Kynurenin väg (KP) är den viktigaste vägen för tryptofan (Trp) katabolism i mänskliga celler. Indoleamin-2,3-dioxygenas (IDO-1) i extrahepatiska celler är det första och begränsande enzymet av KP och omvandlar Trp till N-formylkynurenin1. Ytterligare steg inom KP genererar andra sekundära metaboliter, nämligen kynureniner som uppvisar olika biologiska egenskaper. Kynurenin (Kyn) är den första stabila Trp-kataboliten som visar giftiga egenskaper och reglerar cellulära händelser efter bindning till arylkolvätereceptorn (AhR)2. Därefter omvandlas Kyn till flera molekyler antingen spontant eller i de enzymmedierade processerna, vilket genererar sådana metaboliter som 3-hydroxykynurenin (3HKyn), anthranilic syra (AA), 3-hydroxyanthranilic syra (3-HAA), kynurensyra (Kyna) och xanthurenic syra (XA). En annan nedströms metabolit, 2-amino-3-karboxymukansyra-6-semialdehyd (ACMS), genomgår icke-enzymatisk cyklisering till kinolinsyra (QA) eller picolinsyra (PA)1. Slutligen omvandlas QA ytterligare till nikotinamid-adenin dinukleotid (NAD+)3, KP-slutpunktmetaboliten som är en viktig enzymkofaktor. Vissa kynureniner har neuroprotektiva egenskaper som Kyna och PA, medan de andra, dvs. 3HAA och 3HKyn, är giftiga4. Xanthurenic syra, som bildas av 3HKyn, presenterar antioxidant och vasorelaxation egenskaper5. XA ackumuleras i åldrande linser och leder till apoptos av epitelceller6. KP, som beskrevs i mitten av 1900-talet, fick mer uppmärksamhet när dess engagemang i olika störningar visades. Ökad aktivitet av denna metaboliska väg och ackumulering av vissa kynureniner modulerar immunsvaret och är förknippade med olika patologiska tillstånd som depression, schizofreni, encefalopati, HIV, demens, amyotrofisk lateral skleros, malaria, Alzheimers, Huntingtons sjukdom och cancer4,7. Vissa förändringar i Trp metabolism observeras i tumör microenvironments och cancerceller2,8. Dessutom anses kynureniner vara lovande sjukdomsmarkörer9. Inom cancerforskningen är in vitro-cellkulturmodeller väletablerade och används ofta för preklinisk utvärdering av svar på cancerläkemedel10. Trp metaboliter utsöndras av celler i odlingsmediet och kan mätas för att bedöma statusen på kynureninvägen. Därför finns det ett behov av att utveckla lämpliga metoder för samtidig detektion av så många KP-metaboliter som möjligt i en mängd olika biologiska exemplar med ett enkelt, flexibelt och tillförlitligt protokoll.
I detta dokument beskriver vi ett protokoll för samtidig bestämning av fyra kynurenin utbildningsavsnitt metaboliter: Kyn, 3HKyn, 3HAA och XA av LC-SQ i ett postkulturellt medium samlas in från cancerceller. I ett modernt analytiskt tillvägagångssätt föredrasflytande kromatografi 13,14,15,16 för samtidig detektion och kvantifiering av de enskilda tryptofankataboliterna, i motsats till biokemiska ospecificerade analyser som använder Ehrlich reagens11,12. För närvarande finns det många metoder tillgängliga för kynureninbestämning i mänskliga exemplar, huvudsakligen baserade på flytande kromatografi med ultravioletta eller fluorescensdetektorer13,17,18,19. Flytande kromatografi i kombination med en masspektrometridetektor (LC-MS) verkar mer lämplig för denna typ av analys, på grund av deras högre känslighet (lägre detektionsgränser), selektivitet och repeterbarhet.
Trp metaboliter har redan fastställts i humant serum, plasma och urin13,20,21,22,23, men metoderna för andra biologiska exemplar, som cellkulturmedium önskas också. Tidigare användes LC- MS för Trp- härledda föreningar i ett medium som samlats in efter odling av mänskliga gliomceller, monocyter, dendritiska celler eller astrocyter behandlade med interferon gamma (IFN-γ)24,25,26. För närvarande finns det ett behov av nya validerade protokoll som kan möjliggöra en bedömning av flera metaboliter i olika odlingsmedier, celler och behandlingar som används i cancermodeller.
Syftet med den utvecklade metoden är att kvantifiera (inom en analytisk körning) fyra stora kynureniner som kan indikera avvikelser i KP. Presenteras här är kritiska steg i vårt nyligen publicerade protokoll för kvantitativ LC-SQ analys av utvalda meningsfulla kynureniner med hjälp av en intern standard (3-nitrotyrosin, 3NT) i mediet som samlats in från in vitro odlade mänskliga cancerceller27. Så väl som vi vet är det det första LC-SQ-protokollet för samtidig kvantifiering av 3HKyn, 3HAA, Kyn och XA i ett odlingsmedium som erhållits från de in vitro-odlade cellerna. Vid vissa modifieringar kan metoden tillämpas ytterligare för att studera förändringarna i Trp-metabolismen i ett bredare spektrum av cellkulturmodeller.

simultaneous quantification of selected kynurenines analyzed by liquid chromatography-mass spectrometry in medium collected from cancer cell cultures

Kynureninvägen och tryptofankataboliterna som kallas kynureniner har fått ökad uppmärksamhet för sitt engagemang i immunreglering och cancerbiologi. En in vitro-cellkulturanalys används ofta för att lära sig om bidraget från olika tryptofankataboliter i en sjukdomsmekanism och för att testa terapeutiska strategier. Cellkulturmedium som är rikt på utsöndrade metaboliter och signalmolekyler återspeglar statusen för tryptofanmetabolism och andra cellulära händelser. Nya protokoll för tillförlitlig kvantifiering av flera kynureniner i det komplexa cellkulturmediet önskas möjliggöra en tillförlitlig och snabb analys av flera prover. Detta kan åstadkommas med flytande kromatografi i kombination med masspektrometri. Denna kraftfulla teknik används i många kliniska och forskningslaboratorier för kvantifiering av metaboliter och kan användas för att mäta kynureniner.
Presenteras här är användning av flytande kromatografi i kombination med en fyrdubbel massa spektrometri (LC-SQ) för samtidig bestämning av fyra kynureniner, dvs kynurenin, 3-hydroxykynurenin, 3-hydroxyanthranilic och xanthurenic syra i mediet samlas in från in vitro odlade cancerceller. SQ-detektorn är enkel att använda och billigare jämfört med andra masspektrometrar. I SQ-MS-analysen genereras och separeras flera joner från provet enligt deras specifika mass-till-laddningsförhållande (m / z), följt av detektion med hjälp av ett SIM-läge (Single Ion Monitoring).
Detta dokument uppmärksammar fördelarna med den rapporterade metoden och indikerar några svaga punkter. Det är inriktat på kritiska element i LC-SQ-analys inklusive provberedning tillsammans med kromatografi och masspektrometrianalys. Kvalitetskontroll, metodkalibreringsförhållanden och matriseffektfrågor diskuteras också. Vi beskrev en enkel tillämpning av 3-nitrotyrosin som en analog standard för alla mål analyter. Som bekräftas av experiment med mänskliga äggstocks- och bröstcancerceller genererar den föreslagna LC-SQ-metoden tillförlitliga resultat och kan tillämpas ytterligare på andra cellulära in vitro-modeller.

LC-MS uppvisar obestridliga fördelar vid kvantifiering av biologiskt aktiva molekyler, även om vissa komponenter i komplexa exemplar orsakar så kallade matriseffekter och kompromissjonisering av analyter. Det leder till jondämpning eller jonförbättring, vilket kraftigt minskar noggrannheten och påverkar en gräns för detektion/kvantifiering av LC-MS, vilket anses vara en "svag" punkt i metoden.
