Tilapia lake virus sykdom (TiLVD) er en virussykdom i tilapia (Oreochromis spp.) som angivelig forårsaker tilapia dødsfall i mange regioner av verden, inkludert Asia1,2, Afrika og Amerika. Sykdommen ble først anerkjent under massedødeligheten av tilapia i 2009 i Israel, hvor antall vill tilapia i Kinneretsjøen falt dramatisk fra 257 til 8 tonn per år2. Sykdommen er forårsaket av tilapia innsjø virus (TiLV), som har blitt tildelt familien Amnoonviridae som en ny slekt Tilapinevirus og en ny art Tilapia tilapinevirus3. Genetisk karakterisering av TiLV viste at viruset er en roman innhyllet, negativ-sense, enkelt-strandet RNA virus som har 10 segmenter koding 10 proteiner1,2,4. Foreløpig fortsetter dette viruset å spre seg globalt, muligens gjennom bevegelse av infisert levende fisk6,7, mens risikoen for viral overføring via frossen tilapia eller produktet er begrenset8. Betydelig dødelighet på grunn av TiLV-infeksjon har potensial til å ha en betydelig skadelig økonomisk innvirkning på tilapiaindustrien. For eksempel ble den økonomiske effekten av sommerdødelighetssyndrom i Egypt forbundet med TiLV-infeksjon beregnet til US $ 100 millioner9. Følgelig er det viktig å utvikle en rask og riktig diagnostisk metode for å lette kontrollen av denne sykdommen i oppdrettsanlegg.
Inntil nå har diagnosen TiLVD vært basert på molekylære analyser, viral isolasjon og histopatologi. Ulike PCR-protokoller og primere er utviklet for TiLV diagnose10,11. For eksempel, en SYBR grønn-basert omvendt transkripsjon kvantitativ PCR (RT-qPCR) metode med følsomhet for å oppdage så få som to kopier / μL av viruset er utviklet og validert for TiLV deteksjon10. Andre PCR-metoder for TiLV-deteksjon inkluderer TaqMan kvantitativ PCR11,RT-PCR2,nestet RT-PCR12og halvnestet RT-PCR13. Disse metodene krever imidlertid sofistikert laboratorieutstyr og relativt lengre perioder for å gi resultater på grunn av kompleksiteten i reaksjonene, noe som gjør dem uegnet for feltapplikasjon.
Den loop-medierte isothermal forsterkning (LAMP) analysen er en rask, enkel og praktisk for-feltet søknad14,15. Teknikken benytter prinsippet om en trådforskyvningsreaksjon, mens forsterkningsreaksjonen går under isothermale forhold uten en sofistikert og dyr termisk cycler14,15. Følgelig analyseres forsterkede LAMP-produkter eller RT-LAMP-produkter i stigelignende bånd ved hjelp av agarose gelelektroforese med en fluorescerende flekk for enten sikker visualisering av DNA eller RNA14 eller observasjon med det blotte øye for tilstedeværelse av turbiditet eller en hvit utfelling16,17,18. Av disse grunnene har denne teknikken blitt brukt til påvisning av ulike fiskepatogener17,18,19,20,21,22,23,24,25,26,27. Hensikten med denne studien var å etablere en rask, følsom og nøyaktig RT-LAMP-analyse for TiLV-deteksjon. RT-LAMP-analysen tilbyr screening for TiLV i fiskeprøver innen 30 min. reverse transcription loop-mediated isothermal amplification (rt-lamp) assay for the specific and rapid detection of tilapia lake virus

Tilapia lake virus sykdom (TiLVD), en fremvoksende virussykdom i tilapia forårsaket av tilapia innsjø virus (TiLV), er en vedvarende utfordring i akvakulturindustrien som har resultert i masse sykelighet og dødelighet av tilapia i mange deler av verden. En effektiv, rask og nøyaktig diagnostisk analyse for TiLV-infeksjon er derfor nødvendig for å oppdage den første infeksjonen og for å forhindre spredning av sykdommen i akvakulturoppdrett. I denne studien presenteres en svært følsom og praktisk omvendt transkripsjonloop-mediert isothermal forsterkning (RT-LAMP) metode for å oppdage tilapia innsjøvirus i fiskevev. En sammenligning av RT-qPCR- og RT-LAMP-analyser av infiserte prøver viste positive resultater i 63 (100%) og 51 (80,95 %) prøver, henholdsvis. Videre viste en analyse av uinfiserte prøver at alle 63 uinfiserte vev ga negative resultater for både RT-qPCR og RT-LAMP-analyser. Kryssreaktiviteten med fem patogener i tilapia ble evaluert ved hjelp av RT-LAMP, og alle testene viste negative resultater. Både lever- og slimprøvene hentet fra infisert fisk viste sammenlignbare resultater ved hjelp av RT-LAMP-metoden, noe som tyder på at slim kan brukes i RT-LAMP som en nonlethal analyse for å unngå å drepe fisk. Til slutt viste resultatene at den presenterte RT-LAMP-analysen gir en effektiv metode for TiLV-deteksjon i tilapiavev innen 1 h. Så, for å redusere spredningen av tilapia innsjøvirus, er en umiddelbar screening av disse infiserte fiskene, ved hjelp av en effektivt diagnostisk metode, en av de viktige nøklene. Så, den nyutviklede RT-LAMP analyse bør tilby en rask og følsom diagnostisk verktøy som skal brukes i prosessen med biosecurity tiltak.Analysen kan brukes enten i laboratoriet eller på stedet.
