Bij utero-elektroporatie (IUE) van MAGIC Markers in corticale voorlopers (figuur 1) maakte etikettering van astrocyten in de postnatale hersenschors in verschillende postnatale stadia mogelijk (P4-P7-P21, figuur 2). Interessant is dat het stadium van IUE niet kritiek is, omdat elektroporatie uitgevoerd van E13,5 tot E15,5 vergelijkbare etiketteringspatronen oplevert met betrekking tot corticale astrocyten14. Astrocyten spelen talrijke vitale functies in de ontwikkeling en fysiologie van de hersenen1. Naast hun rol bij de bloed-hersenbarrière waar ze de opname van voedingsstoffen en de bloedstroom reguleren, dragen ze actief bij aan synapsvorming en -functie terwijl ze neuromodulatoren produceren die neuronale activiteit en gedrag kunnen veranderen2. Bovendien draagt astrocytendisfunctie bij aan een verscheidenheid aan neurologische aandoeningen3. Astrocyten in de hersenschors vertonen een uitgebreide morfologie die uitgebreid contact met neuronale processen mogelijk maakt. Deze contacten, essentieel voor circuitfunctie, regelen ook astrocytenmorfogenese en synaptogenese door celhechtingseiwitten4. Neurowetenschappers hebben handige en robuuste tools nodig om de ontwikkeling van astrocyten en morfogenese in hun neurologische interessemodellen te onderzoeken. Vanwege de nauwe aanleg van astrocyten bij hun buren en hun uniforme driedimensionale tegels, is het echter een uitdaging om corticale astrocyten uit te kiezen en hun morfologie uitgebreid te beoordelen met behulp van immunomarkers.
Momenteel maken twee belangrijke genetische manipulatiestrategieën etikettering en individualisering van corticale astrocyten in situ mogelijk: schaarse reporteractivering in transgene muislijnen of somatische transgenese met behulp van elektroporatie van reporter plasmiden. De eerste strategie is gebaseerd op het fokken van een floxed reporter muislijn met muizen die een induceerbare vorm van Cre recombinase uitdrukken die specifiek bij astrocyten wordt geactiveerd bij de levering van tamoxifen (bijv. Aldh1l1-CreERT25). Aan deze strategie zijn verschillende nadelen verbonden. Ten eerste vereist het fokken van transgene muizen een groot aantal dieren en meerdere tests zijn meestal nodig om de juiste dosis tamoxifen te bepalen om voldoende schaarse etikettering van corticale astrocyten te bieden. Het analyseren van corticale astrocyten fenotypen in een genetisch muismodel van belang vereist nog meer fokkerij en muisconsumptie. Bovendien is bekend dat bij utero tamoxifen-injectie de bevalling interfereert, waardoor deze strategie moeilijk toe te passen is op de studie van de vroegste stadia van de ontwikkeling van astrocyten. In vivo DNA-elektroporatie is een alternatieve tamoxifenvrije strategie die afhankelijk is van een minimumaantal dieren6. Deze aanpak wordt uitgevoerd in embryonale of postnatale stadia en bestaat uit het injecteren van reporter plasmiden in de laterale ventrikels van knaagdieren, gevolgd door elektrische pulsen die poriën in het celmembraan creëren, waardoor DNA in stamvadercellen langs de ventrikel kan komen. Vervolgens worden de door de geëlektroporeerde plasmiden gedragen reporter transgenes verwerkt door de doelcelmachines en uitgedrukt7. Twee elektroporatiemethoden zijn eerder beschreven om corticale astrocyten van muizen te labelen: 1) Postnatale astrocytenetikettering door elektroporatie (PALE), die afhankelijk is van de elektroporatie van 1-2 enkelkleurige episomale reporter plasmiden in vroege postnatale stadia4; 2) De StarTrack-strategie gebaseerd op in utero elektroporatie (IUE) van meerdere single-color integratieve reporter plasmids8,9,10. Hoewel deze twee technieken PrA efficiënt labelen in de hersenschors, vertonen ze ook enkele beperkingen. In hun oorspronkelijke versie vertrouwen beide methoden op een gliale fibrillaire zure eiwitpromotor (GFAP) om expressie in astrocyten te stimuleren, wat de etikettering naar radiale glia kan vertekenen, evenals piale en reactieve astrocyten die GFAP sterker uitdrukken dan normale rustende PrA11,12. Wat PALE betreft, zijn andere nadelen het late stadium van elektroporatie, dat etikettering van vroeggeboren PrA (of die afkomstig van vroege delaminerende voorlopers) en analyse van vroege stadia van astroglia-ontwikkeling voorkomt, en het gebruik van episomale vectoren die verdund raken door opeenvolgende divisies tijdens de massale proliferatie die PrA ondergaat tijdens de eerste postnatale week13,14. In tegenstelling tot PALE is StarTrack gebaseerd op de embryonale elektroporatie van integratieve reporter plasmiden die het mogelijk maken om de bijdrage van zowel embryonale als postnatale stamvaders aan PrA te volgen. Een bijgewerkt StarTrack-schema dat vertrouwt op de ubiquitine C-promotor (UbC-StarTrack) bereikt een bredere expressie van fluorescerende verslaggevers in zowel de neuronale als gliale afdaling (inclusief astrocyten) van neurale voorlopers15,16,17. In de huidige versie is de implementatie van deze aanpak echter complex, omdat het steunt op een equimolar-mengsel van 12 verschillende plasmiden die zes fluorescerende eiwitten (FP) uitdrukken met gedeeltelijke excitatie en emissiespectra-overlapping.
Hier gepresenteerd is een eenvoudige in utero elektroporatie-gebaseerde multicolor labeling methode met behulp van integratieve reporter constructies gedreven door een sterke en breed actieve promotor om corticale astrocyten14uit te kiezen . Bovendien wordt een eenvoudige pijplijn voor beeldanalyse met behulp van zowel gelicentieerde (bijv. Imaris) als open access (Vaa3D18,19,20) beeldanalysesoftware geleverd om respectievelijk astrocytenterritoriaal volume en arborisatie te segmenteren. In vergelijking met de eerder beschreven methoden deze strategie is uitsluitend gebaseerd op 1–2 multicolor integratieve transgenes Multiaddressable Genome-Integrating Color Markers (MAGIC Markers of MM21) gericht op het cytoplasmatische en (optioneel) nucleaire celcompartiment waarvan de expressie wordt aangedreven door een synthetische CAG promotor bestaande uit een cytomegalovirus enhancer, kip β-actine promotor, en konijn β-globine splice acceptor site22. Dit maakt etikettering en tracking van corticale astrocyten mogelijk, van embryonale tot late postnatale stadia, onafhankelijk van GFAP-expressie14,23. Elk van deze transgenen heeft de volgende vier verschillende FP's: eBFP, mTurquoise2/mCerulean, EYFP en tdTomato/mCherry, die minimale spectrale overlapping vertonen die gemakkelijk kan worden omzeild met 1) Sequentiële kanaalverwerving; 2) Geoptimaliseerde excitatiekracht en inzamelingswinst; en 3) Specifieke dichroïsche filters om smalle KP-emissievensters te verzamelen. De MM-strategie maakt gebruik vanCre/lox recombinatie met een zelfuitsnijdbare Cre recombinase (seCre) om stochastische expressie van FP in een cellulaire populatie te stimuleren. Een enkele kopie van MM-transgene drukt FP op een wederzijds exclusieve manier uit, terwijl meerdere transgenen aanleiding geven tot FP-combinaties, waardoor tientallen verschillende tinten ontstaan. De genomische integratie van de transgenen wordt aangedreven door het piggyBac (PB) of Tol2-omzettingssysteem24,25,26. Daarom maken de MM-toolkit en het veelkleurige 'mozaïek' dat het genereert, door in utero-elektroporatie gelijktijdige markering van meerdere aangrenzende corticale voorlopers mogelijk en het volgen van hun gliale afdaling, inclusief corticale astrocyten, over lange perioden. Kleurcontrast als gevolg van de expressie van verschillende FP maakt het mogelijk om de contour van PrA af te bakenen en vervolgens belangrijke informatie over hun territoriale volume (met behulp van IMARIS) en complexe morfologie (met vaa3D) te extraheren. De veelkleurige strategie die hier in detail wordt gepresenteerd, is een handige en robuuste methode die snelle en gemakkelijke toegang geeft tot het corticale astrocytenoppervlak en morfologie bij wilde muizen in verschillende ontwikkelingsstadia, en is gemakkelijk aanpasbaar om astrocytenanatomische kenmerken in muismodellen van neurologische ziekten te onderzoeken zonder transgene reporterlijnen te gebruiken.

in utero electroporation of multiaddressable genome-integrating color (magic) markers to individualize cortical mouse astrocytes

Protoplasmatische astrocyten (PrA) in de hersenschors van de muis worden strak naast elkaar geplaatst en vormen een schijnbaar continue driedimensionale matrix in volwassen stadia. Tot nu toe kan geen enkele immunostainingstrategie hen onderscheiden en hun morfologie segmenteren bij volwassen dieren en in de loop van corticogenese. Corticale PrA is afkomstig van voorlopers in het dorsale pallium en kan gemakkelijk worden gericht met behulp van utero-elektroporatie van integratieve vectoren. Hier wordt een protocol gepresenteerd om deze cellen te labelen met de multiaddressable genome-integrating color (MAGIC) Markers-strategie, die afhankelijk is van piggyBac/Tol2-omzetting en Cre /lox-recombinatie om stochastically verschillende fluorescerende eiwitten (blauw, cyaan, geel en rood) uit te drukken die zijn gericht op specifieke subcellulaire compartimenten. Deze veelkleurige fate mapping strategie maakt het mogelijk om in situ nabijgelegen corticale voorlopers te markeren met combinaties van kleurmarkers voorafgaand aan het begin van gliogenese en om hun nakomelingen, waaronder astrocyten, te volgen van embryonale tot volwassen stadia op individueel celniveau. Semi-schaarse etikettering bereikt door het aanpassen van de concentratie van elektroporated vectoren en kleurcontrast geleverd door de Multiaddressable Genome-Integrating Color Markers (MAGIC Markers of MM) maken het mogelijk om astrocyten te individualiseren en hun territorium en complexe morfologie te onderscheiden ondanks hun dichte anatomische opstelling. Hier wordt een uitgebreide experimentele workflow gepresenteerd, inclusief de details van de elektroporatieprocedure, multichannel beeldstapels acquisitie door confocale microscopie en computerondersteunde driedimensionale segmentatie waarmee de experimenteerder het individuele PrA-volume en de morfologie kan beoordelen. Kortom, elektroporatie van MAGIC Markers biedt een handige methode om individueel talrijke astrocyten te labelen en toegang te krijgen tot hun anatomische kenmerken in verschillende ontwikkelingsstadia. Deze techniek zal nuttig zijn om corticale astrocytenmorfologische eigenschappen in verschillende muismodellen te analyseren zonder toevlucht te nemen tot complexe kruisen met transgene reporterlijnen.

Elektroporatie van MAGIC Markers in embryonale corticale voorlopers maakt het mogelijk astrocyten te labelen van vroege tot late stadia van de ontwikkeling van de hersenschors (figuur 1). Deze astrocyten werden gevonden in alle corticale lagen in verschillende postnatale stadia (P4, P7, P21) omdat ze zich wijd verspreidden in de hele hersenschors.
