L’industrie aquacole est continuellement menacée par des infections virales qui causent des pertes économiques importantes9,23,28. Par exemple, le TiLV émergent représente une menace majeure pour les pays producteurs de tilapia dans de nombreuses parties du monde1,6,9. Jusqu’à présent, il n’y avait pas de thérapeutique spé...

La maladie par le virus du lac de Tilapia (TiLVD) est une maladie virale dans le tilapia(Oreochromis spp.) qui causerait des décès de tilapia dans de nombreuses régions du monde, y compris l’Asie1,2, l’Afrique et l’Amérique. La maladie a été identifiée pour la première fois lors de la mortalité massive du tilapia en 2009 en Israel, où le nombre de tilapia sauvage dans le lac Kinneret a chuté de façon spectaculaire, passant de 257 à 8 tonnes par an2. La maladie est causée par le virus du lac de tilapia (TiLV), qui a été attribué à la famille Amnoonviridae comme un nouveau genre Tilapinevirus et une nouvelle espèce Tilapia tilapinevirus3. La caractérisation génétique de TiLV a montré que le virus est un nouveau, négatif-sens, virus de l’ARN à brin unique qui a 10 segments codant 10 protéines1,2,4. Diverses espèces de tilapia dans le genre Sarotherodon, Oreochromis, et Tilapine et d’autres poissons d’eau chaude (par exemple, gourami géant (Osphronemus goramy))ont été montrés pour être sensibles à TiLV2,5. Actuellement, ce virus continue de se propager à l’échelle mondiale, peut-être par le mouvement de poissons vivants infectés6,7, tandis que le risque de transmission virale via le tilapia congelé ou son produit est limité8. La mortalité importante due à l’infection tiLV a le potentiel d’avoir un impact économique considérablement préjudiciable sur l’industrie du tilapia. Par exemple, l’impact économique du syndrome de mortalité estivale en Égypte associé à l’infection par tiLV a été estimé à 100 millions de dollarsEU 9. Par conséquent, il est important de mettre au point une méthode diagnostique rapide et appropriée pour faciliter le contrôle de cette maladie dans les exploitations piscicoles.
Jusqu’à présent, le diagnostic de TiLVD a été basé sur les essais moléculaires, l’isolement viral, et l’histopathologie. Différents protocoles PCR et amorces ont été développés pour le diagnostic TiLV10,11. Par exemple, une méthode quantitative PCR (RT-QPCR) basée sur le vert SYBR avec la sensibilité de détecter aussi peu que deux copies/L du virus a été développée et validée pour la détection TiLV10. D’autres méthodes PCR pour la détection TiLV comprennent TaqMan quantitative PCR11, RT-PCR2, imbriqué RT-PCR12, et semi-niché RT-PCR13. Cependant, ces méthodes nécessitent un équipement de laboratoire sophistiqué et des périodes de temps relativement prolongées pour donner des résultats en raison de la complexité des réactions, ce qui les rend impropres à l’application sur le terrain.
L’essai d’amplification isothermale à médiation en boucle (LAMP) est une application rapide, simple et pratique pour le champ14,15. La technique utilise le principe d’une réaction de déplacement de brin, tandis que la réaction d’amplification fonctionne dans des conditions isothermales sans un cycleur thermique sophistiqué et coûteux14,15. Par conséquent, les produits LAMP amplifiés ou les produits RT-LAMP sont analysés dans des bandes en valeur d’échelle à l’aide d’électrophoresis gel agarose avec une tache fluorescente pour la visualisation sûre de l’ADN ou de l’ARN14 ou l’observation à l’œil nu pour la présence de turbidité ou d’un précipité blanc16,17,18. Pour ces raisons, cette technique a été utilisée pour la détection sur place de différents pathogènes du poisson17,18,19,20,21,22,23,24,25,26,27. Le but de cette étude était d’établir un essai rapide, sensible et précis de RT-LAMP pour la détection de TiLV. La maladie du virus du lac de Tilapia (TiLVD), une maladie virale émergente du tilapia causée par le virus du lac tilapia (TiLV), est un défi persistant dans l’industrie aquacole qui a entraîné la morbidité et la mortalité massives du tilapia dans de nombreuses régions du monde. Un essai diagnostique efficace, rapide et précis pour l’infection de TiLV est donc nécessaire pour détecter l’infection initiale et pour empêcher la propagation de la maladie dans l’agriculture aquacole. Dans cette étude, une méthode très sensible et pratique d’amplification isothermale (RT-LAMP) de transcription inverse très sensible et pratique est présentée pour détecter le virus du lac de tilapia dans les tissus du poisson. Une comparaison des essais RT-qPCR et RT-LAMP d’échantillons infectés a révélé des résultats positifs dans 63 (100%) et 51 (80,95%) échantillons, respectivement. En outre, une analyse des échantillons non infectés a montré que les 63 tissus non infectés ont donné des résultats négatifs pour les essais RT-qPCR et RT-LAMP. La réactivité croisée avec cinq agents pathogènes dans le tilapia a été évaluée à l’aide de RT-LAMP, et tous les tests ont montré des résultats négatifs. Les échantillons de foie et de mucus obtenus à partir de poissons infectés ont montré des résultats comparables en utilisant la méthode RT-LAMP, suggérant que le mucus peut être utilisé dans RT-LAMP comme un essai non létal pour éviter de tuer des poissons. En conclusion, les résultats ont démontré que l’essai RT-LAMP présenté fournit une méthode efficace pour la détection de TiLV dans le tissu de tilapia dans un rayon de 1 h. La méthode est donc recommandée comme outil de dépistage sur les fermes pour le diagnostic rapide de TiLV.

Après l’incubation pendant 30 minutes, la réaction de fluorescence de grain suggérant l’amplification positive de TiLV dans les tissus de poissons, ont été évidemment observées dans les échantillons infectés sous la lumière UV à une longueur d’onde de 365 nanomètres, alors que ceux du contrôle négatif ont eu une solution claire et légèrement incolore.En conséquence, l’amplification des produits RT-LAMP a été confirmée sur l’électrophoresis de gel d’agarose, montrant différentes tailles des amplifications multiples des répétitions inversées d’ADN, formant des structures de tige-boucle sous la lampe UV dans les échantillons TiLV-infectés, alors qu’aucune bandes n’a été montrée dans les puits du contrôle négatif, où les résultats étaient conformément à ceux de l’essai de RT-LAMP. Ainsi, pour réduire la propagation du virus du lac tilapia, un dépistage immédiat de ces poissons infectés, en utilisant une méthode de diagnostic efficace, est l’une des clés importantes. Ainsi, le nouvel essai RT-LAMP devrait offrir un outil de diagnostic rapide et sensible à appliquer dans le processus de mesure de biosécurité.L’essai pourrait être utilisé soit dans le laboratoire, soit sur place. Nous espérons que la méthode sera mise en œuvre pour dépister la population de poissons infectés qui pourrait conduire à la réduction de la propagation du virus du lac tilapia.
