Dit protocol biedt een methode voor succesvolle immunolabeling van volwassen Drosophila-teelballen. Zoals gemeld in andere procedures moeten jonge mannetjes worden gebruikt (we hebben zelfs de leeftijd verkort tot minder dan 20 uur na eclosie) om de aanwezigheid van zich ontwikkelend sperma te minimaliseren, wat de spermatocyten dreigt te belemmeren. De dissectie moet snel zijn en de antilichaamverdunning moet worden aangepast om de signaal-ruisverhouding21,22<su...

Drosophila-teelballen zijn een waardevol model voor de studie van nucleaire architectuur. In mitotische spermatogoniale cellen wordt het nucleaire volume grotendeels ingenomen door chromatine. Deze cellen ondergaan vier delingsrondes en produceren een cyste van 16 primaire spermatocyten. In de komende dagen passeren de spermatocyten 6 ontwikkelingsstadia (S1-S6), sterk toenemend in volume, ongeveer 20-voudig1,2. Ze veranderen ook hun nucleaire morfologie. De omtrek van de kern, onthuld door de lamin Dm0, wordt onregelmatig en vertoont diepe invaginaties1,3. Dit gaat gepaard met uitgebreide genexpressieveranderingen en wijzigingen in chromatineorganisatie1,4,5,6,7. De interfasechromosomen zijn goed gedefinieerd in stadium S4-S5 en vormen drie verschillende massa's die overeenkomen met de 2 grote bivalente autosomen en het X-Y-paar aangemeerd met de nucleolus.
Mad1 werd oorspronkelijk geïsoleerd als een belangrijk onderdeel van het mitotische controlepunt (beoordeeld in8). In interfase wordt beschreven als voornamelijk gelegen aan de nucleaire envelop, maar ook in het nucleoplasma9,10,11. In Drosophila testis meldden we dat Mad1 prominent aanwezig is in het nucleoplasma, nauw verbonden met de twee belangrijkste autosomale chromatinemassa's en in mindere mate met de XY chromatine. We definieerden deze chromatine-geassocieerde structuur als MINT (Mad1-containing IntraNuclear Territory)12. Vier andere eiwitten werden geassocieerd met MINT's in spermatocyten: de nucleoporine Mtor/Tpr, het SUMO peptidase Ulp1, het mitotische checkpoint-eiwit Mad2 en een subeenheid van een Polycomb-achtig complex Raf212.
Om de beschrijving van DE's te voltooien, moesten we antilichamen gebruiken en werden we geconfronteerd met de technische problemen die over het algemeen worden ondervonden met immunostaining bij testis: een slechte permeabiliteit die resulteert in een significante niet-uniformiteit van kleuring in de diepte van de testis, met name in de grote spermatocyten. Svernietigde preparaten verhoogden de toegang tot antilichamen, maar leidden tot gecomprimeerde beelden, waardoor de 3D-analyses werden beperkt en het meten van kwantitatieve fluorescentie, waaronder colocalisatie, werdvoorkomen. Het probleem van antilichaampenetratie kan ook worden aangepakt door permeabiliserende middelen zoals 0,3% Na-deoxycholaat13, maar deze procedure leverde in onze handen geen reproduceerbare resultaten op. Omdat we veel co-labeling experimenten hebben uitgevoerd, hebben we gekeken naar het verbeteren van het permeabilisatieprotocol. De combinatie van 1% NP40 plus heptaan resulteerde in meer reproduceerbaarheid, met name in het bestuderen van de grote spermatocyten.
Dit protocol voert fixatie en immunostaining van teelballen in suspensie uit, verbetert de monsterkwaliteit en verbetert de reproduceerbaarheid en maakt het geschikt voor confocale microscopie. We gebruikten het protocol om de ruimtelijke relaties van bepaalde nucleaire envelopeiwitten met de nucleoplasmatische structuren van grote spermatocyten beter te definiëren. Deze methode zal een beter onderzoek mogelijk maken naar de veranderingen die gepaard gaan met de ontwikkeling van spermatocyt, bijvoorbeeld de dynamische structurele veranderingen van de kern, waaronder chromatine, nucleoplasma en de nucleaire poriën.

Drosophila-teelballen zijn een krachtig modelsysteem voor het bestuderen van biologische processen, waaronder stamcelbiologie, nucleaire architectuur, meiose en spermaontwikkeling. Immunolabeling van de hele Drosophila-testis wordt echter vaak geassocieerd met significante niet-uniformiteit van kleuring als gevolg van antilichaampenetratie. Geplette preparaten overwinnen het probleem slechts gedeeltelijk, omdat het de 3D-kwaliteit van de analyses vermindert. Hierin beschrijven we een whole-mount protocol met NP40 en heptaan tijdens fixatie samen met immunolabeling in vloeibare media. Het behoudt het volume dat geschikt is voor confocale microscopie samen met reproduceerbare en betrouwbare etikettering. We laten verschillende voorbeelden zien van 3D reconstitutie van spermatocytkernen uit confocale secties. De reproduceerbaarheid van intra- en intertesten maakt 3D-kwantificering en vergelijking van fluorescentie tussen afzonderlijke cellen van verschillende genotypen mogelijk. We gebruikten verschillende componenten van de intranucleaire MINT-structuur (Mad1-bevattend Intra Nuclear Territory) en twee componenten die verband houden met het nucleaire poriecomplex om dit protocol en de toepassingen ervan op de grootste cellen van de testis, de S4-S5 spermatocyten, te illustreren.

Het belangrijkste obstakel voor het verkrijgen van adequate kleuring van Drosophila-teelballen is de beperkte penetrante doordringbaarheid van antilichamen, die gedeeltelijk is opgelost door het gebruik van geplette preparaten, maar ten koste van het induceren van een beperking van de 3D-analyses (bijvoorbeeld 3D-representatie of meting van colocalisatie). We presenteren hier een verbeterd protocol voor de hele montering van drosophila-teelballen, geschikt voor confocale microscopie en ook reproduceerbaar en betrouwbaar labelen mogelijk.
