1. 人类 iPSC 线路解冻和维护

	<li>第2阶段：悬架造血细胞的出现

<li>第4天，涂4口6井组织培养板的0.1%v明胶，孵育至少10分钟。</li>
		<li>去除明胶，加入2.5 mL的阶段2介质（X-VIVO15辅以100纳克/mL CSF1，25 ng/mL IL-3，2 mM谷胱甘肽，1%青霉素-链霉素，和0.055 mM 2-麦卡普托乙醇）。</li>
		<li>将形成的EB收集到50mL离心管中，使其通过重力在管底沉降。小心地吸气媒体。</li>

	<li>第 3 阶段：终端巨噬体成熟

	<li>巨噬体极化
	<ol>
<li>要激活M（LPS + IFNg）表型的巨噬细胞，用LPS（最终浓度：100纳克/mL）和IFNg（最终浓度：10 U/mL）刺激细胞48小时。要激活细胞到M（IL4）表型，用IL4刺激细胞（最终浓度：20纳克/mL）。要激活到 M（IL10） 表型，使用 IL10 刺激巨噬细胞（最终浓度：5 纳克/mL）。</li>

</ol>4. iPSC 衍生的巨噬细胞质量控制检查
<ol><li>使用造血细胞计计数每6孔EBs中产生的造血悬浮细胞数量。</li>
	<li>评估大噬菌体形态（例如，商业套件染色）8，,9.</li>
	<li>
使用基因表达分析和流细胞测定检测巨噬细胞特异性标记和偏振标记的表达，如前所述的8、99。

<li>在6孔大噬细胞的一口井上进行流细胞测定实验，通过吸气其成熟介质来收获细胞，用2 mL的PBS清洗，并在室温（RT）下用2 mL的酶自由细胞分离缓冲液孵育5分钟。移液反复分离和收获巨噬细胞。</li>
		<li>用造血计计数细胞，并在80μL中重新悬浮在2%BSA、0.5M乙烯二甲酸（EDTA）-PBS溶液中。</li>

</ol>5. 高通量噬菌体测定

	<li>板 8 x 10 4 iPSC-DM 在成像组织培养等级 96 井板（例如，细胞载体 Ultra，Perkin Elmer）的孔中至少 2 天，在 200 μL 的阶段 3 介质的高通量成像之前。4</li>
	<li>通过在 2 mL PBS 中重新悬挂一个小瓶（"解决方案 1"）来制备 pHrodoGreen Zymosan-A 生物颗粒。涡旋溶液为10 s。</li>

总之，我们提出了用于生产功能性 iPSC 衍生巨噬细胞的详细协议。与单核细胞衍生巨噬细胞（MdM）的实验，利用iPSC衍生的巨噬细胞进行体外实验，研究健康与疾病中的巨噬细胞生物学具有许多优势。这些优点包括易于获得材料（例如，不需要捐赠者），可以产生大量巨噬细胞，而且产生转基因巨噬细胞是可行的，而且相对简单。此外，iPSC衍生的巨噬细胞可能是研究组织-居民巨噬细胞生物学的更好资源。

production and characterization of human macrophages from pluripotent stem cells

造血悬浮细胞的前2周收获（第16~28天）的一个完整的6孔板的EBs平均含有2.59 x 106 = 0.54细胞。第28天之后，平均每6口液的液位板产生4.64 x 106 ± 0.94 的悬浮细胞。从第80天开始，悬浮细胞的数量开始下降，因为EB变得耗尽（图2C）。建议在每6口EB的收获量低于3 x 106前体后停止分化协议。
IPSC派生的巨噬细胞表面标记表达式 流细胞学仍然是用于评估人类巨噬细胞表的最常见的方法。评估细胞表面标记表达式的浇注策略包括使用物理参数（如大小和粒度）对单元格的主要组门，然后对单个单元格进行门控，然后对活单元格进行浇注（图 3A）。成熟的iPSC衍生巨噬细胞应表示谱系标记CD45和巨噬细胞成熟标记25F9，对单核细胞/未成熟的巨噬细胞标记CD93为负数。这与我们的观测结果（图3A）一致。IPSC衍生的巨噬细胞对系系骨髓标记CD11b、CD14、CD43、CD64、CD115、CD163和CD169（图3B）也呈阳性。它们对免疫调节标记CD86呈阳性，其中一小部分在幼化状态下表示化学素受体CX3CR1、CCR2、CCR5和CCR8（图3B）。这些地块是从我们的实验室8先前公布的数据中获得的。

巨噬细胞可以响应和改变其表型，具体取决于环境线索。为了评估它们对环境刺激的反应和变化的能力，可以使用 LPS 和 IFNg、IL4 或 IL10 处理 iPSC-DM。经过48小时的治疗，他们改变了表型，这里称为M（LPS + IFNg），M（IL4）和M（IL10），分别29。基因表达分析是检验巨噬细胞极化状态的有用工具。在LPS和IFNg刺激下，巨噬细胞对CD40、VCAM1VCAM1和TNFA基因的mrRNA表达进行调节（图5A）。在IL4刺激下，细胞对CD68、CD84和MRC1基因的mRNA表达进行调节（图5B）。 CD84在IL10刺激下，iPSC-DM对MRC1的调节表达（图5B）。在噬菌体方面，与天真的巨噬细胞相比，用LPS+IFNg或IL4治疗的巨噬细胞的百分比明显较低。使用IL10治疗的iPSC-DM显示噬菌细胞和噬菌体指数的百分比增加（图5C+E）。E
