels negatieve resultaten opleverden voor zowel de RT-qPCR als de RT-LAMP-test. De kruisreactiviteit met vijf ziekteverwekkers in tilapia werd geëvalueerd met behulp van RT-LAMP, en alle tests toonden negatieve resultaten. Zowel de lever en slijm monsters verkregen van besmette vissen toonde vergelijkbare resultaten met behulp van de RT-LAMP methode, wat suggereert dat slijm kan worden gebruikt in RT-LAMP als een niet-dodelijke test om te voorkomen dat het doden van vis. Tot slot toonden de resultaten aan dat de gepresenteerde RT-LAMP-test een effectieve methode biedt voor TiLV-detectie in tilapiaweefsel binnen 1 uur. De methode wordt daarom aanbevolen als een screening instrument op boerderijen voor de snelle diagnose van TiLV.

In deze studie werd een RT-LAMP test ontwikkeld om TiLV-infectie in tilapia op te sporen. De geteste monsters werden verzameld van 14 bedrijven in verschillende delen van Thailand tussen 2015 en 2016. De geïnfecteerde en niet-geïnfecteerde vissen werden voornamelijk gegroepeerd op basis van fysieke diagnoses en de verschijningen van symptomatische TiLVD. Deel twee, RNA extractie. Na de incubatie gedurende 30 minuten, de korrel fluorescentie reactie suggereert de positieve versterking van TiLV in visweefsels, werden uiteraard waargenomen in geïnfecteerde monsters onder UV-licht op een golflengte van 365 nanometer, terwijl die van negatieve controle had een duidelijke en licht kleurloze oplossing.De versterking van RT-LAMP-producten werd overeenkomstig bevestigd op de agarose gel elektroforese, met verschillende maten van de meervoudige versterkingen van omgekeerde DNA-herhalingen, die stamlusstructuren onder UV-lamp vormen in met TiLV geïnfecteerde monsters, terwijl er geen banden werden weergegeven in de putten van de negatieve controle, waar de resultaten in overeenstemming waren met die van de RT-LAMP-test. Dus, om de verspreiding van tilapia lake virus te verminderen, een onmiddellijke screening van deze geïnfecteerde vissen, met behulp van een effectief diagnostische methode, is een van de belangrijke sleutels. De nieuw ontwikkelde RT-LAMP-test moet dus een snel en gevoelig diagnostisch instrument bieden dat moet worden toegepast tijdens het proces van bioveiligheidsmaatregel.De test kan worden gebruikt in het laboratorium, of faciliteiten ter plaatse. We hopen dat de methode zal worden uitgevoerd om besmette vispopulaties te screenen die kunnen leiden tot de vermindering van de verspreiding van het tilapiameervirus.
