ここでは、新開発のECLIAシステムを用いて、ヒトおよびマウス由来のサンプルおよび組換えタンパク質の異なる変異体におけるメチルCpG結合タンパク質2(MeCP2)レベルを分析した。MecP2は、メチル化DNA配列と相互作用することが知られているX結合型核酸結合タンパク質である。このタンパク質は、遺伝子発現44,55の調節に関与している。このタンパク質をコードする遺伝子の機能喪失変異は、Tett症候群(RTT)を引き起こす主な原因である、重度の神経発達障害6である。別のMeCP2関連障害、MECP2重複症候群は、RTT 7のものと重なり合う神経学的症状にもつながりる。特に、女性は主にRTTの影響を受け、男性は主にMECP2複製症候群66、77に苦しんでいます。
これらの障害は、中枢神経系(CNS)においてそれぞれ不十分または過剰なMeCP2レベルに関連している。したがって、CNSでMeCP2レベルを増加させるRTTの治療オプションは、MeCP27の過剰に関連する有害な影響を避ける必要があります。この事実により、ECLIAシステムによって提供されるMeCP2タンパク質レベルの高感度かつ正確な定量化は、RTTの進歩とMECP2重複症候群の研究にとって非常に重要です。ヒト細胞株およびマウス組織サンプルからの内因性および外因性MeCP2レベルの正確な測定と、ヒトMeCP2アイソフォームB(アイソフォームe1とも呼ばれる)からなる組換えタンパク質、および血液脳関門8を横断する可能性を有する最小N末端HIV-TAT伝達ドメイン(TAT-MeCP2)が提示される。

ECLIAは、MeCP2の細胞レベルを定量化するシンプルで効率的な方法です。ECLIAは、ウェスタンブロットやELISAなどの他の定量方法と比較して、高速で、より敏感で、扱いやすくなります。PBSで0.5%Tween 20である洗浄液を調製するには、500マイクロリットルのTween 20をPBSの1リットルに加え、激しく混合する。3%ブロッカーAとPBSのブロッキング溶液を調製し、穏やかにかき混ぜます。ろ過によりブロッキング溶液を殺菌する。アッセイ希釈液を調製するために、1%ブロッカーA、およびPBS、10ミリリットルのPBSに5ミリリットルのブロッキング溶液を加える。次に、モノクローナルマウスの抗MeCP2抗体を氷上で解凍する。0.67マイクロリットルの抗体を4ミリリットルのPBSと混合し、溶液をボルテックスします。96ウェル高結合プレートを溶解するには、マルチチャネルピペッタを使用して、各ウェルの下隅に25マイクロリットルのコーティング溶液を慎重に分配します。コーティング溶液が各ウェルの底部を覆うことを確認するために、両側に穏やかに96ウェルプレートをダブ。粘着ホイルでプレートを密封し、摂氏4度で一晩インキュベートします。冷蔵庫から96ウェルプレートを取り出し、ホイルを取り外します。廃容器にフリックして、ウェル内の抗体コーティング溶液を除去します。その後、ペーパータオルのプレートをタップして、残りの溶液を取り除きます。次に、各ウェルに125マイクロリットルのブロッキング溶液を加えます。その後、粘着ホイルでプレートを再び密封します。プレートを軌道マイクロプレートシェーカーに置き、800 RPMに設定し、室温で90分間インキュベートします。MeCP2またはTat-MeCP2タンパク質ストック溶液、マウス脳溶解物、およびHDF溶解物の氷上の1バイアルで解凍する。原稿の表2に記載されているように、きれいなチューブでタンパク質ストック溶液を希釈します。次に、ライセートサンプルを希釈します。マウスの脳のライゼの場合は、25マイクロリットルのライシスバッファーあたり1〜20マイクログラムを使用してください。HDFライセートの場合、25マイクロリットルのライシスバッファーあたり1マイクログラムに0.25を加えます。96ウェルプレートを90分間インキュベートした後、廃棄物容器にフリックして、ブロッキング溶液をウェルから取り除きます。その後、ペーパータオルのプレートをタップして、残りの溶液を取り除きます。次いで、150マイクロリットルの洗浄液でプレートを3回洗浄し、洗浄液を添加し、直ちに除去する。96ウェルプレートの井戸に25マイクロリットルの標準とサンプルを加えます。プレートを密封し、一定の800 RPMの揺れで室温でさらに4時間インキュベートします。非標識検出抗体、ポリクローナルウサギ抗MeCP2を氷上で解凍します。アッセイ希釈液を用いて、抗体を1~6,000の割合で希釈する。96ウェルプレートがシェーカーでのインキュベートを終了したら、プレートを廃棄物容器にフリックして標準とサンプルを取り除きます。その後、ペーパータオルのプレートをタップして、残りの溶液を取り除きます。洗浄液を加えてすぐに取り除いて、150マイクロリットルの洗浄液でプレートを3回洗います。マルチチャネル・ピペッタを用いて、25マイクロリットルの未標識検出抗体溶液を各ウェルに加えます。プレートを密封し、800 RPMで一定の揺れで室温で1時間インキュベートします。冷蔵庫から特異的な二次抗体を入手し、1つの氷を置きます。二次体をアッセイ希釈液で希釈し、軽く混ぜます。96ウェルプレートから無ラベル検出抗体を除去するには、廃棄物容器にフリックし、ペーパータオルのプレートをタップします。プレートを浸透して3回洗浄した後、マルチチャネルピペッタを用いて各ウェルに25マイクロリットルの二次抗体を加える。プレートを密封し、800 RPMで一定の揺れで室温で1時間インキュベートします。廃容器にフリックしてペーパータオルのプレートをタップし、前述のようにプレートを3回洗浄して、無料の二次抗体を取り除きます。プレートの各ウェルに、1X Trisベースゴールドリードバッファーの150マイクロリットルを界面活性剤付き、光発生用のコアラクタントとして含みます。気泡を発生させないために、逆ピペット法を使用してください。96ウェルプレートを電気化学発光検出システムを備えたマイクロプレートプラットフォームに配置します。内蔵のCCDカメラを使用して、すぐにデータのキャプチャを開始します。相対光単位に対応し、光の強度に直接比例する記録信号数。MeCP2標準曲線は、ヒトMeCP2から複数の測定で生成された。ECLIAは、1ミリリットル当たり1ナノグラムから1ミリリットル当たり1800ナノグラムまでの範囲で、組み換えヒトMeCP2を正確に定量することができます。ECLIAを用いて、MeCP2タンパク質レベルをヘテロ接合マウスおよび雌野生型マウスおよび1つのMeCP2ノックアウトマウスから脳核ライセートで測定した。ECLIAは、健康なコントロールから、およびレット症候群のモデルとして使用されるc.806delG細胞株から細胞ライセートに行われた。ECLIAまたは免疫蛍光によって変異細胞株中でMeCP2タンパク質が検出されなかった。ECLIAは、MeCP2欠損セルライン残業によるTat-MeCP2の取り込み調査にも使用されました。ECLIAインターアッセイとイントラアッセイ精度は3日間連続して決定した。将来のタンパク質置換療法のために、MeCP2の送達はECLIAによるタンパク質レベル評価に続いて繰り返し最適化することができる。
