La enfermedad por el virus del lago Tilapia (TiLVD) es una enfermedad viral en la tilapia (Oreochromis spp.) que al parecer causa muertes por tilapia en muchas regiones del mundo, incluyendo Asia1,2, Africa, y América. La enfermedad fue reconocida por primera vez durante la mortalidad masiva de tilapia en 2009 en Israel, donde el número de tilapia silvestre en el lago Kinneret se desplomó dramáticamente de 257 a 8 toneladas por año2. La enfermedad es causada por el virus del lago tilapia (TiLV), que ha sido asignado a la familia Amnoonviridae como un nuevo género Tilapinevirus y una nueva especie Tilapia tilapinevirus3. Por ejemplo, el impacto económico del síndrome de mortalidad de verano en Egipto asociado con la infección por TiLV se calculó en US$100 millones9. En consecuencia, es importante desarrollar un método de diagnóstico rápido y adecuado para facilitar el control de esta enfermedad en las piscifactorías.
Hasta ahora, el diagnóstico de TiLVD se ha basado en ensayos moleculares, aislamiento viral e histopatología. Se han desarrollado diferentes protocolos e imprimaciones pcR para el diagnóstico TiLV10,,11. Por ejemplo, se ha desarrollado y validado un método de PCR cuantitativo de transcripción inversa basado en verde SYBR (RT-qPCR) con la sensibilidad de detectar tan solo dos copias/L del virus para la deteccióntiLV 10. Otros métodos de PCR para la detección de TiLV incluyen TaqMan cuantitativoPCR 11, RT-PCR2, RT-PCR12anidado y RT-PCR13semianidado. Sin embargo, estos métodos requieren equipos de laboratorio sofisticados y períodos de tiempo relativamente prolongados para producir resultados debido a la complejidad de las reacciones, lo que los hace inadecuados para la aplicación en campo.
El ensayo de amplificación isotérmica (LAMP) mediado por bucle es una aplicación for-field rápida, sencilla y práctica14,15. La técnica emplea el principio de una reacción de desplazamiento de la hebra, mientras que la reacción de amplificación se ejecuta en condiciones isotérmicas sin un sofisticado y costoso ciclor térmico14,,15. En consecuencia, los productos LAMP amplificados o RT-LAMP se analizan en bandas tipo escalera utilizando electroforesis de gel de agarosa con una mancha fluorescente para la visualización segura del ADN o del ARN14 u observación a simple vista para la presencia de turbidez o un precipitado blanco16,,17,,18. Por estas razones, esta técnica se ha utilizado para la detección in situ de diferentes patógenos de peces17,,18,,19,,20,,21,,22,,23,,24,,25,,26,,27. El propósito de este estudio fue establecer un ensayo RT-LAMP rápido, sensible y preciso para la detección de TiLV. La enfermedad por el virus del lago Tilapia (TiLVD), una enfermedad viral emergente en la tilapia causada por el virus del lago tilapia (TiLV), es un desafío persistente en la industria de la acuicultura que ha dado lugar a la morbilidad masiva y mortalidad de tilapia en muchas partes del mundo. Por lo tanto, es necesario un ensayo diagnóstico eficaz, rápido y preciso para la infección por TiLV para detectar la infección inicial y prevenir la propagación de la enfermedad en la acuicultura. En este estudio, se presenta un método de amplificación isotérmica mediada por bucle de transcripción inversa (RT-LAMP) altamente sensible y práctico para detectar el virus del lago tilapia en el tejido de los peces. Una comparación de los ensayos RT-qPCR y RT-LAMP de muestras infectadas reveló resultados positivos en 63 (100%) y 51 (80,95%) muestras, respectivamente. Además, un análisis de muestras no infectadas mostró que los 63 tejidos no infectados arrojaron resultados negativos tanto para los ensayos RT-qPCR como para RT-LAMP. La reactividad cruzada con cinco patógenos en la tilapia se evaluó utilizando RT-LAMP, y todas las pruebas mostraron resultados negativos. Tanto las muestras de hígado como de moco obtenidas de peces infectados mostraron resultados comparables utilizando el método RT-LAMP, lo que sugiere que la mucosidad se puede utilizar en RT-LAMP como un ensayo no letal para evitar la matanza de peces.
