</ol>4. iPSC 유래 대식세포 품질 관리 검사

production and characterization of human macrophages from pluripotent stem cells

분화 진행, 대식세포 수 및 형태학 제시된 결과는 SFCi55 인간 iPSC 라인의 분화로부터 기술되고 다수의 연구에서88,9,10,,26을기술하고 사용한다.9 대식세포로 IPSC 분화의 과정은 광학 현미경 검사법에 의해 감시될 수 있었습니다. iPSC 콜로니, 배아체(EBs), 조혈 현탁액 세포 및 성숙한 대식세포는 형태학적으로 구별되었다(도2A). 성숙한 대식세포 형태는 원심 분리된 사이토스핀 제제의 염색에 의해 추가로 검증될 수 있었다. IPSC 유래 대식세포는 크고, 많은 소포를 함유하는 단일 작은 타원형 핵 및 풍부한세포질(도 2B)을가졌다.
조혈 현탁액 세포의 처음 2주(일 16-28일)는 평균 2.59 x 10 6±0.54 세포를6 함유한 1개의 전체 6개의 웰 플레이트를 함유하였다. 28일 이후, 평균 4.64 x6 10 6±0.94의 현탁액 세포가 6개의 웰 플레이트당 EBs를 생성하였다. 80일째부터, 현탁액 세포의 수는 EB가 고갈됨에 따라 떨어지기시작했다(그림 2C). EB의 6 웰 플레이트 당 수확 당 3 x 106 전구체 이하로 숫자가 떨어지면 분화 프로토콜을 중지하는 것이 좋습니다.
