따라서, AFM의 주요 원리는 AFM 프로브와 샘플의 표적 구조 사이에서 작용하는 힘의 검출이다. 얻어진 힘 곡선은 탄성, 전하 분포, 자화, 항복 응력 및 탄성 소성 변형 역학8과같이 샘플 표면에 대상 구조의 기계적 특성을 설명합니다. 다른 이미징 기술에 비해 AFM의 중요한 장점은 AFM이 조직을 손상시키지 않고 기본 상태에서 중간 또는 조직에서 살아있는 세포의 기계적 특성을 측정하는 데 사용될 수 있다는 것입니다. AFM은 액체 또는 건조 조건에서 모두 작동 할 수 있습니다. 샘플 준비에 대한 요구 사항은 없습니다. AFM을 사용하면 시료를 손상시키지 않고 고해상도, 신뢰성 및 감도로 셀룰러 수준에서 측정을 수집할 수 있습니다.생체 역학적 특성 변화는 골관절염 초기에 발생합니다. 관절 연골의 국부적 탄력성을 측정하면 국부 조직 변성의 정도에 대한 유효한 결론을 도출할 수 있습니다. 인간 무릎 관절 내의 퇴행성 변화를 평가하기 위해 먼저 메스를 사용하여 관절 연골을 아천드콘드랄 뼈에서 전체적으로 자르고 극저온 장치에서 저온하에서 동결되는 극저온 노브에 수용성 임베딩 배지에 연골 샘플을 포함시한다.시료가 동결되면 표준 Cryotome을 사용하여 관절 연골의 맨 위 층에서 조직의 35 마이크로 미터 두께의 섹션을 획득하여 각 섹션을 개별 유리 슬라이드에 수집합니다. 모든 단면을 수득한 경우, 세척당 5분간 신선한 PBS에서 시료를 3회 헹구어 수용성 포함 매체를 제거한다. 세척에서 첫 번째 슬라이드를 제거합니다.그 후 섹션당 2~3방울의 생체 적합성 샘플 접착제를 개별 AFM 호환 페트리 접시에 넣습니다. 샘플에서 여분의 용액을 제거하고 건조 된 섹션의 가장자리를 접착제 반점에 부드럽게 누릅니다. 2 분 후, L-글루타민없이 Leibovitz의 L-15 배지와 결합 된 조직을 신중하게 커버하고 샘플을 분석 할 때까지 세포 배양 인큐베이터에 접시를 배치합니다.AFM 장치를 준비하려면, 상부 표면이 홀더와 평행하다는 AFM 홀더의 액체 환경에서 측정하기 위한 가스 블록을 조정하고, 핀셋을 사용하여 선택한 캔틸레버를 유리 블록 표면에 신중하게 장착하여 미세구가 연마된 광학 평면위에 돌출되도록 한다. 유리 블록의 캔틸레버를 안정화하려면 금속 스프링을 블록의 홈으로 밀어 넣고 핀셋을 사용하여 스프링과 함께 캔틸레버의 상단을 고정시하십시오. 조심스럽게 AFM 헤드에 캔틸레버와 유리 블록을 배치하고 통합 잠금 메커니즘으로 블록을 확보.그런 다음 캔틸레버를 사용하여 AFM 헤드를 AFM 장치에 장착하고 스프링이 왼쪽으로 향합니다. 샘플을 AFM 장치에 장착하려면 샘플 페트리 접시를 AFM 샘플 홀더에 놓고 페트리 접시 히터를 섭씨 37도로 설정합니다. 약 20분 후 장치 소프트웨어와 레이저 정렬 및 접근 매개 변수 설정 창을 엽니다.다음으로 스테퍼 모터, 레이저 라이트 및 CCD 카메라를 켜고 스테퍼 모터 기능을 사용하여 캔틸레버가 매체에 완전히 잠길 때까지 100 마이크로미터 단계로 캔틸레버를 낮춥니다. 조정 나사를 사용하여 캔틸레버 위에 레이저를 지시하고 AFM 장치 나사를 사용하여 레이저 빔을 조정하여 반사된 빔이 광검출기의 중앙에 떨어지도록 합니다. 레이저 캔틸레버가 조정되면 필요에 따라 광검출기를 조정하여 합계 신호를 1볼트 이상으로 설정하고 측면 및 수직 편향을 0에 가깝게 설정합니다.교정 힘 곡선을 얻으려면 테이블에 표시된 대로 접근 매개 변수로 스캐너 접근 방식을 실행합니다. 조직 배양 접시의 바닥에 도달하면 캔틸레버를 100 마이크로미터로 철회하십시오. 테이블에 표시된 대로 실행 매개 변수를 설정합니다.그리고 실행을 클릭하여 측정을 시작하고 교정 힘 거리 곡선을 얻습니다. 힘 곡선은 그림에 도시된 대로 얻어진다. 교정 힘 거리 곡선에서 리포트된 곡선의 선형 핏을 위해 영역을 선택합니다.선형 맞춤이 완료되면 소프트웨어에서 값을 저장합니다. 그런 다음 측정이 섭씨 37도에서 중간에서 수행됨에 따라 소프트웨어의 온도 변수를 가능한 한 가깝게 생리적 조건을 모방하도록 37도로 설정합니다. 연골 샘플 섹션에서 수렴성 패턴을 식별하기 위해, 건강한 조직 영역을 나타내는 단일 문자열, 조직 변성의 시작을 나타내는 이중 문자열, 조직 변성의 시작을 나타내는 이중 문자열, 고급 조직 변성의 징후, 및 조직 파괴의 끝을 나타내는 작은 클러스터를 포함 할 수있는 위상 대비 현미경에 골관절염 무릎에서 관절 연골의 특정 세포 패턴을 찾아 식별합니다.특정 원하는 패턴이 확인되면, 백세포 매트릭스 측정을 위해 캔틸레버를 세포에 근접하게 배치하고 매트릭스 유형당 선택한 패턴당 2개의 부위를 측정하고, 가능한 부정확성을 고려할 수 있을 만큼 큰 샘플 크기를 포함하여 측정 부위당 9회 측정한다. 측정할 패턴의 셀룰러 매트릭스에 초점을 맞추고 해당 시점에서 컴퓨터 마우스를 수정합니다. 다음으로 프로브에 초점을 맞추고 프로브 끝을 컴퓨터 화살표로 수정한 지점으로 이동합니다.세포 외 매트릭스의 측정을 수행하려면 세포가없는 영역을 선택하고 캔틸레버가 조직 위에 100 마이크로 미터를 배치되도록 철회 다음 접근 방식을 수행합니다. 그런 다음 캔틸레버교정에 의해 얻은 세트포인트 매개변수를 사용하여 실행을 클릭하여 측정을 시작합니다. 획득한 데이터를 처리하려면 AFM 장치에서 얻은 데이터와 호환되는 데이터 처리 소프트웨어를 열고 힘 거리 곡선을 처리하기 위한 소프트웨어에서 허스트 모델을 선택합니다.푸아송 비율을 0.5로 설정하고 팁 모양을 구형으로 설정하고 팁 반지름을 12.5 마이크로미터로 설정합니다. 모든 매개 변수가 조정되면 결과가 장착되고 영의 Modulus는 소프트웨어에 의해 계산됩니다. 문자열에서 이중 문자열에 이르기까지 생리학적 모델을 따라 작은 클러스터에서 큰 클러스터까지, 세포 외 및 세포 매트릭스 탄성 계골 계열은 문자열과 이중 문자열 사이를 제외하고 각 패턴 변화 사이에 크게 감소합니다.또한, 세포외 세포 외 매트릭스 비율은 크게 변하지 않는 반면 세포외 및 세포 간 행렬 사이의 탄성에 있는 절대적인 차이의 현저한 감소가 관찰된다. 탄력 값은 들여쓰기 깊이 또는 캔틸레버 팁 속성과 같은 다양한 조건에 따라 달라집니다. 따라서 AFM은 동일한 실험 설정 내에서 다른 조건을 비교하는 데 가장 적합합니다.AFM이 시료의 로컬 탄성을 측정하기 때문에 정확한 측정을 허용하고 캔틸레버 손상을 방지하기 위해 섹션을 제자리에 고정해야 합니다. 조직 탄력 변화에 대한 책임을 지는 공정을 더욱 분석하기 위해 관심 있는 단백질 구조의 생화학적 정량화는 서양 얼룩 또는 ELISA를 통해 수행될 수 있다.
